Tiopronin, formally 2‐mercaptopropionylglycine (MPG), is currently prescribed to treat cystinuria and rheumatoid arthritis, and its antioxidant properties have led to its investigation as a treatment for cataracts, a condition in which oxidative stress is strongly implicated. To study its accumulation in the eye, a reliable, isocratic HPLC method was developed for the determination of MPG and its primary metabolite 2‐mercaptopropionic acid (MPA) in plasma and relevant ocular tissues. This method utilizes pre‐column derivatization and fluorescence detection. The 3.5 min separation enables high‐throughput analysis, and validation experiments demonstrated that this method is suitable for evaluating ocular accumulation of MPG and MPA at concentrations as low as 66 and 33 nm , respectively. Excellent linearity was achieved over the working concentration range with R2 > 0.997. Extraction recovery was reproducible within each matrix and exceeded 97%. Accuracy was within 13.3% relative error, and intra‐ and inter‐day precisions were within 6% CV and 7% CV, respectively. Sample stability was demonstrated under various storage conditions, and the use of an internal standard conferred exceptional ruggedness. This method has been successfully applied for the determination of MPG and MPA in plasma, cornea, lens and retina following intraperitoneal administration of the drug in Wistar rats.  相似文献

As part of an ongoing effort to expand the genetic alphabet for in vitro and eventual in vivo applications, we have synthesized a wide variety of predominantly hydrophobic unnatural base pairs and evaluated their replication in DNA. Collectively, the results have led us to propose that these base pairs, which lack stabilizing edge‐on interactions, are replicated by means of a unique intercalative mechanism. Here, we report the synthesis and characterization of three novel derivatives of the nucleotide analogue d MMO2 , which forms an unnatural base pair with the nucleotide analogue d 5SICS . Replacing the para‐methyl substituent of d MMO2 with an annulated furan ring (yielding d FMO ) has a dramatically negative effect on replication, while replacing it with a methoxy (d DMO ) or with a thiomethyl group (d TMO ) improves replication in both steady‐state assays and during PCR amplification. Thus, d TMO –d 5SICS , and especially d DMO –d 5SICS , represent significant progress toward the expansion of the genetic alphabet. To elucidate the structure–activity relationships governing unnatural base pair replication, we determined the solution structure of duplex DNA containing the parental d MMO2 –d 5SICS pair, and also used this structure to generate models of the derivative base pairs. The results strongly support the intercalative mechanism of replication, reveal a surprisingly high level of specificity that may be achieved by optimizing packing interactions, and should prove invaluable for the further optimization of the unnatural base pair.  相似文献

Ni(III)-intermediates are trapped by EPR and UV/Vis spectroscopy in the reaction of a Ni(II) salt with mCPBA. On the basis of their oxo-transfer and hydrogen-atom abstraction abilities the intermediates are assigned as the elusive terminal Ni(III)-oxo/hydroxo species. The findings suggest that Ni(III)-O(H) moieties are viable reactants in oxidation catalysis.  相似文献

Herein, we report the formation of a highly reactive nickel–oxygen species that has been trapped following reaction of a NiII precursor bearing a macrocyclic bis(amidate) ligand with meta‐chloroperbenzoic acid (HmCPBA). This compound is only detectable at temperatures below 250 K and is much more reactive toward organic substrates (i.e., C?H bonds, C?C bonds, and sulfides) than previously reported well‐defined nickel–oxygen species. Remarkably, this species is formed by heterolytic O?O bond cleavage of a Ni–HmCPBA precursor, which is concluded from experimental and computational data. On the basis of spectroscopy and DFT calculations, this reactive species is proposed to be a NiIII–oxyl compound.  相似文献

We report the synthesis and characterization of sugar-containing microspheres consisting of poly(divinylbenzene) (PDVB) cores onto which chains of galactose- or mannose-bearing polymers have been grafted. PDVB particles prepared by distillation polymerization with a diameter of 2.4 μm containing residual surface vinyl groups were used as starting material. “Grafting from”, “grafting through” and “grafting to” techniques were performed and special interest was laid towards the resulting grafting densities. The surface modification via “grafting from” was conducted by reversible addition fragmentation chain transfer (RAFT) polymerization directly from the surface, whereas thiol-ene chemistry was used to affix glycopolymer chains onto the particle surface. The resulting sugar-covered microspheres were analyzed towards their protein recognition activity with a series of lectins.  相似文献

The products and dynamics of the reactions (18)O((3)P)+NO(2) and (18)O((1)D)+NO(2) have been investigated using crossed beams and provide new constraints on the structures and lifetimes of the reactive nitrogen trioxide intermediates formed in collisions of O((3)P) and O((1)D) with NO(2). For each reaction, two product channels are observed - isotope exchange and O(2)+NO formation. From the measured product signal intensities at collision energies of ~6 to 9.5 kcal∕mol, the branching ratio for O(2)+NO formation vs. isotope exchange for the O((3)P)+NO(2) reaction is 52(+6∕-2)% to 48(+2∕-6)%, while that for O((1)D)+NO(2) is 97(+2∕-12)% to 3(+12∕-2)%. The branching ratio for the O((3)P)+NO(2) reaction derived here is similar to the ratio measured in previous kinetics studies, while this is the first study in which the products of the O((1)D)+NO(2) reaction have been determined experimentally. Product energy and angular distributions are derived for the O((3)P)+NO(2) isotope exchange and the O((1)D)+NO(2)→O(2)+NO reactions. The results demonstrate that the O((3)P)+NO(2) isotope exchange reaction proceeds by an NO(3)? complex that is long-lived with respect to its rotational period and suggest that statistical incorporation of the reactant (18)O into the product NO(2) (apart from zero point energy isotope effects) likely occurs. In contrast, the (18)O((1)D)+NO(2)→O(2)+NO reaction proceeds by a direct "stripping" mechanism via a short-lived (18)O-O-NO? complex that results in the occurrence of (18)O in the product O(2) but not in the product NO. Similarly, (18)O is detected in O(2) but not NO for the O((3)P)+NO(2)→O(2)+NO reaction. Thus, even though the product energy and angular distributions for O((3)P)+NO(2)→O(2)+NO derived from the experimental data are uncertain, these results for isotope labeling under single collision conditions support previous kinetics studies that concluded that this reaction proceeds by an asymmetric (18)O-O-NO? intermediate and not by a long-lived symmetric NO(3)? complex, as earlier bulk isotope labeling experiments had concluded. Applicability of these results to atmospheric chemistry is also discussed.  相似文献
