A validated kinetic spectrophotometric method has been developed for the determination of losartan potassium in pure and dosage forms. The method is based on oxidation of the losartan potassium with alkaline potassium permanganate at room temperature (25 ± 1 °C). The reaction is followed spectrophotometrically by measuring the increase in absorbance with time at 603 nm, and the initial rate, fixed time (at 12.0 min) and equilibrium time (at 90.0 min) methods are adopted for constructing the calibration graphs. All the calibration graphs are linear in the concentration range of 7.5–60.0 μg mL?1 and the calibration data resulted in the linear regression equations of n? = ?6.422 × 10?7 + 1.173 × 10?5 C, A =3.30 × 10?4 + 5.28 × 10?3 C and A = ?2.09 × 10?2 + 1.05 × 10?1 C for initial‐rate, fixed time and equilibrium time methods, respectively. The limits of detection for initial rate, fixed time and equilibrium time methods are 0.71, 0.21 and 0.19 μg mL?1, respectively. The activation parameters such as Ea, ΔH?, ΔS?, and ΔG? are also determined for the reaction and found to be 87.34 KJ mol?1, 84.86 KJ mol?1, 50.96 JK?1 mol?1 and ?15.10 KJ mol?1, respectively. The variables are optimized and the proposed methods are validated as per ICH guidelines. The method has been applied successfully to the estimation of losartan potassium in commercial tablets. The performance of the proposed methods was judged by calculating paired t‐ and F‐ values. The analytical results of the proposed methods when compared with those of the reference method show no significant difference in accuracy and precision and have acceptable bias.  相似文献

Potential genotoxic impurities in pharmaceuticals at trace levels are of increasing concern to both pharmaceutical industries and regulatory agencies due to their possibility for human carcinogenesis. Molecular functional groups that render starting materials and synthetic intermediates as reactive building blocks for small molecules may also be responsible for their genotoxicity. Determination of these genotoxic impurities at trace levels requires highly sensitive and selective analytical methodologies, which poses tremendous challenges on analytical communities in pharmaceutical research and development. Experimental guidance for the analytical determination of some important classes of genotoxic impurities is still unavailable in the literature. Therefore, the present review explores the structural alerts of commonly encountered potential genotoxic impurities, draft guidance of various regulatory authorities in order to control the level of impurities in drug substances and to assess their toxicity. This review also describes the analytical considerations for the determination of potential genotoxic impurities at trace levels and finally few case studies are also discussed for the determination of some important classes of potential genotoxic impurities. It is the authors’ intention to provide a complete strategy that helps analytical scientists for the analysis of such potential genotoxic impurities in pharmaceuticals.  相似文献

The objective of this work is to develop and validate spectrophotometric method for the determination of piroxicam in commercial dosage forms. The method is based on the chelation of the drug with Fe(III) to form pink coloured metal chelate at room temperature which absorbs maximally at 504 nm. Beer's law is obeyed over the concentration range of 8–160 μg mL?1 (A = 1.07 × 10?3 + 7.75 × 10?3 C). Under the optimized experimental conditions, proposed method is validated as per the International Conference on Harmonisation guidelines. The limits of detection and quantitation for the proposed method are 0.775 and 2.348 μg mL?1, respectively. The proposed method has been successfully applied to the determination of piroxicam in commercial dosage forms. The results are compared with the reference El‐Ries et al. spectrophotometric method.  相似文献

An analysis to disentangle information about the occurrence of dynamical fluctuations in multiparticle production in high energy nucleus-nucleus collisions has been carried out in terms of fractal moments and entropy for the experimental and simulated data using FRITIOF, UrQMD and HIJING generators. Although there is a possibility to thermodynamically interpret the final state of multiparticle production by calculating the values of specific heat, c, using G q - and F q -moments. However, both these moments give markedly different values of c. The constancy observed in the values of specific heats calculated from either G q - or F q -moments is in accord with the predictions of constant specific heat approximation. Variations of multifractal and factorial moments and various other parameters calculated from these moments predict the presence of non-statistical fluctuations in high energy nuclear collisions.  相似文献

The Heck cross-coupling reaction is a well-established chemical tool for the synthesis of unsaturated compounds by formation of a new C-C bond. In this study, 1,3-diarylpropene derivatives, designed as structural analogues of stilbenoids and dihydrostilbenoids, were synthesised by the palladium-catalysed reactions of 2-amidoiodobenzene derivatives with either estragole or eugenol. The products were obtained with high (E) stereoselectivity but as two regioisomers. The ratios of isomers were found to be dependent on the nature of the allylbenzene partner and were rationalised by electronic effects exercising a determining influence in the β-hydride elimination step. In addition, the cytotoxic effects of all the Heck reaction products were evaluated against MCF-7 and MDA-MB-231 human breast cancer cells, with unpromising results. Among all, compound 7d exhibited weak cytotoxic activity towards MCF-7 cell lines with IC50 values of 47.92 µM in comparison with tamoxifen and was considered to have general toxicity (SI value < 2).  相似文献

This paper proposes and investigates the use of several factors for portfolio selection of international mutual funds. Three of the selected factors are specific to mutual funds, additional three factors are taken from Macroeconomics and one factor represents regional and country preferences. Two different flexible osmium redox polymers; poly(1-vinylimidazole)12-[Os-(4,4'-dimethyl-2,2'-di'pyridyl)2Cl2](2+/+) (osmium redox polymer I) and poly(vinylpyridine)-[Os-(N,N'-methylated-2,2'-biimidazole)3](2+/3+) (osmium redox polymer II) were investigated for their ability to efficiently "wire" Pseudomonas putida ATCC 126633 and Pseudomonas fluorescens (P. putida DSM 6521), which are well-known phenol degrading organisms, when entrapped onto cysteamine modified gold electrodes. The two Os-polymers differ in redox potential and the length of the side chains, where the Os(2+/3+)-functionalities are located. The bacterial cells were adapted to grow in the presence of phenol as the sole source of organic carbon. The performance of the redox polymers as mediators was investigated for making microbial sensors. The analytical characteristics of the microbial sensors were evaluated for determination of catechol, phenol and glucose as substrates in both batch analysis and flow analysis mode.  相似文献

The objective of this work is to develop a new kinetic spectrophotometric method for the determination of irbesartan in pharmaceutical formulations. The method is based on the reaction of carboxylic acid group of the oxidized irbesartan with a mixture of potassium iodate (KIO(3)) and iodide (KI) to form yellow colored triiodide ions in aqueous medium at 30+/-1 degrees C. The reaction is followed spectrophotometrically by measuring the rate of change of absorbance at 352 nm. The initial-rate and fixed-time (DeltaA) methods are adopted for constructing the calibration curves, which were found to be linear over the concentration ranges of 10.0-60.0 and 7.5-60.0 microg ml(-1) respectively. The regression analysis of calibration data yielded the linear equations: rate=-2.138 x 10(-6)+1.058 x 10(-4)C and DeltaA=-3.75 x 10(-3)+3.25 x 10(-3)C for initial rate and fixed time (DeltaA) methods, respectively. The limit of detection for initial rate and fixed time methods are 0.21 and 2.40 mug ml(-1), respectively. The various activation parameters such as E(a), DeltaH++, DeltaS++ and DeltaG++ are also calculated for the reaction and found to be 70.95+/-0.43 kJ mol(-1), 68.48+/-0.21 kJ mol(-1), 16.54+/-0.24 J K(-1) mol(-1) and -4.94+/-0.07 kJ mol(-1), respectively. The proposed methods are optimized and validated as per the guidelines of International Conference on Harmonisation (U.S.A.). The point and interval hypothesis tests have been performed which indicate that there is no significant difference between the proposed methods and the reference method. The methods have been successfully applied to the determination of irbesartan in commercial dosage forms.  相似文献
