The α-D -allo-diol 9 possesses an intramolecular H-bond (HO? C(3) to O? C(1)) in solution and in the solid state (Fig. 2). In solution, it exists as a mixture of the tautomers 9a and 9b (Fig. 3), which possess a bifurcated H-bond, connecting HO? C(2) with both O? C(1) and O? C(3). In addition, 9a possesses the same intramolecular H-bond as in the solid state, while 9b is characterized by an intramolecular H-bond between HO? C(3) and O? C(4). In solution, the β-D -anomer 12 is also a mixture of tautomers, 12a and presumably a dimer. The H-bonding in 9 and 12 is evidenced by their IR and 1H-NMR spectra and by a comparison with those of 3–8, 10 , and 11 . The expected regioselectivity of glycosidation of 9 and 12 by the diazirine 1 or the trichloroacetimidate 2 is discussed on the basis of the relative degree of acidity/nucleophilicity of individual OH groups, as governed by H-bonding. Additional factors determining the regioselectivity of glycosidation by 1 are the direction of carbene approach/proton transfer by H-bonded OH groups, and the stereoelectronic control of both the proton transfer to the alkoxy-alkyl carbene (in the σ-plane) and the combination of the thereby formed ions (π-plane of the oxycarbenium ion). Glycosidation of 9 by the diazirine 1 or the trichloroacetimidate 2 proceeded in good yields (75–94%) and with high regioselectivity. Glycosidation of 9 and 12 by 1 or 2 gave mixtures of the disaccharides 14–17 and 18–21 , respectively (Scheme 2). As expected, glycosidation of 12 by 1 or by 2 gave a nearly 1:1 mixture of regioisomers and a slight preference for the β-D -anomers (Table 4). Glycosidation of the α-D -anomer 9 gave mostly the 1,3-linked disaccharides 16 and 17 (α-D β-D ) along with the 1,2-linked disaccharides 14 and 15 (α-D < β-D , 1,2-/1,3-linked glycosides ca. 1:4), except in THF and at low temperature, where the β-D -configurated 1,2-linked disaccharide 15 is predominantly formed. Similarly, glycosidation of 9 with 2 yielded mainly the 1,3-linked disaccharides (1,2-/1,3-linked products ca. 1:3 and α-D /β-D ca. 1:4). Yields and selectivity depend upon the solvent and the temperature. The regioselectivity and the unexpected stereoselectivity of the glycosidation of 9 by 1 evidences the combined effect of the above mentioned factors, which also explain the lack of regio-complementarity in the glycosidation of 9 by 1 and by 2 (Scheme 3). THF solvates the intermediate oxycarbenium ion, as evidenced by the strong influence of this solvent on the regio- and stereoselectivity, particularly at low temperatures, where kinetic control leads to a stereoelectronically preferred axial attack of THF on the oxycarbenium ion.  相似文献

A liquid chromatographic/tandem mass spectrometric method was developed and validated for the quantitation of capecitabine and its metabolite 5-fluorouracil in human plasma. The simultaneous determination of both analytes was achieved by a column switching method using a trapping column and two analytical columns with different stationary phases. Isocratic elution was used for the separation of capecitabine on a C18 column whereas 5-fluorouracil was separated using gradient elution on an non-polar carbon phase. The calibration curves were linear for both compounds with a correlation factor (R2) > 0.9993 for 5-fluorouracil and >0.9942 for capecitabine. The assay was validated in the concentration range 5.00-1000 ng ml(-1) for both compounds. The intra-day precision was better than 10% for 5-fluorouracil and better than 11% for capecitabine whereas the inter-day precision was better than 8% for 5-fluorouracil and better than 14% for capecitabine.  相似文献

Addition of dialkyl phosphites to the nitrone 6 , formed in situ from the oxime 5 and formaldehyde gave the hydroxylamines 7 (86%) and 8 (88%), which reacted with p-benzoquinone in the presence of ethylene via the C-dialkoxyphosphonoylnitrones 9 and 10 to yield the cycloaddition products 11 – 14 (80–85%) with a diastereoselectivity of about 50%. The cycloaddition products were transformed into the monoisopropylidene derivatives 15 – 18 and the diacetates 19 – 22 . Comparison of the NMR spectra and the specific rotations of the compounds 19 – 22 with those of the corresponding α-ammo-acid derivatives 23 – 26 of known configuration indicated preferential formation of the L -isomers. The cycloaddition products were transformed in good yield into the L -α-aminophosphonic acids 29 , 30 , 36 , and 39 .  相似文献

The chromophore structures in the parent states Pr and Pfr as well as in the photocycle intermediate Lumi-R of oat phytochrome phyA are determined by comparison of the experimental resonance Raman spectra with calculated Raman spectra that have been obtained by density functional theory calculations (B3LYP) using scaled force fields. The spectra were calculated for various tetrapyrrole geometries including more than twenty different methine bridge isomers. For the parent states Pr and Pfr the best agreement in terms of vibrational frequencies, isotopic shifts, and Raman intensities was achieved with the ZZZasa and ZZEssa geometry, respectively. For the first intermediate Lumi-R, the chromophore geometry is concluded to be the ZZEasa configuration. These finding imply that the primary step of the photoactivation of phytochrome is the Z/E isomerization of the C-D methine bridge double bond, whereas the single bond remains in the anti conformation. The subsequent transition to the physiologically active state Pfr includes a (partial) single bond rotation of the A-B methine bridge.  相似文献

The density of the 2-chloroethanol (CE) + 2-methoxyethanol (ME) + 1,2-dimethoxye- thane (DME) ternary mixtures has been measured at different temperatures ranging from –10 to 80°C, and over the entire composition range. The experimental data have been used to check the validity of some relationships accounting for the dependence of the density on temperature and composition domains. Starting from the primary data, some derived quantities, such as excess molar volumes V E, partial molar volumes and partial excess molar volumes , have been obtained. In these mixtures, V E is always positive for the [CE(1) + ME(2)] binaries, while it is generally negative at all other experimental conditions, showing the greatest deviations along the binary axes corresponding to the binary subsystems in the sequence [CE(1) + DME(2)] < [CE(1) + ME(2)] < [ME(1) + DME(2)]. The results are compared and discussed to in terms of changes in molecular association and structural effects in these solvent systems.  相似文献

The glucose‐, mannose‐, and galactose‐derived spirocyclic cyclopropylammonium chlorides 1a – 1d, 2a – 2d and 3a – 3d were prepared as potential glycosidase inhibitors. Cyclopropanation of the diazirine 5 with ethyl acrylate led in 71% yield to a 4 : 5 : 1 : 20 mixture of the ethyl cyclopropanecarboxylates 7a – 7d , while the Cu‐catalysed cycloaddition of ethyl diazoacetate to the exo‐glycal 6 afforded 7a – 7d (6 : 2 : 5 : 3) in 93–98% yield (Scheme 1). Saponification, Curtius degradation, and subsequent addition of BnOH or t‐BuOH led in 60–80% overall yield to the Z‐ or Boc‐carbamates 11a – 11d and 12a – 12d , respectively. Hydrogenolysis of 11a – 11d afforded 1a – 1d , while 12a – 12d was debenzylated to 13a – 13d prior to acidic cleavage of the N‐Boc group. The manno‐ and galacto‐isomers 2a – 2d and 3a – 3d , respectively, were similarly obtained in comparable yields (Schemes 2 and 4). Also prepared were the differentially protected manno‐configured esters 24a – 24d ; they are intermediates for the synthesis of analogous N‐acetylglucosamine‐derived cyclopropanes (Scheme 3). The cyclopropylammonium chlorides 1a – 1d, 2a – 2d and 3a – 3d are very weak inhibitors of several glycosidases (Tables 1 and 2). Traces of Pd compounds, however, generated upon catalytic debenzylation, proved to be strong inhibitors. PdCl is, indeed, a reversible, micromolar inhibitor for the β‐glucosidases from C. saccharolyticum and sweet almonds (non‐competitive), the β‐galactosidases from bovine liver and from E. coli (both non‐competitive), the α‐galactosidase from Aspergillus niger (competitive), and an irreversible inhibitor of the α‐glucosidase from yeast and the α‐galactosidase from coffee beans. The cyclopropylamines derived from 1a – 1d or 3a – 3d significantly enhance the inhibition of the β‐glucosidase from C. saccharolyticum by PdCl , lowering the Ki value from 40 μM (PdCl ) to 0.5 μM for a 1 : 1 mixture of PdCl and 1d . A similar effect is shown by cyclopropylamine, but not by several other amines.  相似文献

The use of high-performance ion chromatography (HPIC) for the determination of N-nitrosoglyphosate [N-nitroso-N-(phosphonomethyl)glycine] is reported. NMR, spectrophotometric and electroanalytical measurements showed that two conformers of N-nitrosoglyphosate in slow equilibrium are always present. Separation of these conformers is achieved by using appropriate chromatographic conditions. The conductimetric detection of N-nitrosoglyphosate and glyphosate and application to the determination of traces of N-nitrosoglyphosate in complex matrices are also reported.  相似文献

The reaction of 3-benzoyl-5-perfluoroalkyl-1,2,4-oxadiazoles with hydrazine has been investigated, evidencing the possibility of competitive reaction paths. Nucleophilic addition of the hydrazine to the electrophilic C(5) of the 1,2,4-oxadiazole ring, followed by ring opening and ring closure with enlargement, leads with high yield and in very mild experimental conditions to the formation of Z-oximes of 3-perfluoroalkyl-6-phenyl-2H-1,2,4-triazin-5-ones (11a-c) as major products of the reaction. In turn, the hydrazine can attack the electrophilic carbonyl carbon giving 4-perfluoroacylamino-5-phenyl-2H-1,2,3-triazoles (13a-c) through the well-known Boulton-Katritzky rearrangement of the intermediate hydrazones.  相似文献

The relative permittivities for the ternary 1,2-ethanediol (component 1) + 2-methoxyethanol (2) + water (3) solvent system have been measured for 66 mixtures covering the whole mole fraction composition 0X1/X2/X3 1 range at –10, –5 and 0 °C. The experimental data were used to test some empirical relations stating the dependence of = (X1, X2, X3). A comparison between the calculated and experimental data show that these equations can be usefully employed to predict values in correspondence of the experimental data gaps.  相似文献
