This study evaluated the effect of Mucuna pruriens (MP) administration on neuroinflammation and behavioral and murinometric parameters in obese rats. Proximate composition, oligosaccharide and phenolic compound profile of MP were determined. Wistar adult male rats were randomized into healthy (HG) and obese group (OG). The HG consumed a control chow diet while OG consumed a cafeteria diet for eight weeks. Then, they were subdivided into: Healthy (HG); Healthy with MP administration (HGMP); Obese (OG); Obese with MP administration (OGMP), with the consumption of the respective diets remaining for another eight weeks, in addition to gavage with MP extract to supplemented groups (750 mg/kg weight). MP presented a composition rich in proteins and phenolic compounds, especially catechin, in addition to 1-kestose and levodopa. Supplementation reduced food intake, body weight, and thoracic and abdominal circumferences in obese rats. MP showed anxiolytic and antidepressant effects and reduced morphological damage and expression of interleukin 6 in the hippocampus of obese rats. MP treatment showed satietogenic, slimming, anxiolytic and antidepressant effects, besides to minimizing hippocampal neuroinflammation in obese rats. Our results demonstrated the potential anti-obesity of MP which are probably related to the high content of bioactive compounds present in this plant extract. 相似文献

Trimethylsilylation of anabolic agents and their metabolites is frequently achieved by using the derivatization mixture N-methyl-N-(trimethylsilyl)trifluoroacetamide (MSTFA)/NH(4)I/2-mercaptoethanol. Nevertheless, artifacts were formed when this mixture was employed in the monitoring of exemestane and its main metabolite 17β-hydroxyexemestane prior to GC-MS analysis. These artifacts were identified as the N-methyltrifluoroacetamide (MTFA) and trimethylsiloxyethylmercapto products of the respective trimethylsilyl (TMS) derivatives. Furthermore, artifact formation was evaluated taking the structure (1,4-diene-3-keto-6-exomethylene) of the compounds into account. Although these artifacts are relevant for investigations regarding the derivatization process and may be of interest in many fields, they are detrimental to cope with the requirements of the World Anti-Doping Agency (WADA) in terms of the limits of detection (LODs) required. To overcome this issue, a method using an alternative derivatization was proposed: formation of methyloxime-TMS derivatives through double derivatization using O-methylhydroxylamine/pyridine and MSTFA/TMS imidazole after enzymatic hydrolysis and liquid-liquid extraction. Samples from an excretion study after administration of exemestane to healthy volunteers were analyzed by the proposed method and detection of both exemestane and its main metabolite was possible. This method showed excellent results for both analytes meeting the LODs required for antiestrogenic agents (50 ng/mL) established by WADA. The method was validated for the main metabolite, it was robust and cost-effective for qualitative and quantitative purposes, with LOD and LOQ of 10 ng/mL and 25 ng/mL, respectively. 相似文献

Photoelectrochemical cells with TiO2 electrodes to convert sunlight and water into gaseous hydrogen and oxygen are a source of clean and renewable fuel. Despite their great potential, far-from-ideal performance and poor utilization of the solar spectrum have prevented them from becoming a widespread and practical technology. We review recent experimental work that uses dynamics measurements to study limitations of photoelectrochemical cells from a fundamental level and the use of TiO2 nanotube arrays as a superior alternative to TiO2 nanoparticles. Through a combination of nanoscale size control, doping, composite materials, and the incorporation of noble-metal nanoparticles, improved performance and light harvesting are demonstrated. 相似文献

The kinetic behavior of the enzyme laccase in solution and immobilized onto carbon platforms using poly(amido amine) (PAMAM) dendrimers has been investigated. The results with the immobilized enzymes have demonstrated that almost ten times more enzyme on the carbon support is required for satisfactory kinetic rates to be achieved. Furthermore, the study as a function of the substrate concentration revealed that the kinetic behavior of the enzyme in solution fits the Michaelis?CMenten model. However, when the enzyme is immobilized onto the carbon surface, the catalyzed reaction follows a particular kinetic behavior with apparent positive cooperativity. The highest activity with laccase (in solution or immobilized) is achieved around pH?4.5, and the substrate conversion rate clearly diminishes with rising pH. The optimum temperature lies around 60?°C. The enzyme displays good catalytic activity in a wide range of pH and temperature values. The stability tests evidenced that there is no appreciable reduction in the enzymatic activity after immobilization within the first 30?days. Taking into account both the kinetic and stability tests, one can infer that the use of PAMAM dendrimers seems to be a very attractive approach for the immobilization of enzymes, as well as a feasible and useful methodology for the anchoring of enzymes with potential application in many biotechnological areas. 相似文献

Doping control screening based on the enhanced resolution of comprehensive two-dimensional (2D) gas chromatography hyphenated to time of flight mass spectrometer was investigated. The identification of anabolic agents (clenbuterol, norandrosterone, epimetendiol, two methyltestosterone metabolites and 3′-hydroxystanozolol) contained in a spiked urine sample (2 ng/ml) was demonstrated. Special emphasis was given to 3′-hydroxystanozolol, mainly considering the difficulty in its detection. In contrast to conventional GC–MS approaches that must use single-ion monitoring, the GC × GC–TOFMS method enabled the identification of that metabolite through the deconvolution of the full mass spectrum and also resolved the co-eluted peaks of 3′-hydroxystanozolol and an endogenous component. 相似文献

HTHRGC was found essential for optimizing representative preparation of high molecular weight samples and enabling fingerprinting of heavy fractions of crude oils and associated paraffinic deposits in production pipelines. Laboratory experiments aimed at simulating the process of paraffin deposition could also be easily evaluated. The Fe(II) of the binuclear Fe(II)Fe(III) active site of pig purple acid phosphatase (uteroferrin) has been replaced in turn by five M(II) ions (Mn(II), Co(II), Ni(II), Cu(II), and Zn(II)). An uptake of 1 equiv of M(II) is observed in all cases except that of Cu(II), when a second more loosely bound Cu(II) is removed by treatment with edta. The products have been characterized by different analytical procedures and by UV-vis spectrophotometry. At 25 degrees C, I = 0.100 M (NaCl), the nonenzymatic reactions with H(2)PO(4)(-) give the mu-phosphato product, and formation constants K/M(-1) show an 8-fold spread at pH 4.9 of 740 (Mn), 165 (Fe), 190 (Co), 90 (Ni), 800 (Cu), 380 (Zn). The variations in K correlate well with stability constants for the complexing of H(2)PO(4)(-) and (CH(3)O)HPO(3)(-) with M(II) hexaaqua ions. At pH 4.9 with [H(2)PO(4)(-)] > or = 3.5 mM rate constants k(obs) decrease, and an inhibition process in which a second [H(2)PO(4)(-)] coordinates to the dinuclear center is proposed. The mechanism considered accounts for most but not all of the features displayed. Thus K(1) values for the coordination of phosphate to M(II) are in the range10-60 M(-1), whereas K(2) values for the bridging of the phosphate to Fe(III) are in the narrower range 7.8-12.4. From the fits described K(i) approximately 10(3) M(-1) for the inhibition step, which is independent of the identity of M(II). Values of k(obs) decrease with increasing pH, giving pK(a) values which are close to 3.8 and independent of M(II) (Fe(II), Zn(II), Mn(II)). The acid dissociation process is assigned to Fe(III)-OH(2) to Fe(III)-OH(-), where OH(-) is less readily displaced by phosphate. 相似文献

By a combination of theoretical and experimental design, we probed the effect of a quasi‐single electron on the surface plasmon resonance (SPR)‐mediated catalytic activities of Ag nanoparticles. Specifically, we started by theoretically investigating how the E‐field distribution around the surface of a Ag nanosphere was influenced by static electric field induced by one, two, or three extra fixed electrons embedded in graphene oxide (GO) next to the Ag nanosphere. We found that the presence of the extra electron(s) changed the E‐field distributions and led to higher electric field intensities. Then, we experimentally observed that a quasi‐single electron trapped at the interface between GO and Ag NPs in Ag NPs supported on graphene oxide (GO‐Ag NPs) led to higher catalytic activities as compared to Ag and GO‐Ag NPs without electrons trapped at the interface, representing the first observation of catalytic enhancement promoted by a quasi‐single electron. 相似文献
