An indicator-based and indicator-free magnetic assays connected with a disposable pencil graphite electrode (PGE) were successfully developed, and also compared for the electrochemical detection of DNA hybridization. The oxidation signals of echinomycin (ECHI) and electroactive DNA bases, guanine and adenine, respectively were monitored in the presence of DNA hybridization by using differential pulse voltammetry (DPV) technique. The biotinylated probe was immobilized onto the magnetic beads (magnetic particles, microspheres) and hybridization with its complementary target at the surface of particles within the medium was exhibited successfully using electrochemical sensor system. For the selectivity studies, the results represent that both indicator-based and indicator-free magnetic assays provide a better discrimination for DNA hybridization compared to duplex with one-base or more mismatches. The detection limits (S/N = 3) of the magnetic assays based on indicator or indicator-free were found in nM concentration level of target using disposable sensor technology with good reproducibility. The characterization and advantages of both proposed magnetic assays connected with a disposable electrochemical sensor are also discussed and compared with those methods previously reported in the literature. 相似文献

In the title compounds, {2,2′‐[2,2‐dimethyl‐1,3‐propanediylbis(nitrilomethylidyne)]diphenolato‐κ4N,N′,O,O′}nickel(II), [Ni(C19H20N2O2)], and {2,2′‐[2,2‐dimethyl‐1,3‐propanediylbis(nitrilomethylidyne)]diphenolato‐κ4N,N′,O,O′}copper(II), [Cu(C19H20N2O2)], the NiII and CuII atoms are coordinated by two iminic N and two phenolic O atoms of the N,N′‐bis(salicylidene)‐2,2‐dimethyl‐1,3‐propanediaminate (SALPD2?, C17H16N2O22?) ligand. The geometry of the coordination sphere is planar in the case of the NiII complex and distorted towards tetrahedral for the CuII complex. Both complexes have a cis configuration imposed by the chelate ligand. The dihedral angles between the N/Ni/O and N/Cu/O coordination planes are 17.20 (6) and 35.13 (7)°, respectively. 相似文献

Four complexes of the nuclear structure NiII–ZnII were prepared with bis-N,N′-(salicylidene)-1,3-propanediamine (LH2), bis-N,N′-(salicylidene)-2,2′-dimethyl-1,3-propanediamine (LDMH2) and the reduced derivatives of these Schiff bases, bis-N,N′-(2-hydroxybenzyl)-1,3-propanediamine (LHH2), bis-N,N′-(2-hydroxybenzyl)-2,2′-dimethyl-1,3-propanediamine (LDMHH2). The complexes were characterized using IR spectroscopy, elemental analysis and thermogravimetric methods. The stoichiometry of the complex molecules were found to be NiL·ZnCl2·(DMF)2, NiLDM·ZnCl2·(DMF)2, NiLH·ZnCl2·(DMF)2 and NiLDMH·ZnCl2·(DMF)2. The molecular models of the complexes prepared with the reduced Schiff bases were determined according to the X-ray diffraction method. It is seen that in these complexes Ni(II) is in octahedral and Zn(II) is in tetrahedral coordination sphere. Ni(II) ion is coordinated between two nitrogen and two oxygen donors of the ligand and oxygen donors of the two DMF molecules. Zn(II) ion on the other hand is coordinated between two oxygen of the organic ligand forming two μ bonds. It also coordinates two Cl ions. The thermogravimetric analysis showed that the complex NiLDMH·ZnCl2·(DMF)2 containing methyl groups is more stable than the other complex NiLH·ZnCl2·(DMF)2 containing reduced Schiff base. The coordinative DMF molecules in NiLDMH·ZnCl2·(DMF)2 were thermally cleaved. However, the cleavage of DMF molecules NiLH·ZnCl2·(DMF)2 resulted in the thermal degradation of the complex. In order to explain the TG data of the ligands were titrated in non-aqueous medium and their basicity strengths were determined. It was found that the basicity of the ligands containing two methyl groups were stronger. It is understood that the two methyl groups increase the negative charge density on nitrogen causing an increase in complex stability. 相似文献

A novel UV-VIS spectrophotometric method was developed in this study by using solid phase extraction procedure for the simultaneous preconcentration, separation and determination of trace levels of Pb (II), Cd (II) and Zn (II) ions in various water samples by using Amberlite N,N-bis(salicylidene)cyclohexanediamine (SCHD) resin. This study presents the results of experimental procedures carried out like the adsorption of analytes to the resin, influences of some analytical parameters that effect the recovery such as pH, sample volume, sample flow rate, eluent type and concentration, eluent volume, eluent flow rate and the effects of alkaline metals, earth alkaline metals and some other transition metals. The analytes in the samples with the adjusted pH range of 4–7 were adsorbed on XAD-4-SCHD resin and eluted by using 1.0 mol L?1 nitric acid. The amounts of ions were determined by using UV-VIS spectrometer. The limits of detection were 0.03, 0.07 and 0.05 µg mL?1 for Pb (II), Cd (II) and Zn (II), respectively. The accuracy of the method was assured by the analysis of the certified standard water sample NW-TMDA-70.2 and the observed recoveries were above 93%. Different environmental water samples that contain trace amounts of Pb (II), Cd (II) and Zn (II) were analysed by using the method developed in this study. Same samples were also analysed by ICP-MS for comparison and almost the similar results were observed. The method developed in this study was successfully applied to the various environmental water samples to determine the trace levels of Pb (II), Cd (II) and Zn (II) ions. 相似文献

Mononuclear nickel(II) complexes were prepared by reaction of the three ONNO type reduced Schiff bases bis‐N,N′‐(2‐hydroxybenzyl)‐1,3‐propanediamine (LHH2), bis‐N,N′‐(2‐hydroxybenzyl)‐2,2′‐dimethyl‐1,3‐propanediamine (LDMHH2), and bis‐N,N′‐[1‐(2‐hydroxyphenyl)ethyl]‐1,3‐propanediamine (LACHH2) with NiII ions in the presence of pseudo halides (OCN–, SCN– and N3–). The complexes were characterized with the use of elemental analyses, IR spectroscopy, and thermal analyses. The molecular structure of one of the complexes was obtained by single‐crystal X‐ray diffraction. The obtained complexes are mononuclear, and a pseudo halide molecule is attached. One of the oxygen atoms of the ligand is in phenolate and the other was in phenol form. According to the thermogravimetry results, it was thought that the pseudo halide thermally detaches from the structure as hydropseudo halide. In azide‐containing complexes an endothermic reaction was observed although the azide group usually decomposes with an exothermic reaction. 相似文献

Determination of 90Sr in environmental solid samples is a challenging task because of the presence of so many other radionuclides in samples of interest. This problem was dealt with by radiochemical separation of strontium followed by yttrium separation and Cerenkov counting of the high-energy ??-particle emissions of 90Y in order to quantitate 90Sr. In this work, an improved method is described for the determination of 90Sr in soil samples, through the separation of the daughter 90Y at equilibrium. The procedure is based on the HDEHP solvent extraction in combination with liquid scintillation spectrometry (LSS). A low background Quantulus has been optimized for low level counting of Cerenkov radiation emitted by the hard ??-emitter 90Y. The analytical quality of the method has been checked by analyzing IAEA Soil-375 reference materials. The analytical method has also been successfully applied to the determination of 90Sr for moss-soil samples in inter-laboratory exercises through IAEA??s ALMERA network. The chemical recovery for 90Y extraction ranged from 80 to 85% and the counting efficiency was 73% in the window 25?C400 keV. 相似文献

The polymer, poly(vinylferrocenium) (PVF+) modified electrode was developed as the first time herein for the improved electrochemical sensing of DNA based on the oxidation signals of polymer and guanine. The morphologies of polymer film and DNA immobilized polymer film were examined using scanning electron microscope (SEM). The electrochemical behavior of polymer modified electrode was investigated by using cyclic voltammetry (CV) and differential pulse voltammetry (DPV) in the absence/presence of DNA. Experimental parameters, such as the polymeric film thickness, the DNA immobilization time, the concentration of buffer solution, pH and DNA concentration were examined in order to obtain more sensitive and selective electrochemical signals. Metal determinations at low concentration levels (≤ng mL−1) comprise one of most important targets in analytical chemistry. This interest also increases in different areas such as biology, medicine, environment and food samples. In spite of inherent high sensitivities obtained for electrothermal atomic absorption spectrometry (ETAAS) and inductively coupled plasma-mass spectrometry (ICP-MS), these techniques have some limitations depending on the concomitants. As a result, interest in preconcentration techniques still continues increasingly for trace metal determinations by flame atomic absorption spectrometry (FAAS) due to the high accuracy of this method.In this work, thioureasulfonamide resin was synthesized, characterized and applied as a new sorption material for determinations of cadmium and lead in water samples. The method is based on the sorption of Cd and Pb ions on the synthesized resin without using any complexing reagent. The optimization of experimental conditions was performed using factorial design including pH, amount of resin, contact time, first sample volume and final eluent volume. Using the experimental conditions defined in the optimization, the method was applied to the determination and preconcentration of Cd and Pb at ng mL−1 level in natural water. Flame AAS was used for trace metal determinations. This method exhibits the superiority in compared to the other adsorption reagents because of the fact that there is no necessity of any complexing reagent and optimum pH of solution presents in acidic media. Consequently, 600- and 360-fold improvements in the sensitivity of FAAS were achieved by combining the slotted tube atom trap-atomic absorption spectrometry (STAT-FAAS) and the purposed enrichment method for Cd and Pb, respectively. 相似文献

In the present work, diethanol amine modified polystyrene based polymer (PVBC-Diethanol amine) was synthesized and characterized, then surface properties of the polymer were examined by inverse gas chromatography method at infinite dilution. The retention diagrams obtained based on the interaction of polar and nonpolar probes with the polymer were drawn over a temperature range from 30 to 55 °C. Through the diagrams, the dispersive component of the surface free energy, g S D of the polymer surface, and the specific enthalpy of adsorption, DH A S , of probes on the polymer were also calculated. Lewis acid, K A , and Lewis base, K D , parameters of PVBC-Diethanol amine surface were determined. The values of K A and K D indicated that PVBC-Diethanol amine surface exhibited a basic behavior. 相似文献
