A simple, rapid, and sensitive liquid chromatography–mass spectrometric (LC–MS) method was developed and validated for the determination of isoniazid in dog plasma. Plasma samples were deproteined with methanol and separated on a C18 column interfaced with a single quadrupole mass spectrometer, using 0.1% formic acid–acetonitrile (91:9 v/v) as mobile phase. Detection was performed by positive electrospray ionization with selected ion monitoring at m/z 138 for isoniazid and 152 for entecavir maleate internal standard. Linearity was obtained over the range of 25–5,000 ng mL?1, with a lower limit of quantification of 25 ng mL?1. The intra- and inter-day precision was less than 2.7% in terms of relative standard deviation. Accuracy, expressed as relative error, ranged from ?2.0 to 8.0%. Plasma samples were analysed within 5 min. The method was successfully applied to the evaluation of the pharmacokinetics of isoniazid in dog plasma.  相似文献

1,10-N,N′-bis-(β-d-ureidoglucopyranosyl)-4,7,13-trioxa-1,10-diazacyclopentadecane is a new, recently synthesized compound, which exhibits complexation ability towards drugs. In the present study various theoretical methods, including molecular mechanics, computer simulations, semiempirical and DFT calculations, are applied to find the lowest energy conformers of this molecule in vacuo and in aqueous solution. For the most stable structures the vibrational frequencies as well as the C and H chemical shifts were computed. Along with the theoretical investigation the IR in the KBr discs and the NMR spectra in water and in pyridine were experimentally recorded. It is shown that in the lowest energy structures the two glucosyl units are placed on the same side of the diazacrown ring with their mutual orientations favoring formation of hydrogen bonds. The “open” structure, in which no such hydrogen bonds can be formed, is found to have much higher energy. The computed and measured IR spectra and NMR chemical shifts are compared and discussed in detail. The most stable structures are analyzed with respect to the possible mechanism of complexation of drugs.  相似文献

Summary The combined application of thermogravimetry reactiongas chromatography and gel-permeation chromatography permits to follow the heat degradation of polyurethane polymers in inert gas, air and water-saturated environment. The examinations give information on the rate of thermal degradation, the individual volatile degradation components, the critical points of the polymer chains and on the change of their molecular-weight distribution. A series of novel nanocomposites constituted of FeCo nanoparticles dispersed in an ordered cubic Im3m mesoporous silica matrix (SBA-16) have been successfully synthesized using the wet impregnation method. SBA-16, prepared using the non-ionic Pluronic 127 triblock copolymer as a structure-directing agent, is an excellent support for catalytic nanoparticles because of its peculiar three-dimensional cage-like structure, high surface area, thick walls, and high thermal stability. Low-angle X-ray diffraction, N2 physisorption, and transmission electron microscopy analyses show that after metal loading, calcination at 500 °C, and reduction in H2 flux at 800 °C, the nanocomposites retain the well-ordered structure of the matrix with cubic symmetry of pores. FeCo alloy nanoparticles with spherical shape and narrow size distribution (4–8 nm) are homogeneoulsy distributed throughout the matrix and they seem in a large extent to be allocated inside the pores.  相似文献

A novel series of ligands for the recombinant human AT2 receptor has been synthesized utilizing a fast and efficient palladium-catalyzed procedure for aminocarbonylation as the key reaction. Molybdenum hexacarbonyl [Mo(CO)6] was employed as the carbon monoxide source, and controlled microwave heating was applied. The prepared N-aryl isoleucine derivatives, encompassing a variety of amide groups attached to the aromatic system, exhibit binding affinities at best with Ki values in the low micromolar range versus the recombinant human AT2 receptor. Some of the new nonpeptidic isoleucine derivatives may serve as starting points for further structural optimization. The presented data emphasize the importance of using human receptors in drug discovery programs.  相似文献

The pH effect of potato, apple, and soil matrices on the chromatographic response of nine pesticides was evaluated. All chromatographic analyses were performed in duplicate on a gas chromatograph with electron capture detection. The matrix effect observed in the chromatographic response of the pesticides was evaluated by comparison. We compared the chromatographic response of each pesticide in pure solvent and in organic extract obtained for the matrices. The organic extracts were obtained by solid–liquid extraction with partition at low temperature. Depending on the matrix pH, a greater or lesser amount of co-extractives can be extracted into the organic phase, which affects the matrix effect. The pH of the samples before the extraction process was modified in order to check their influence on pesticide responses. Statistical analyses involving principal component analysis and marginal means revealed that, in the potato and apple matrices, the co-extractives exerted positive effects on the chromatographic response of the analytes. At lower pH, the extraction of co-extractives from potato and apple was favored, thus increasing the matrix effect for these samples.  相似文献
