Radiolabeled GX1 Peptide for Tumor Angiogenesis Imaging

Early and accurate detection of primary or metastatic tumors is of great value in staging, treatment management, and prognosis. Tumor angiogenesis plays an essential role in the growth, invasion, and metastatic spread of solid cancers, and so, is a promising approach for tumor imaging. The GX1 (CGNSNPKSC) peptide was identified by phage display library and has been investigated as a marker for human cancers. This study aims to evaluate the ^99mTc-HYNIC-PEG₄-c (GX1) as a biomarker for tumor imaging. Our results showed that GX1 specifically binds to tumor cells in vitro. SKMEL28 and MDA-MB231 cells achieved total binding peak at 60 min of incubation. For B16F10 and MKN45 cells, the total and specific binding were similar during all time points, while A549 cell line showed rapid cellular total uptake of the tracer at 30 min of incubation. Biodistribution showed low non-specific uptakes and rapid renal excretion. Melanoma tumors showed enhanced GX1 uptake in animal model at 60 min, and it was significantly blocked by cold peptide. The radiotracer showed tumor specificity, especially in melanomas that are highly vascularized tumors. In this sense, it should be considered in future studies, aiming to evaluate degree of angiogenesis, progression, and invasion of tumors.     

Palladium nanoparticles stabilized by an ionic polymer and ionic liquid: a versatile system for C-C cross-coupling reactions

Highly stable palladium nanoparticles (Pd NPs), protected by an imidazolium-based ionic polymer (IP) in a functionalized ionic liquid (IL), have been prepared and characterized by transmission electron microscopy (TEM). These Pd NPs are excellent precatalysts for Suzuki, Heck, and Stille coupling reactions and can be stored without undergoing degradation for at least 2 years. The NP-IP-IL system may therefore be considered as an alternative to the traditional palladium on carbon (Pd/C) precatalyst employed in many C-C coupling reactions, also allowing reactions to be conducted under "solvent-free" conditions.     

Crystal structure and photochemical behavior in solution of the 3'-N-sulfamate analogue of thymidylyl(3'-5')thymidine

The 3'-N-sulfamate analogue of thymidylyl(3'-5')thymidine (TnsoT, 1) exhibits a preference for a C3'-endo conformation in the solution and solid states. Its photochemical behavior in solution is compared to that of its natural counterpart, thymidylyl(3'-5')thymidine (TpT, 2), to get further insight into the significance of the C3'-endo conformation on the photoproduct formation at the single-stranded dinucleotide level. Irradiation at 254 nm of 1 led to the same type of photoproducts as observed with 2. However, 1 was significantly more photoreactive than 2, and accordingly, the initial rate of photoproduct formation was enhanced in accordance with its propensity to base stack compared to 2. The corresponding quantum yields were determined and showed that the enhancement factor (1 compared to 2) is moderate for the cyclobutane pyrimidine dimer (CPD) (1.26) and much higher for the (6-4) photoproduct (1.8). These data strongly suggest that the CPD and (6-4) photoproduct arise from distinct minor stacked conformations.     

Pd-catalyzed, Cu(I)-mediated cross-couplings of bisarylthiocyclobutenediones with boronic acids and organostannanes

Bisarylthiocyclobutenedione 7 reacted smoothly with a variety of both organostannanes and (hetero)arylboronic acids in the presence of a catalytic amount of Pd and a stoichiometric amount of CuTC to produce symmetrical disubstituted cyclobutenediones in yields that range from 37 to 94% (18 examples).

Identification of Cavities Inside Two-Dimensional Heterogeneous Isotropic Elastic Bodies

In this paper, we discuss the uniqueness in determining cavities (i.e., nonrectilinear cracks) in a heterogeneous isotropic elastic medium in two dimensions. Our main result asserts that there is at most one cavity in the elastic medium which yields the same surface displacements and stresses on an arbitarily small portion of the boundary. The boundaries of cavities are assumed to be piecewise smooth and admit edges where no net force is exerted. The key of the proof is the unique continuation for the isotropic Lamé system and geometric considerations. This revised version was published online in August 2006 with corrections to the Cover Date.
