We developed a novel method to fabricate a crab-like microrobot that can actuate for a long period in a physiological condition. The microrobot backbone was built with a biocompatible and elastic material-polydimethylsiloxane (PDMS)-by using a specially designed 3D molding aligner, and consisted of three strips of PDMS "legs" connected across a "body." Cardiomyocytes were then plated on the grooved top surface of the backbone, resulting in a high concentration of pulsating cells. These key techniques enabled the microrobot to walk continuously for over ten days. The performance of our crab-like microrobot was measured at an average velocity of 100 microm s(-1), and the estimated total distance it travelled was 50 m over a one-week period. Thus, we have demonstrated for the first time a walking robot that exhibited reliable and long-term actuation performances.  相似文献

Protein–ligand docking is a useful tool for providing atomic-level understanding of protein functions in nature and design principles for artificial ligands or proteins with desired properties. The ability to identify the true binding pose of a ligand to a target protein among numerous possible candidate poses is an essential requirement for successful protein–ligand docking. Many previously developed docking scoring functions were trained to reproduce experimental binding affinities and were also used for scoring binding poses. However, in this study, we developed a new docking scoring function, called GalaxyDock BP2 Score, by directly training the scoring power of binding poses. This function is a hybrid of physics-based, empirical, and knowledge-based score terms that are balanced to strengthen the advantages of each component. The performance of the new scoring function exhibits significant improvement over existing scoring functions in decoy pose discrimination tests. In addition, when the score is used with the GalaxyDock2 protein–ligand docking program, it outperformed other state-of-the-art docking programs in docking tests on the Astex diverse set, the Cross2009 benchmark set, and the Astex non-native set. Ligand-dependent particle size control of PbSe quantum dots   总被引:1,自引:0,他引:1  
Colloidal solutions of monodisperse PbSe quantum dots (QDs) were synthesized by a hot solution chemical method from a reaction mixture of lead oleate and TOPSe (TOP: tri-n-octylphosphine). The synthesis was carried out at a fixed temperature (170 degrees C) and time, while the particle sizes of the PbSe QDs were controlled by using two different kinds of organic ligands with varied chain length. It was seen that the tuning of PbSe QDs are possible by using the proper molar ratio of the co-ligands, such as acetic acid or hexanoic acid, at a fixed reaction temperature and time, verified by TEM and XRD as well as NIR absorption analysis. The effects of different organic acids were studied and the role of additional organic acids might be due to the extent of ligand exchange efficiency between the Pb oleate and acetic/hexanoic acid in the initial stage, which is caused by the steric hindrance effects of the acids.  相似文献

We report here a novel strategy for the high-sensitive detection of target biomolecules with very low concentrations on a quartz crystal microbalance (QCM) device using gold nanoparticles as signal enhancement probes. By employing a streptavidin-biotin interaction as a model system, we could prepare biotin-conjugated gold nanoparticles maintaining good dispersion and long-term stability by controlling the biotin density on the surface of gold nanoparticles that have been investigated by UV-vis spectra and AFM images. These results showed that 10 microM N-(6-[biotinamido]hexyl)-3'-(2'-pyridyldithio)propionamide (biotin-HPDP) was the critical concentration to prevent the nonspecific aggregation of gold nanoparticles in this system. For sensing streptavidin target molecules by QCM, biotinylated BSA was absorbed on the Au surface of the QCM electrode and subsequent coupling of the target streptavidin to the biotin in the sensing interface followed. Amplification of the sensing process was performed by the interaction of the target streptavidin on the sensing surface with gold nanoparticles modified with 10 microM biotin-HPDP. The biotinylated gold nanoparticles were used as signal amplification probes to improve the detection limit, which was 50 ng/ml, of the streptavidin detection system without signal enhancement, and the calibration curve determined for the net frequency changes showed good linearity over a wide range from 1 ng/ml to 10 microg/ml for the quantitative streptavidin target molecule analysis. In addition, the measured dissipation changes suggested that the layer of biotin-BSA adsorbed on the Au electrode and the streptavidin layer assembled on the biotin-BSA surface were highly compact and rigid. On the other hand, the structure formed by the biotinylated gold nanoparticles on the streptavidin layer was flexible and dissipative, being elongated outward from the sensing surface.  相似文献

As an alternative to strong acid reaction media for the Friedel–Crafts acylation for a polymer‐forming reaction, a mild polyphosphoric acid (PPA) with optimized amount of phosphorous pentoxide (P2O5) has been tested for the polymerization of AB monomers 4‐(2‐phenoxyethoxy)benzoic acid and 3‐(2‐phenoxyethoxy)benzoic acid, and an AB2 monomer 3,5‐bis(2‐phenoxyethoxy)benzoic acid. The reaction progress of AB2 monomer was conveniently traced by FTIR spectroscopy monitoring aromatic ketone (C?O) stretching bands arisen from carboxylic acid groups at the chain ends and carbonyl groups in the backbone as a function of reaction time at 110 °C. The resultant linear and hyperbranched polymers containing flexible oxyethylene spacers, which were prone to be hydrolyzed in strong acids at elevated temperature, displayed high intrinsic viscosities. Thus, the reaction medium PPA/P2O5 mixture as an electrophilic substitution reaction was indeed benign not to depolymerize growing polymer molecules but strong enough for the direct generation of carbonium ion from carboxylic acid to promote efficient polymerization. The resultant hyperbranched poly(etherketone) (PEK) displayed the best solubility among samples. All PEKs showed good thermal stability; glass transition temperatures were in the range of 90–117 °C; 5% weight loss generally occurred at greater than 345 °C in air. © 2007 Wiley Periodicals, Inc. Chaga mushroom (Inonotus obliquus) comprises polyphenolic compounds, triterpenoids, polysaccharides, and sterols. Among the triterpenoid components, inotodiol has been broadly examined because of its various biological activities. The purpose of this study is to examine inotodiol from a safety point of view and to present the potential possibilities of inotodiol for medical usage. From chaga mushroom extract, crude inotodiol (INO20) and pure inotodiol (INO95) were produced. Mice were treated with either INO20 or INO95 once daily using oral administration for repeated dose toxicity evaluation. Serum biochemistry parameters were analyzed, and the level of pro-inflammatory cytokines in the serum was quantified. In parallel, the effect of inotodiol on food allergic symptoms was investigated. Repeated administration of inotodiol did not show any mortality or abnormalities in organs. In food allergy studies, the symptoms of diarrhea were ameliorated by administration with INO95 and INO20. Furthermore, the level of MCPT-1 decreased by treatment with inotodiol. In this study, we demonstrated for the first time that inotodiol does not cause any detrimental effect by showing anti-allergic activities in vivo by inhibiting mast cell function. Our data highlight the potential to use inotodiol as an immune modulator for diseases related to inflammation.  相似文献
