In vitro production of sperm is a desirable idea for fertility preservation in azoospermic men and prepubertal boys suffering from cancer. In this study, a biocompatible porous scaffold based on a triad mixture of silk fibroin (SF), alginate (Alg), and laminin (LM) is developed to facilitate the differentiation of mouse spermatogonia stem cells (SSCs). Following SF extraction, the content is analyzed by SDS-PAGE and stable porous 3D scaffolds are successfully prepared by merely Alg, SF, and a combination of Alg-SF, or Alg-SF-LM through freeze-drying. Then, the biomimetic scaffolds are characterized regarding the structural and biological properties, water absorption capacity, biocompatibility, biodegradability, and mechanical behavior. Neonatal mice testicular cells are seeded on three-dimensional scaffolds and their differentiation efficiency is evaluated using real-time PCR, flow cytometry, immunohistochemistry. Blend matrices showed uniform porous microstructures with interconnected networks, which maintained long-term stability and mechanical properties better than homogenous structures. Molecular analysis of the cells after 21 days of culture showed that the expression of differentiation-related proteins in cells that are developed in composite scaffolds is significantly higher than in other groups. The application of a composite system can lead to the differentiation of SSCs, paving the way for a novel infertility treatment landscape in the future.  相似文献

A glassy carbon electrode (GCE) modified with polymeric nanocomposite consisting of palladium nanoparticles and a conductive polymeric ionic liquid was prepared. The modified GCE was applied to sensitive and fairly selective electrochemical determination of the mycotoxin zearalenone. Electrocatalytic oxidation is performed in a solution containing 20 % (V/V) acetonitrile and 80 % (V/V) of 1 M perchloric acid. Cyclic voltammetry and square wave voltammetry revealed a well-defined electrocatalytic peak current at overpotential of +0.69 V versus Ag/AgCl. Under optimized experimental conditions, there is a linear relationship between anodic peak current and zearalenone concentration in the range from 0.03 to 35 ng?mL ̄1, and the detection limit is 0.01 ng?mL ̄1. The method was successfully applied to the analysis of zearalenone in spiked food samples and gave recoveries between 95.6 and 104.0 %.

In this article, a new ligandless dispersive liquid-liquid microextraction method has been developed for preconcentration of trace quantities of silver as a prior step to its determination by flame atomic absorption spectrometry. In the proposed approach, carbon tetrachloride and ethanol were used as extraction and dispersive solvents. Several factors that may be affected on the extraction process, like, extraction solvent, disperser solvent, the volume of extraction and disperser solvent, pH of the aqueous solution and extraction time were optimized. Under the optimal conditions, the calibration curve was linear in the range of 5.0 ng mL−1 to 2.0 μg mL−1 of silver with R2 = 0.9995 (n = 9) and detection limit based on three times the standard deviation of the blank (3Sb) was 1.2 ng mL−1 in original solution. The relative standard deviation for eight replicate determination of 0.5 μg mL−1 silver was ±1.5%. The high efficiency of dispersive liquid-liquid microextraction to carry out the determination of silver in complex matrices was demonstrated. The proposed method has been applied for determination of trace amount of silver in standard and water samples with satisfactory results.  相似文献

This study describes the effect of covalent derivatization of source and drain electrodes with monolayers of organic semiconductors. These monolayers form a template on the metal surface and provide better electronic coupling between the electrode and the semiconductor. We see a large improvement in nanoscale (40-100 nm) transistors only when the monolayer presents functionality that is complementary to the chemical and electronic structure of the molecular semiconductor.  相似文献

Recent developments in the field of applied nanoscience and nanotechnology have heightened the need for categorizing various characteristics of nanostructures. In this regard, this paper establishes a novel method to investigate magnetic properties (phase diagram and spontaneous magnetization) of a cylindrical Ising nanotube. Using a two-layer Ising model and the core-shell concept, the interactions within nanotube has been modelled. In the model, both ferromagnetic and antiferromagnetic cases have been considered. Furthermore, the effect of nanotube's length on the critical temperature is investigated. The model has been simulated using cellular automata approach and phase diagrams were constructed for different values of inter- and intra-layer couplings. For the antiferromagnetic case, the possibility of existence of compensation point is observed.  相似文献

Recent preclinical and clinical testing of hypocrellin-based photosensitizer SL052 for use in photodynamic therapy (PDT) of cancer has shown encouraging results. Further optimization of its formulation for delivery could considerably extend the therapeutic efficiency of this drug. A nanoformulation encapsulating SL052 into biodegradable polymer poly(lactic-co-glycolic acid) (PLGA) was developed using a single-emulsion solvent evaporation technique and characterized in terms of particle size and loading of the photosensitizing agent. This nanoformulation, SL052-PLGA-nanoparticles (NPs), was compared with recently created nanoformulation based on polyvinylpyrrolidone (SL052-PVP-NPs) and standard liposomal SL052 preparation in terms of efficacy when used for PDT treatment of squamous cell carcinomas SCCVII growing subcutaneously in syngeneic mice. The therapeutic effect of PDT using these three different SL052 formulations was tested for both 1 and 4 h intervals between drug injection and tumor light exposure. The longer time interval produced higher tumor cure rates with all SL052 preparations. With both drug-light intervals, PDT based on SL052-PLGA-NPs produced superior therapeutic benefit compared with the other two SL052 formulations.  相似文献

We demonstrate that modifying pentacene to incorporate an acid-labile moiety into its molecular structure leads to a new precursor that can be easily deposited, photopatterned, and processed via wet-chemical methods to produce organic semiconducting devices exhibiting good electrical characteristics. Acidic conditions produced by ultraviolet illumination of a co-deposited photoacid generator greatly accelerate the local conversion of this N-sulfinyl-tert-butylcarbamate pentacene adduct back to pentacene. Photopatterned thin-film transistors exhibit carrier mobilities in excess of 0.1 cm2 V-1 s-1, making this an attractive precursor for fabrication of large-area organic electronics via solution-phase methods.  相似文献

A highly selective and sensitive anodic stripping differential pulse polarographic method has been developed for the determination of trace amount of bismuth in various samples after adsorption of its 2-(5-bromo-2-pyridylazo)-5-diethylaminophenol complex on amberlite XAD-2 resin in the pH range of 2.0-3.0. The retained analyte on the resin recovered with 10.0 ml of 2 M hydrochloric acid and bismuth is determined by anodic stripping differential pulse polarography. In this case, 0.15 μg of bismuth can be concentrated in the column from 1100 ml of aqueous sample, where its concentration is as low as 0.14 ng ml−1 and the linearity is maintained in the concentration range 0.05-160 μg ml−1 in final solution with a correlation coefficient of 0.999 and relative standard deviation of ±1.2%. Various parameters such as the effect of pH, flow rate, and interference of number of metal ions on the determination of bismuth have been studied in detail to optimize the conditions for determination of bismuth in standard alloys: Nippon Keikinzoku Kogyo (NKK CRM No. 916 aluminum alloy); (NKK CRM No. 1021 Al, Si, Cu, Zn alloy); and (NKK CRM No. 920 aluminum alloy), biological samples: National Institute for Environmental Studies (NIES, No. 5 human hair; NIES, No. 7 tea leaves) and water samples: (spring water, river water; Kerman water; and Shiraz water).  相似文献
