CryoTEM demonstrates that a CNA35-bearing liposomal MRI contrast agent selectively binds to poorly assembled collagen type I as opposed to well-assembled collagen fibrils, whereas monomeric CNA35 binds to all forms of collagen. It is shown that upon conjugation to liposomes and micelles CNA35 loses its ability to dissociate ordered collagen fibrils and thereby to create its own binding sites.  相似文献

An apparatus for the production of hydrogen for hydrogenation-induced polarization studies was developed. The apparatus provides hydrogen gas from a solid source, thereby simplifying the requirement for siting of the apparatus. The produced hydrogen can be either isotopically enriched (with deuterium) or spin-enriched (with parahydrogen). These specialty gases were produced at small predetermined quantities and ambient pressure. The properties of the hydrogen mixtures were characterized by gas-phase nuclear magnetic resonance. The T 1 of the hydrogen mixtures (3.7 ms) was not affected by para-enrichment. The line width of the hydrogen signal in the para-enriched mixture was 34% larger. The reaction of ethyl propiolate hydrogenation served to evaluate the performance in terms of the ability to create hyperpolarized states. In this reaction, a polarization of 11.2% was measured for protons. Consecutive alkene hydrogenations as well as hydrogenations of the catalyst ligand resulted in additional hyperpolarized signals which were systematically assigned.  相似文献

Deuterated 13C sites in sugars (D-glucose and 2-deoxy-D-glucose) showed 6.3-to-17.5-fold higher solid-state dynamic nuclear polarization (DNP) levels than their respective protonated sites at 3.35T. This effect was found to be unrelated to the protonation of the bath. Deuterated 15N in sites bound to exchangeable protons ([15N2]urea) showed a 1.3-fold higher polarization than their respective protonated sites at the same magnetic field. This relatively smaller effect was attributed to incomplete deuteration of the 15N sites due to the solvent mixture. For a 15N site that is not bound to protons or deuterons ([15N]nitrate), deuteration of the bath did not affect the polarization level. These findings suggest a phenomenon related to DNP of X-nuclei directly bound to deuteron(s) as opposed to proton(s). It appears that direct binding to deuterons increases the solid-state DNP polarization level of X-nuclei which are otherwise bound to protons.  相似文献
