Prenylated proteins with non-native functionalities are generally very difficult to obtain by recombinant or enzymatic means. The semisynthesis of preparative amounts of prenylated Rab guanosine triphosphatases (GTPases) from recombinant proteins and synthetic prenylated peptides depends largely on the availability of functionalised prenylated peptides corresponding to the proteins' native structure or modifications thereof. Here, we describe and compare solution-phase and solid-phase strategies for the generation of peptides corresponding to the prenylated C terminus of Rab7 GTPase. The solid-phase with utilisation of a hydrazide linker emerges as the more favourable approach. It allows a fast and practical synthesis of pure peptides and gives a high degree of flexibility in their modification. To facilitate the analysis of semisynthetic proteins, the synthesised peptides were equipped with a fluorescent group. Using the described approach, we introduced fluorophores at several different positions of the Rab7 C terminus. The position of the incorporated fluorescent groups in the peptides did not influence the protein-ligation reaction, as the generated peptides could be ligated onto thioester-tagged Rab7. However, it was found that the positioning of the fluorescent group had an influence on the functionality of the Rab7 proteins; analysis of the interaction of the semisynthetic Rab7 proteins with REP (Rab escort protein) and GDI (guanosine diphosphate dissociation inhibitor) molecules revealed that modification of the peptide side chains or of the C-terminal isoprenoid did not significantly interfere with complex formation. However, functionalisation of the C terminus was found to have an adverse effect on complex formation and stability, possibly reflecting low structural flexibility of the Rab GDI/REP molecules in the vicinity of the lipid-binding site.  相似文献

Hydrogenases are H2 converting enzymes that harbor catalytic cofactors in which iron (Fe) ions are coordinated by biologically unusual carbon monoxide (CO) and cyanide (CN) ligands. Extrinsic CO and CN, however, inhibit hydrogenases. The mechanism by which CN binds to [FeFe]-hydrogenases is not known. Here, we obtained crystal structures of the CN-treated [FeFe]-hydrogenase CpI from Clostridium pasteurianum. The high resolution of 1.39 Å allowed us to distinguish intrinsic CN and CO ligands and to show that extrinsic CN binds to the open coordination site of the cofactor where CO is known to bind. In contrast to other inhibitors, CN treated crystals show conformational changes of conserved residues within the proton transfer pathway which could allow a direct proton transfer between E279 and S319. This configuration has been proposed to be vital for efficient proton transfer, but has never been observed structurally.  相似文献

Branched poly(ethylene imine) (bPEI) is frequently used in RNA interference (RNAi) experiments as a cationic polymer for the delivery of small interfering RNA (siRNA) because of its ability to form stable polyplexes that facilitate siRNA uptake. However, the use of bPEI in gene delivery is limited by its cytotoxicity and a need for target specificity. In this work, bPEI is modified with d- fructose to improve biocompatibility and target breast cancer cells through the overexpressed GLUT5 transporter. Fructose-substituted bPEI (Fru−bPEI) is accessible in three steps starting from commercially available protected fructopyranosides and bPEI. Several polymers with varying molecular weights, degrees of substitution, and linker positions on d- fructose (C1 and C3) are synthesized and characterized with NMR spectroscopy, size exclusion chromatography, and elemental analysis. In vitro biological screenings show significantly reduced cytotoxicity of 10 kDa bPEI after fructose functionalization, specific uptake of siRNA polyplexes, and targeted knockdown of green fluorescent protein (GFP) in triple-negative breast cancer cells (MDA-MB-231) compared to noncancer cells (HEK293T).  相似文献

Chemical and environmental engineering and biotechnology are among the fields now being transformed by continually increasing levels of automation. Whereas the objective in other sectors of industry is simply to increase efficiency, here considerations of system theory or safety demand a high level of automation. Either the processes are too complex and require multifunctional control with feedback, or an analysis of the safety requirements shows the necessity for a certain degree of redundancy in the safety measures, and for elimination of human error as a risk factor. With regard to quality control, cost-benefit analyses lead to striking conclusions which again indicate the need for highly automated, and above all reliable, systems to eliminate rejects. The crux of any automated system is the measurement and control technology; of central importance is the rapid, reliable, and in some cases continuous, measurement and interpretation of key processes or control variables. For this purpose a wide variety of recording instruments and sensors are used to give as accurate a picture as possible of the state of the system. It is obvious from this that the performance of the control system is critically dependent on the sensors. Errors in the measured quantities can become amplified in the control variables or, in dynamic systems, can lead to undesirable operating conditions. Moreover, as a consequence of great advances in microelectronics, “intelligent sensors” which can calibrate and control themselves will be one of the key technologies of the nineties. Unless fast and immediate information on the true current status of a system is available, microprocessors as control devices react blindly and unpredictably to errors in input information. New discoveries in the fields of electronic, electrochemical, and optical transducers are now being applied in heterogeneous catalysis and surface physics, and in biochemistry (enzymology and immunology); in these fields new chemical sensor principles are being tested, which could revolutionize instrumental methods of molecular analysis in particular, owing to their very favorable cost-performance relationship. This article aims to give an up-to-date overview of the current state of the art in these developments, with emphasis on their importance for analysis and their significance in relation to the chemist's interest in mechanisms for identifying substances.  相似文献

Reaction of A2CO3 (A = K, Rb) with Sn and Se in an H2O/CH3OH mixture at 115–130°C affords the isotypic selenidostannates(IV) A6Sn4Se11 _. xH2O (A = K, x = 8) 1 and 2 whose discrete [Sn4Se11]6– anions each contain two corner‐bridged ditetrahedral [Sn2Se6]4– species. Similar reaction conditions with A = Cs afford Cs2Sn2Se5 _. H2O ( 3a ) and Cs2Sn2Se5 ( 3b ) in which such [Sn2Se6]4– building blocks are connected through common Se atoms into infinite [Sn2Se52–] chains. The [Sn3Se72–] ribbons of (Et4N)2Sn3Se7 ( 4 ), formed by treating (Et4N)I with Sn and Se in methanol at 130°C, can be regarded as resulting from the condensation of [Sn2Se52–] chains with molecular [SnSe4]4– anions. The anions [Sn4Se11]6–, [Sn2Se52–], and [Sn3Se72–] represent the products of individual reaction steps on the potential condensation pathway of [Sn2Se6]4– to the lamellar selenidostannates(IV) [Sn4Se92–] or [Sn3Se72–].  相似文献
