N-Phenylsulfonyl (S)-proline catalyzes the direct aldol reaction of 3-substituted cyclobutanones and aryl aldehydes in good yield and with excellent diastereoselectivity and enantioselectivity. Seven synthetic analogues of the PXR (pregnane-X-receptor) potent natural agonist solomonsterol A were prepared by total synthesis. Their activity toward PXR was assessed by transactivation and RT-PCR assays. The study discloses cholestan disulfate (8) as a new, simplified agonist of PXR. By in vitro studies on hepatic cells we have demonstrated that this compound is a potent PXR agonist and functional characterization in human macrophages and hepatic stellate cells provided evidence that cholestan disulfate (8) has the ability to modulate the immune response triggered by bacterial endotoxin as well as to counter-activate hepatic stellate cell activation induced by thrombin. Because inhibition of immune-driven circuits might have relevance in the treatment of inflammation and liver fibrosis, the present data support the development of cholestan disulfate (8) in preclinical models of inflammatory diseases.  相似文献

A concise, enantioselective synthesis of the potent dual orexin inhibitor suvorexant (1) is reported. Key features of the synthesis include a mild copper-catalyzed amination, a highly chemoselective conjugate addition, and a tandem enantioselective transamination/seven-membered ring annulation. The use of Fourier transform infrared spectromicroscopy and mass spectrometry (MS) allowed us to characterize the composition of polar and non-polar binders present in sporadic wall paint fragments taken from Pompeii’s archaeological excavation. The analyses of the polar and non-polar binder components extracted from paint powder layer showed the presence of amino acids, sugars, and fatty acids but the absence of proteinaceous material. These results are consistent with a water tempera painting mixture composed of pigments, flours, gums, and oils and are in agreement with those obtained from a simulated wall paint sample made for mimicking an ancient “a secco” technique. Notably, for the first time, we report the capability to discriminate by tandem MS the presence of free amino acids in the paint layer.  相似文献

Poly(ether amide)s containing naphthalene unit were prepared either by the polyaddition reaction of aromatic bis(2‐oxazoline)s with the different dihydroxynaphthalenes or by the homopolyaddition of a monomer containing an oxazoline, a hydroxy, and naphthalene moieties. First, polymerization method represents AA + BB mode where 1,4‐phenylene‐2,2′‐bis(2‐oxazoline) (A) and 1,3‐phenylene‐2,2′‐bis(2‐oxazoline) (B) were used as AA monomers and four different dihydroxynaphthalenes 1–4 were used as BB monomers. In the second case, 2‐(6‐hydroxynaphthalene‐2‐yl)‐2‐oxazoline (5) was used as AB‐type monomer in thermally induced polymerizations. The time dependences of polyadditions in bulk as well as in the solution were examined. The reduction of molar mass was observed after the initial fast increase of molar mass. This can be explained by the presence of side and degradation reactions. In both cases, polyadditions resulted in the linear poly(ether amide)s, which were characterized by 1H and 13C NMR spectroscopy. Thermal properties of the prepared polymers were studied by differential scanning calorimetry (DSC) measurements. Comparison of the temperatures of glass transition for polymers prepared in AA + BB mode shows the strong dependence of thermal properties on the structure of the polymers. The values were in the range of 136–171°C. The glass‐transition temperature (Tg) of poly[2‐(6‐hydroxynaphthalene‐2‐yl)‐2‐oxazoline] prepared by AB‐type polyaddition is 183°C, which corresponds to the higher contents of hard aromatic segments in the latter type of polymers compared to the polymers prepared in the AA + BB‐type polyadditions. Over the past years, LC–MS‐based approaches have gained a growing interest in food analysis by using different platforms and methodologies. In particular, enhanced selectivity and sensitivity of multiple reaction monitoring (MRM) scan function offer powerful capabilities in detecting and quantifying specific analytes within complex mixtures such as food matrices. The MRM approach, traditionally applied in biomedical research, is particularly suitable for the detection of food adulteration and for the verification of authenticity to assure food safety and quality, both recognized as top priorities by the European Union Commission. Increasingly stringent legislation ensure products safety along every step ‘from farm to fork’, especially for traditional foods designed with the Protected Designation of Origin certification. Therefore, there is a growing demand of new methodologies for defining food authenticity in order to preserve their unique traits against frauds. In this work, an ultra performance liquid chromatopgraphy‐electrospray ionization‐tandem mass spectrometry (MS/MS) methodology based on MRM has been developed for the sensitive and selective detection of buffalo mozzarella adulteration. The targeted quantitative analysis was performed by monitoring specific transitions of the phosphorylated β‐casein f33‐48 peptide, identified as a novel species‐specific proteotypic marker. The high sensitivity of MRM‐based MS and the wide dynamic range of triple quadrupole spectrometers have proved to be a valuable tool for the analysis of food matrices such as dairy products, thus offering new opportunities for monitoring food quality and adulterations. Raman spectra were recorded experimentally and calculated theoretically for bithiophene, terthiophene, and quaterthiophene samples as a function of excitation polarization. Distinct spectral signatures were assigned and correlated to the molecular/unit cell orientation as determined by X-ray diffraction. The ability to predict molecular/unit cell orientation within organic crystals using polarized Raman spectroscopy was evaluated by predicting the unit cell orientation in a simulated terthiophene crystal given a random set of simulated polarized Raman spectra. Polarized Raman spectroscopy offers a promising tool to quickly and economically determine the unit cell orientation in known organic crystals and crystalline thin films. Implications of our methodologies for studying individual molecule conformations are discussed.  相似文献

The recently developed correlation consistent Composite Approach for transition metals (ccCA-TM) was utilized to compute the thermochemical properties for a collection of 225 inorganic molecules containing first row (3d) transition metals, ranging from the monohydrides to larger organometallics such as Sc(C(5)H(5))(3) and clusters such as (CrO(3))(3). Ostentatiously large deviations of ccCA-TM predictions stem mainly from aging and unreliable experimental data. For a subset of 70 molecules with reported experimental uncertainties less than or equal to 2.0 kcal mol(-1), regardless of the presence of moderate multireference character in some molecules, ccCA-TM achieves transition metal chemical accuracy of ±3.0 kcal mol(-1) as defined in our earlier work [J. Phys. Chem. A2007, 111, 11269-11277] by giving a mean absolute deviation of 2.90 kcal mol(-1) and a root-mean-square deviation of 3.91 kcal mol(-1). As subsets are constructed with decreasing upper limits of reported experimental uncertainties (5.0, 4.0, 3.0, 2.0, and 1.0 kcal mol(-1)), the ccCA-TM mean absolute deviations were observed to monotonically drop off from 4.35 to 2.37 kcal mol(-1). In contrast, such a trend is missing for DFT methods as exemplified by B3LYP and M06 with mean absolute deviations in the range 12.9-14.1 and 10.5-11.0 kcal mol(-1), respectively. Salient multireference character, as demonstrated by the T(1)/D(1) diagnostics and the weights (C(0)(2)) of leading electron configuration in the complete active self-consistent field wave function, was found in a significant amount of molecules, which can still be accurately described by the single reference ccCA-TM. The ccCA-TM algorithm has been demonstrated as an accurate, robust, and widely applicable model chemistry for 3d transition metal-containing species with versatile bonding features.  相似文献
