Two simple and fast C18 and HILIC liquid chromatography–electrospray mass spectrometry methods for the determination of hyaluronic acid (HA) in a mucoadhesive chitosan-based formulation were developed and validated. The performances of both methods were compared in terms of validation parameters and matrix effect. A simple sample preparation method based on sulphuric acid-based degradation was optimized for the detection of HA fragments (i.e. m/z 380 2-mer, m/z 759 4-mer, m/z 1,138 8-mer and m/z 1,518 16-mer). By operating under selected ion-monitoring mode, excellent selectivity towards chitosan fragments was obtained. For validation, good linearity, detection limits (<4 μg mL−1) and precision (RSD % < 16 %) were generally obtained on matrix with both columns. However, HILIC column exhibited improved performances in terms of HA fragment separation and selectivity. By analyzing on the C18 column the chitosan-based formulation and sample extracts from pig mucosa treated with the formulation, matrix effects exhibited a dependence of signal suppression degree (ranging from 37 to 83 %) as a function of the HA fragment dimension. The HILIC column afforded instead a significantly reduced suppression degree (ranging from 1 to 16 %) and a better separation. These findings demonstrated the improved performances of the HILIC column with respect to conventional C18 mechanism for the analysis of HA fragments in complex matrices.
The de novo synthesis of piperidine nucleosides from our homologating agent 5,6-dihydro-1,4-dithiin is herein reported. The structure and conformation of nucleosides were conceived to faithfully resemble the well-known nucleoside drugs Immucillins H and A in their bioactive conformation. NMR analysis of the synthesized compounds confirmed that they adopt an iminosugar conformation bearing the nucleobases and the hydroxyl groups in the appropriate orientation. 相似文献
The reactions of 4,5,6,7‐tetrathiocino‐[1,2‐b:3,4‐b′]‐1,3,8,10‐tetrasubstituted‐diimidazolyl‐2,9‐dithiones (R2,R′2‐todit; 1 : R=R′=Et; 2 : R=R′=Ph; 3 : R=Et, R′=Ph) with Br2 exclusively afforded 1:1 and 1:2 “T‐shaped” adducts, as established by FT‐Raman spectroscopy and single‐crystal X‐ray diffraction in the case of complex 1? 2 Br2. On the other hand, the reactions of compounds 1 – 3 with molecular I2 provided charge‐transfer (CT) “spoke” adducts, among which the solvated species 3? 2 I2 ? (1?x)I2 ? x CH2Cl2 (x=0.94) and ( 3 )2 ? 7 I2 ? x CH2Cl2, (x=0.66) were structurally characterized. Mushrooms can be considered a valuable source of natural bioactive compounds with potential polypharmacological effects due to their proven antimicrobial, antiviral, antitumor, and antioxidant activities. In order to identify new potential anticancer compounds, an in-house chemical database of molecules extracted from both edible and non-edible fungal species was employed in a virtual screening against the isoform 7 of the Histone deacetylase (HDAC). This target is known to be implicated in different cancer processes, and in particular in both breast and ovarian tumors. In this work, we proposed the ibotenic acid as lead compound for the development of novel HDAC7 inhibitors, due to its antiproliferative activity in human breast cancer cells (MCF-7). These promising results represent the starting point for the discovery and the optimization of new HDAC7 inhibitors and highlight the interesting opportunity to apply the “drug repositioning” paradigm also to natural compounds deriving from mushrooms. 相似文献
Vincent van Gogh used fugitive red lake pigments that have faded in some paintings. Mapping their distribution is key to understanding how his paintings have changed with time. While red lake pigments can be identified from microsamples, in situ identification and mapping remain challenging. This paper explores the ability of molecular fluorescence imaging spectroscopy to identify and, more importantly, map residual non‐degraded red lakes. The high sensitivity of this method enabled identification of the emission spectra of eosin (tetrabromine fluorescein) lake mixed with lead or zinc white at lower concentrations than elemental X‐ray fluorescence (XRF) spectroscopy used on account of bromine. The molecular fluorescence mapping of residual eosin and two carmine red lakes in van Gogh's The Olive Orchard is demonstrated and compared with XRF imaging spectroscopy. The red lakes are consistent with the composition of paint tubes known to have been used by van Gogh. 相似文献
The orientation of the microdomains at the free surface of solvent-cast films of a polystyrene-block-polybutadiene-block-polystyrene triblock copolymer with cyclindrical morphology was studied by coupling transmission electron and scanning force microscopies (SFM-TEM). It was found that the cylinders of polystyrene, which are assembled in grains randomly oriented in the bulk, tend to reorient with the main axis perpendicular to the surface. SFM investigation indicates that the hexagonal symmetry of the cylinders is also maintained at the free surface which is characterized by protrusions and corrugation with size and characteristic distances closely related to the cylindrical morphology visible in the bulk. Good quantitative agreement between TEM and SFM measurements is observed. 相似文献
A new formulation of a pomegranate-peel extract (PEm) obtained by PUAE (Pulsed Ultrasound-Assisted Extraction) and titrated in both ellagic acid (EA) and punicalagin is proposed, characterized and then analyzed for potential health properties in mice suffering from the experimental autoimmune encephalomyelitis (EAE). PEm effects were compared to those elicited by a formulation containing EA (EAm). Control and EAE mice were chronically administered EAm and Pem dissolved in the drinking water, starting from the day 10 post-immunization (d.p.i.), with a “therapeutic” protocol to deliver daily 50 mg/kg of EA. Treated EAE mice did not limit their daily access to the beverage, nor did they show changes in body weight, but they displayed a significant amelioration of “in vivo” clinical symptoms. “Ex vivo” histochemical analysis showed that spinal-cord demyelination and inflammation in PEm and EAm-treated EAE mice at 23 ± 1 d.p.i. were comparable to those in the untreated EAE animals, while microglia activation (measured as Ionized Calcium Binding Adaptor 1, Iba1 staining) and astrocytosis (quantified as glial fibrillar acid protein, GFAP immunopositivity) significantly recovered, particularly in the gray matter. EAm and PEm displayed comparable efficiencies in controlling the spinal pathological cellular hallmarks in EAE mice, and this would support their delivery as dietary supplementation in patients suffering from multiple sclerosis (MS). 相似文献
