Chemical and environmental engineering and biotechnology are among the fields now being transformed by continually increasing levels of automation. Whereas the objective in other sectors of industry is simply to increase efficiency, here considerations of system theory or safety demand a high level of automation. Either the processes are too complex and require multifunctional control with feedback, or an analysis of the safety requirements shows the necessity for a certain degree of redundancy in the safety measures, and for elimination of human error as a risk factor. With regard to quality control, cost-benefit analyses lead to striking conclusions which again indicate the need for highly automated, and above all reliable, systems to eliminate rejects. The crux of any automated system is the measurement and control technology; of central importance is the rapid, reliable, and in some cases continuous, measurement and interpretation of key processes or control variables. For this purpose a wide variety of recording instruments and sensors are used to give as accurate a picture as possible of the state of the system. It is obvious from this that the performance of the control system is critically dependent on the sensors. Errors in the measured quantities can become amplified in the control variables or, in dynamic systems, can lead to undesirable operating conditions. Moreover, as a consequence of great advances in microelectronics, “intelligent sensors” which can calibrate and control themselves will be one of the key technologies of the nineties. Unless fast and immediate information on the true current status of a system is available, microprocessors as control devices react blindly and unpredictably to errors in input information. New discoveries in the fields of electronic, electrochemical, and optical transducers are now being applied in heterogeneous catalysis and surface physics, and in biochemistry (enzymology and immunology); in these fields new chemical sensor principles are being tested, which could revolutionize instrumental methods of molecular analysis in particular, owing to their very favorable cost-performance relationship. This article aims to give an up-to-date overview of the current state of the art in these developments, with emphasis on their importance for analysis and their significance in relation to the chemist's interest in mechanisms for identifying substances.  相似文献

High‐pressure μSR experiments on ferromagnetic nickel and \alpha‐iron and antiferromagnetic chromium are reported. In Ni above 260 K BFermi was found to be proportional to the saturation magnetization, whereas at lower temperatures it is temperature independent apart from a small anomaly below 30 K which is presumably caused by a magnetoelastic interaction. There was no evidence for an occupation of metastable sites by the μ+ below the Curie temperature. By contrast, in \alpha‐Fe the temperature dependence of \curpartialBμ/\curpartialp shows a structure which might be attributed to the occupation of excited muon states at elevated temperatures. High‐pressure zero‐field experiments on Cr performed in the temperature regime between 4.5 K and 8 K revealed a pressure dependence of Bμ as large as \curpartialBμ/\curpartialp=-(89.15\pm 0.06)\times 10-12 T/Pa. In terms of volume dependence a very large negative Grüneisen parameter \gamma =-27 was obtained. This revised version was published online in July 2006 with corrections to the Cover Date.  相似文献

The use of plasmid DNA in gene therapy and genetic vaccination has increased the need for scalable and sustainable production processes. One key challenge for bioprocess engineering is the separation of plasmid DNA from structurally related impurities. Affinity purification procedures allow a highly selective capturing of the target molecule. In this paper, we present the isolation of a his-tagged lac repressor, its non-covalent immobilisation to different matrices and binding of DNA, thus enabling us to screen for combinations of ligands and stationary phases by using a building block principle.  相似文献

The migration of epoxidized soy bean oil (ESBO) from the gasket in the lids of glass jars into foods, particularly those rich in edible oil, often far exceeds the legal limit (60 mg/kg). ESBO was determined through a methyl ester isomer of diepoxy linoleic acid. Transesterification occurred directly in the homogenized food. From the extracted methyl esters, the diepoxy components were isolated by normal-phase LC and transferred on-line to gas chromatography with flame ionization detection using the on-column interface in the concurrent solvent evaporation mode. The method involves verification elements to ensure the reliability of the results for every sample analyzed. The detection limit is 2-5 mg/kg, depending on the food. Uncertainty of the procedure is below 10%.  相似文献

The topic of this paper is a mathematically rigorous derivation of the continuum limit of the magnetic force between two parts of a rigid magnetized body. For this we start from a discrete setting of magnetic dipoles fixed to a scaled Bravais lattice, The limit as l corresponds to the passage to the continuum. The magnetic dipole moments are scaled in such a way that we obtain a finite total magnetic moment per unit volume. Under certain regularity assumptions on the magnetization and the boundaries we derive a force formula in the passage from the discrete setting to the continuum. Compared with a corresponding magnetic-force formula which has been previously discussed in the literature, the limiting force consists of an additional explicit local surface term, which is due to short-range effects and which reflects the lattice approximation of the underlying hypersingular integral.  相似文献

The chloro and azido complexes trans-[PdCl(4-C5NF4)(PiPr3)2] (3) and trans-[Pd(N3)(4-C5NF4)(PiPr3)2] (4) can be prepared by reaction of [PdF(4-C5NF4)(PiPr3)2] (2) with Et3SiCl or MeSiN3, respectively. In contrast, reactions of 2 with Ph3SiH or Me2FSiSiFMe2 give the products of reductive elimination 2,3,5,6-tetrafluoropyridine (5) or 4-(fluorodimethylsilyl)tetrafluoropyridine (6) as well as [Pd(PiPr3)2] (1). In a catalytic experiment, pentafluoropyridine can be converted with Ph3SiH into 5 in 62% yield, when 10% of 2 is employed as catalyst. Treatment of trans-[PdF(4-C5NF4)(PiPr3)2] (2) with Bu3SnCH=CH2 in THF at 50 degrees C results in the formation of [Pd(PiPr3)2] (1) and 4-vinyltetrafluoropyridine (7). Complex 2 is also active as a catalyst towards a Stille cross-coupling reaction of pentafluoropyridine with Bu3SnCH=CH2 to give 4-vinyltetrafluoropyridine (7) with a TON of 6. The molecular structure of the complex 3 has been determined by X-ray crystallography.  相似文献

Two series of tricyclic inhibitors of the serine protease thrombin, imides (+/-)-1-(+/-)-8 and lactams (+/-)-9-(+/-)-13, were analysed to evaluate contributions of orthogonal multipolar interactions with the backbone C=O moiety of Asn98 to the free enthalpy of protein-ligand complexation. The lactam derivatives are much more potent and more selective inhibitors (K(i) values between 0.065 and 0.005 microM, selectivity for thrombin over trypsin between 361- and 1609-fold) than the imide compounds (Ki values between 0.057 and 23.7 microM, selectivity for thrombin over trypsin between 3- and 67-fold). The increase in potency and selectivity is explained by the favorable occupancy of the P-pocket of thrombin by the additional isopropyl substituent in the lactam derivatives. The nature of the substituent on the benzyl ring filling the D pocket strongly influences binding potency in the imide series, with Ki values increasing in the sequence: F < OCH2O < Cl < H < OMe < OH < N(pyr)< Br. This sequence can be explained by both steric fit and the occurrence of orthogonal multipolar interactions with the backbone C[double bond, length as m-dash]O moiety of Asn98. In contrast, the substituent on the benzyl ring hardly affects the ligand potency in the lactam series. This discrepancy was clarified by the comparison of X-ray structures solved for co-crystals of thrombin with imide and lactam ligands. Whereas the benzyl substituents in the imide inhibitors are sufficiently close (< or =3.5 Angstroms) to the C=O group of Asn98 to allow for attractive orthogonal multipolar interactions, the distances in the lactam series are too large (> or =4 Angstroms) for attractive dipolar contacts to be effective.  相似文献
