A series of thirty eight 2,4-diaminoquinazolines having diverse substitution patterns on the aromatic ring was evaluated for inhibitory activity against dihydrofolate reductase (DHFR) obtained from a human lymphoblast cell line. Many of these compounds were also evaluated as inhibitors of rat liver DHFR under the same experimental conditions. In most instances the results obtained with each enzyme were comparable indicating that the rodent enzyme is a suitable model for the human DHFR as far as the determination of I50 values is concerned. The results demonstrate that relatively simple 5-substituted- or 5,6-disubstituted-2,4-diaminoquinazolines can be potent DHFR inhibitors. The presence of a nonpolar substituent at position 7 or 8 was highly detrimental to inhibitory potency.  相似文献

The redox system of potassium persulfate–thiomalic acid (I1–I2) was used to initiate the polymerization of acrylamide (M) in aqueous medium. For 20–30% conversion the rate equation is where Rp is the rate of polymerization. Activation energy is 8.34 kcal deg?1 mole?1 in the investigated range of temperature 25–45°C. Mn is directly proportional to [M] and inversely to [I1]. The range of concentrations for which these observations hold at 35°C and pH 4.2 are [I1] = (1.0–3.0) × 10?3, [I2] = (3.0–7.5) × 10?3, and [M] = 5.0 × 10?2–3.0 × 10?1 mole/liter.  相似文献

This paper describes a novel clustering methodology for classifying over 700 conformations of a flexible analogue of GBR 12909, a dopamine reuptake inhibitor that has completed phase I clinical trials as a treatment for cocaine abuse. The major aspect of the clustering methodology includes an efficient data-conditioning scheme where a systematic feature extraction procedure based on the structural properties of the molecule was used to reduce the associated feature space. This allowed region-specific clustering that focused on individual pharmacophore elements of the molecule. For clustering of the reduced feature set, the fuzzy clustering partitional method was utilized. Due to the relational nature of the feature data, fuzzy relational clustering was employed, and it successfully detected natural groups defined by rotational minima around N(sp(3))-C(sp(3)), O(sp(3))-C(sp(3)), and C(sp(3))-C(sp(2)) bonds. The proposed clustering methodology also employed several cluster validity measures, which corroborated the partitions produced by the clustering technique and agreed with the results of hierarchical clustering using the XCluster program. Representative structures which exhibited a reasonable spread of energies and showed good spatial coverage of the conformational space were identified for use as putative bioactive conformations in a future Comparative Molecular Field Analysis of GBR 12909 analogues. The clustering methodology developed here is capable of handling other computational chemistry problems, and the feature extraction technique can be easily generalized to other molecules.  相似文献

Summary Alkaline sodium borohydride has been employed as a reducing reagent for the titrimetric determination of compounds containing the carbonyl function. The sample is dissolved in methanol and is then reduced with alkaline sodium borohydride. After completion of the reaction, the excess sodium borohydride is back titrated against standardized hydrochloric acid solution using methyl red as indicator. The stoichiometry between the carbonyl function and sodium borohydride is 41.

Small-angle x-ray scattering methods were applied to calculate macromolecular parameters like specific inner surface (O/V), length of coherence (lc), and the percentage of air present in agave hybrid, treating it as a densely packed colloidal system belonging to a general micelle system. The parameters were equal to O/V = 8.50 × 10?6 Å?1, lc = 150 Å, and the percentage of air = 0.03. The Kratky camera with latest design was used for the experimental measurement. Because the sample agave ybrid is a natural fiber and a highly oriented system that is celluosic in character, the theories of Kratky and Porod have been used to evaluate these parameters.  相似文献

The kinetics of oxidation of ethyldigol by vanadium(V) in aqueous acidic medium has been carried out. The reaction is first order with respect to vanadium(V) and the substrate and is acid catalysed.Hammett acidity function (H 0) andBunnett hypothesis have been applied. The formation of free radicals during the course of the reaction has been indicated. A probable reaction mechanism is proposed.

Zusammenfassung Es wurde die Kinetik der Oxidation von Ethyldigol mittels Vanadium(V) in wäßriger saurer Lösung untersucht. Die Reaktion ist erster Ordnung bezüglich Vanadium(V) und Substrat und ist säurekatalysiert. Es wurden dieHammett-Aciditätsfunktion (H 0) und dieBunnett-Hypothese angewandt. Die Bildung von freien Radikalen während der Reaktion konnte bestätigt werden. Es wird ein Reaktionsmechanismus vorgeschlagen.

The optimal current difference lattice hydrodynamic model is extended to investigate the traffic flow dynamics on a unidirectional single lane gradient highway. The effect of slope on uphill/downhill highway is examined through linear stability analysis and shown that the slope significantly affects the stability region on the phase diagram.Using nonlinear stability analysis, the Burgers, Korteweg-deVries(KdV) and modified Korteweg-deVries(mKdV) equations are derived in stable, metastable and unstable region, respectively. The effect of reaction coefficient is examined and concluded that it plays an important role in suppressing the traffic jams on a gradient highway. The theoretical findings have been verified through numerical simulation which confirm that the slope on a gradient highway significantly influence the traffic dynamics and traffic jam can be suppressed efficiently by considering the optimal current difference effect in the new lattice model.  相似文献
