Measurement of cortisol levels in body fluids is important for monitoring pituitary gland and adrenal functions. To develop a specific and standardized enzyme-linked immunosorbent assay (ELISA), a novel monoclonal anti-cortisol antibody has been generated using a reasonably designed haptenic derivative. Spleen cells were prepared from the BALB/c or A/J mouse, which had repeatedly been immunized with a conjugate of 4-(2-carboxyethylthio)cortisol (CET) and bovine serum albumin, to be fused with P3/NS1/1-Ag4-1 myeloma cells. After four fusion experiments, one hybridoma clone secreting a practical antibody has been established. The resulting monoclonal antibody CS#38 (isotype gamma1, kappa) showed an affinity constant (K(a)) for cortisol of 1 x 10(9) M(-1) and provided a practical calibration curve (detection limit, 0.26 ng per assay) in a homologous ELISA system employing horseradish peroxidase-labeled CET as a labeled antigen. Cross-reactivities with related C-21 steroids were acceptably low: 11-deoxycortisol (4.3%), cortisone (4.0%), corticosterone (1.9%), progesterone (1.6%), 17alpha-hydroxyprogesterone (12%), 6beta-hydroxycortisol (8.4%), and tetrahydrocortisol (< 0.1%). Urinary and serum cortisol levels of healthy volunteers were determined by this method after methylene chloride extraction to be 39.0 +/- 17.0 microg/day (n = 7) and 80.8 +/- 38.9 ng/mL (n = 10), respectively, both of which are in the reference range. 相似文献

Let H 0 and H I be a self-adjoint and a symmetric operator on a complex Hilbert space, respectively, and suppose that H 0 is bounded below and the infimum E 0 of the spectrum of H 0 is a simple eigenvalue of H 0 which is not necessarily isolated. In this paper, we present a new asymptotic perturbation theory for an eigenvalue E(λ) of the operator ${H(\lambda)\,:=\,H_0 + \lambda H_{I}\,(\lambda \in \mathbb{R} \setminus \{0\})}$ satisfying lim λ → 0 E(λ) = E 0. The point of the theory is in that it covers also the case where E 0 is a non-isolated eigenvalue of H 0. Under a suitable set of assumptions, we derive an asymptotic expansion of E(λ) up to an arbitrary finite order of λ as λ → 0. We apply the abstract results to a model of massless quantum fields, called the generalized spin-boson model (Arai and Hirokawa in J Funct Anal 151:455–503, 1997) and show that the ground-state energy of the model has asymptotic expansions in the coupling constant λ as λ → 0. 相似文献

The incompressible flow around bluff bodies (a square cylinder and a cube) is investigated numerically using turbulence models. A non‐linear k–ε model, which can take into account the anisotropy of turbulence with less CPU time and computer memory then RSM or LES, is adopted as a turbulence model. In tuning of the model coefficients of the non‐linear terms are adjusted through the examination of previous experimental studies in simple shear flows. For the tuning of the coefficient in the eddy viscosity (=Cμ), the realizability constraints are derived in three types of basic 2D flow patterns, namely, a simple shear flow, flow around a saddle and a focal point. Cμ is then determined as a function of the strain and rotation parameters to satisfy the realizability. The turbulence model is first applied to a 2D flow around a square cylinder and the model performance for unsteady flows is examined focussing on the period and the amplitude of the flow oscillation induced by Karman vortex shedding. The applicability of the model to 3D flows is examined through the computation of the flow around a surface‐mounted cubic obstacle. Pure rotational spectra of the sodium chloride-water complexes, NaCl-(H(2)O)(n) (n = 1, 2, and 3), in the vibronic ground state have been observed by a Fourier- transform microwave spectrometer coupled with a laser ablation source. The (37)Cl-isotopic species and a few deuterated species have also been observed. From the analyses of the spectra, the rotational constants, the centrifugal distortion constants, and the nuclear quadrupole coupling constants of the Na and Cl nuclei were determined precisely for all the species. The molecular structures of NaCl-(H(2)O)(n) were determined using the rotational constants and the molecular symmetry. The charge distributions around Na and Cl nuclei in NaCl are dramatically changed by the complex formation with H(2)O. Prominent dependences of the bond lengths r(Na-Cl) on the number of H(2)O were also observed. By a comparison with results of theoretical studies, it is shown that the structure of NaCl-(H(2)O)(3) is approaching to that of the contact ion-pair, which is considered to be an intermediate species in the incipient solvation process. 相似文献
