Metabolic activation of drug candidates to electrophilic reactive metabolites that can covalently modify cellular macromolecules may result in acute and/or idiosyncratic immune system-mediated toxicities in humans. This presents a significant potential liability for the future development of these compounds as safe therapeutic agents. We present here an example of an approach where sites of metabolic activation within a new drug candidate series were rapidly identified using online liquid chromatography/multi-stage mass spectrometry on an ion trap mass spectrometer. This was accomplished by trapping the reactive intermediates formed upon incubation of compounds with rat and human liver microsomes as their corresponding glutathione conjugates and mass spectral characterization of these thiol adducts. Based on the structures of the GSH adducts identified, potential sites and mechanisms of bioactivation within the chemical structure were proposed. These metabolism studies were interfaced with iterative structural modifications of the chemical series in order to block these bioactivation sites within the molecule. This strategy led to a significant reduction in the propensity of the compounds to undergo metabolic activation as evidenced by reductions in the irreversible binding of radioactivity to liver microsomal material upon incubation of tritium-labeled compounds with this in vitro system. With the efficiency and throughput achievable with such an approach, it appears feasible to identify and address the metabolic activation potential of new drug leads during routine metabolite identification studies in an early drug discovery setting. 相似文献

A new tetradentate dihydrogen perchlorate macrocyclic ligand (2,4,9,11-tetraphenyl-1,5,8,12-tetraazacyclotetradeca-1,4,8,11-tetraene dihydrogen perchlorate) was prepared and characterised. The macrocycle behaves as a selective chelating ion-exchanger for some metal ions. The polystyrene-based membrane electrode is found to exhibit quite promising selectivity for Cr3+ ions. It can be used to estimate chromium concentrations in the range 3.16x 10(-6)-1.00x10(-1) M with a near-Nernstian slope of 17.5 mV per decade of concentration between pH 3.0 to 6.5. The electrode is found to possess a fast response time of 15 s and was used over a period of three months with good reproducibility (s = +/- 0.3 mV). The selectivity coefficient values for mono-, di- and trivalent cations indicate excellent selectivity for Cr3+ ions over a large number of other cations. Anions such as Cl- and SO4(2-) do not interfere and the electrode also works satisfactorily in a mixed organic-water solution. The sensor has been used as an indicator electrode for the potentiometric titration of Cr3+ with EDTA. The practical utility of the membrane sensor has also been demonstrated in solutions contaminated with detergents (CTAB and SDS). Several polyesters containing thiaxanthone rings were prepared from 2,7-dichloroformylthiaxanthone-5,5′-dioxide ( IVa ), 2,8-dichloroformylthiaxanthone-5,5′-dioxide ( IVb ), and bisphenols by solution polycondensation. The 2,8-diethoxycarbonylthiaxanthone-5,5′-dioxide ( V ) was prepared and characterized by spectral methods to confirm the formation of 2,8-thiaxanthonedicarboxylic acid-5,5′-dioxide ( IVb ). Prior to polymer synthesis two model compounds,2,7-diphenoxycarbonylthiaxanthone-5,5′-dioxide (MDE-1) and 2,8-diphenoxycarbonylthiaxanthone-5,5′-dioxide (MDE-2), were synthesized and characterized by spectral methods. The polyesters were obtained in 62–78% yield and had inherent viscosities in the range 0.42–0.90 dL/g. The effect of thiaxanthone rings on solubility, crystallinity, and thermal stability of the polyesters are also discussed. The polyesters have decomposition temperatures in the range 372–438°C. 相似文献

A rapid reversed-phase high-performance liquid chromatographic procedure is developed and validated for the resolution of
the cis-isomer of 1-[[p-[2-(3-ethyl-4-methyl-2-oxo-3-pyrroline-1-carboxamido) ethyl] phenyl] sulfonyl]-3-(trans-4-methylcyclohexyl) urea, a dopamine agonist in bulk drugs. The cis-isomer and glimepiride were baseline resolved on a Waters Symmetry column (50 × 4.6 mm, 3.5 μm) using a mobile phase system
containing water: tetrahydrofuran (75:25; v/v. The chromatographic resolutions between cis-isomer and glimepiride were found to be greater than two. The developed method was extensively validated and proved to be
robust. The limit of detection and the limit of quantification of cis-isomer were 500 and 1,500 ng mL−1, respectively, for 10 μL injection volume. The percentage recovery of the cis-isomer ranged from 97.3 to 102.0 in bulk drug samples of glimepiride. Glimepiride sample solution and mobile phase were found
to be stable for at least 48 h. The proposed method was found to be suitable and accurate for the quantitative determination

The title complex was prepared and characterized by elemental and spectral analyses. The formation constants of various heterocyclic nitrogen base adducts with the NiII–di(2,4-dibromophenyl)carbazonate have been determined in a monophase by spectrophotometry at 25 ± 0.1 °C. The monodenatate and bidentate bases form penta- and hexa-coordinated adducts respectively with 1:1 stoichiometry for the metal chelate-base, whereas the unsaturated monodentate bases form hexa-coordinated adducts with 1:2 stoichiometry. The kinetics of oxidation of three aldoses (glucose, mannose and galactose) byN-bromoacetamide (NBA) in the presence of an alkaline solution of RuO4 as catalyst and Hg(OAc)2 as co-catalyst and as a scavenger for bromide have been investigated. The main products of the oxidation are the corresponding
aldonic acids. The reaction is zero order with respect to aldose and OH−. First order dependence of the reaction on both NBA and RuO4 at low concentrations shifts to zero order at higher concentrations. Addition of acetamide decreases the reaction rate, while
addition of Hg(OAc)2 has the opposite effect. No significant effect of ionic strength was observed. OBr− is postulated as the reactive oxidising species and a mechanism involving co-catalysis by RuO4 and HgII is proposed.

A procedure has been developed for the direct fourth-derivative spectrophotometric determination of iron(III) dimethyldithiocarbamate by converting it into an iron(II) 2,2'-bipyridyl complex, which is then dissolved in Triton X-100. Beer's law is obeyed over the concentration range 0.5-20 microg mL(-1 )in the final solution. Various parameters such as the effect of pH and interference of large number of ions on the determination of ferbam have been studied in detail. The method is sensitive, highly selective and can be used for the determination of ferbam in a commercial sample and in mixtures with various dithiocarbamates (ziram, zineb, maneb, etc.) and from wheat grains. 相似文献

Summary The kinetics of iridium(III)-catalysed oxidation of 1,2-ethanediol and 1,4-butanediol by N-bromoacetamide (NBA) in HClO4 in the presence of [Hg(OAc)2] as a scavenger for Br– have been investigated. The reactions are zero-order with respect to both diols, and first-order in NBA at low NBA concentrations, tending to zero order at high concentrations. The order in IrIII decreases from unity to zero at high iridium(III) concentrations. A positive effect on the oxidation rate is observed for [H+] and [HgII] whereas a negative effect is observed for acetamide and [Cl–]. Ionic strength does not influence the oxidation rate. (H2OBr)+ is postulated as the oxidizing species. A mechanism consistent with the observed kinetic data is proposed. 相似文献
