Cationic nitrile complexes and neutral halide and cyanide complexes, with the general formula [MnL1L2(NO)(eta-C5H4Me)]z, undergo one-electron oxidation at a Pt electrode in CH2Cl2. Linear plots of oxidation potential, Eo', vs. nu(NO) or the Lever parameters, EL, for L1 and L2, allow Eo' to be estimated for unknown analogues. In the presence of TlPF6, [MnIL'(NO)(eta-C5H4Me)] reacts with [Mn(CN)L(NO)(eta-C5H4Me)] to give [(eta5-C5H4Me)(ON)LMn(mu-CN)MnL'(NO)(eta5-C5H4Me)][PF6] which undergoes two reversible one-electron oxidations; DeltaE, the difference between the potentials for the two processes, differs significantly for stable cyanide-bridged linkage isomers. Novel pentametallic complexes such as [Mn[(mu-NC)Mn(CNBut)(NO)(eta5-C5H4Me)]4(OEt2)][PF6]2 and [Mn[(mu-NC)Mn(CNXyl)(NO)(eta5-C5H4Me)]4(NO3-O,O')][PF6], containing a trigonal bipyramidal and a distorted octahedral Mn(II) centre, respectively, result either from slow decomposition of the binuclear cyanide-bridged species or from the reaction of anhydrous MnI2 with four equivalents of [Mn(CN)L(NO)(eta5-C5H4Me)] in the presence of TlPF6.  相似文献

Elemental selenium generated by the photobleaching of selenomerocyanine dyes forms conjugates with serum albumin and serum lipoproteins that are toxic to leukemia and selected solid tumor cells but well tolerated by normal CD34-positive hematopoietic stem and progenitor cells. Serum albumin and lipoproteins act as Trojan horses that deliver the cytotoxic entity (elemental selenium) to tumor cells as part of a physiological process. They exploit the fact that many tumors have an increased demand for albumin and/or low-density lipoprotein. Se(0)-protein conjugates are more toxic than selenium dioxide, sodium selenite, selenomethionine, or selenocystine. They are only minimally affected by drug resistance mechanism, and they potentiate the cytotoxic effect of ionizing radiation and several standard chemotherapeutic agents. The cytotoxic mechanism of Se(0)-protein conjugates is not yet fully understood. Currently available data are consistent with the notion that Se(0)-protein conjugates act as air oxidation catalysts that cause a rapid depletion of intracellular glutathione and induce apoptosis. Drugs modeled after our Se(0)-protein conjugates may prove useful for the local and/or systemic therapy of cancer.  相似文献

Deprotonation of the tridentate isoindoline ligand 1,3-bis[2-(4-methylpyridyl)imino]-isoindoline, 4'-MeLH, and reaction with hydrated zinc(II) perchlorate produces an unexpected trinuclear Zn(II) complex, [Zn(3)(4'-MeL)(4)](ClO(4))(2).5H(2)O (1), whereas reaction with hydrated copper(II) perchlorate in methanol produces the expected mononuclear product, [Cu(4'-MeL)(H(2)O)(2)]ClO(4) (2). X-ray diffraction shows that the trinuclear Zn(II) complex (1) contains a linear zinc backbone, and the arrangement of ligands about the outer chiral zinc(II) atoms is helical. The two terminal zinc ions exhibit approximate C(2) site symmetry, with tetrahedral coordination by two pyrrole and two pyridyl nitrogen atoms of the potentially tridentate isoindoline ligands. The central zinc ion exhibits approximate tetrahedral symmetry, with coordination by four pyridyl nitrogen atoms of four different isoindoline ligands. Pyridyl-pyrrole intramolecular pi-stacking interactions contribute to the stability of the trinuclear cation. The structure of the mononuclear copper(II) complex cation in 2 is best described as a distorted trigonal bipyramid. The isoindoline anion binds Cu(II) in both axial positions and one of the equatorial positions; water molecules occupy the other two equatorial positions.  相似文献

The conditions and premises of the kinetic evaluation of thermoanalytical curves on the basis of chemical kinetics are demonstrated, and the limits of their availability are pointed out. For reactions with an unclear structural and chemical course, some appropriate definitions of terms are proposed.The possibilities of the use of the inflection points of the reaction rate versus time curve are presented. New methods of determination of kinetic parameters are described, including the ratio of the degrees of reactionx, the ratio of the reaction ratesx and the ratio of the tangent slopesx at these points.
Zusammenfassung Bedingungen und Voraussetzungen der kinetischen Auswertung thermoanalytischer Meßkurven auf der Basis der chemischen Kinetik werden dargestellt, gleichzeitig wird auf die Grenzen ihrer Gültigkeit hingewiesen. Für Reaktionen mit unklarem chemischem oder strukturellem Ablauf werden geeignete neue Bezeichnungen definiert.Die Möglichkeit zur Verwendung der Wendepunkte der Kurve Reaktionsgeschwindigkeit gegen Zeit vorgestellt. Neue Methoden werden beschrieben, die zur Bestimmung kinetischer Parameter das Verhältnis der Umsätzex 1 x 2, das Verhältnis der Reaktionsgeschwindigkeitenx 1/x 2, und das Verhältnis der Tangentensteigungenx 1/x 2 an den Wendepunkten verwenden.

Theory suggests that thermocapillary flow about neighboring bubbles in liquids on hot walls pulls the bubbles together. A temperature gradient perpendicular to the wall establishes a surface tension gradient at the bubble-liquid interface, which in turn sustains a shear stress gradient that pumps adjacent fluid away from the wall. Neighboring bubbles are mutually entrained in this flow and also respond thermophoretically to lateral temperature gradients in the temperature near field. The theory predicts that the aggregation velocity scales with the temperature gradient, the radius of the bubbles, the derivative of the surface tension with respect to temperature, and the reciprocal of the liquid's viscosity. Bubble aggregation experiments under controlled conditions were performed to test the theory. Scaling the experimental bubble trajectories according to the theory substantially collapses all of the data onto a master curve when the interbubble separation is greater than 3 radii, which suggests that the theory is correct. Calculated velocities agree with the experimental results when hindrance of bubble motion due to the wall is included. Values for the parameter that describes the hindrance effect are obtained from fitting the data to the theory, from independent measurements, and from direct hydrodynamic calculation. A nonaqueous capillary electrophoresis/electrospray mass spectrometry method for the separation of lidocaine (LID) and two of its metabolites, monoethylglycinexylidide (MEGX) and glycinexylidide (GX), has been developed. The separation medium was: 70 mM ammonium formate and 2.0 M formic acid in acetonitrile/methanol (60:40 v/v). With a sheath liquid of methanol/water (80:20 v/v) containing 2% formic acid and positive ion detection, reproducible determinations (8-11% relative standard deviation (RSD)) of lidocaine and its metabolites were performed in spiked human plasma. The limits of detection (LODs) were between 69.1 and 337 nM. The influences of sheath liquid composition, nebulizing gas pressure and drying gas temperature on the separation were examined.  相似文献

The mechanism by which a benzotriazine 1,4-dioxide class of anticancer drugs produce oxidizing radicals following their one-electron reduction has been investigated using tirapazamine (3-amino-1,2,4-benzotriazine 1,4-dioxide, 1) and its 6-methoxy (6), 7-dimethylamino (7), and 8-methyl (8) analogues. By measuring the changes in absorption with pH, we found that the radical anions undergo protonation with radical pK(r) values of 6.19 +/- 0.05, 6.10 +/- 0.03, 6.45 +/- 0.04, and 6.60 +/- 0.04, respectively. The one-electron reduced species underwent a first-order reaction, with increased rate constants from 112 +/- 23 s(-)(1) for 1 to 777 +/- 12 s(-)(1)(6), 1120 +/- 29 s(-)(1) (7), and 825 +/- 89 s(-)(1) (8) at pH 7. No overall change in conductance was observed following the one-electron reduction of 6, and 8 at pH 4.5, consistent with the protonation of the radical anions, but a loss in conductance was seen for one-electron reduced 7 because of further protonation of the initially formed radical. This is assigned to the protonation of the dimethylamino group of the radical species, which has a pK(a) of 8.8 +/- 0.3. All conductance changes take place on a time-scale shorter than those of the above first-order reactions, which are not associated with the formation or loss of charged species. The absorption spectra present at the end of the unimolecular reactions were found to be similar to those formed immediately upon the one-electron oxidation of the respective substituted 3-amino-1,2,4-benzotriazine 1-oxides, and it is suggested that common benzotriazinyl radicals are formed by both routes. All these intermediate radicals underwent dismutation to produce final spectra matched by equal contributions of the parent compound and their respective substituted 3-amino-1,2,4-benzotriazine 1-oxides. By establishing redox equilibria between the intermediate radicals formed on the one-electron oxidation of the respective 3-amino-1,2,4-benzotriazine 1-oxides of the compounds and reference compounds, we found the one-electron reduction potential of the oxidizing radicals to range from 0.94 to 1.31 V. The benzotriazinyl radical of tirapazamine was found to oxidize dGMP and 2-deoxyribose with rate constants of (1.4 +/- 0.2) x 10(8) M(-)(1) s(-)(1) and (3.7 +/- 0.5) x 10(6) M(-)(1) s(-)(1), respectively.  相似文献

Among the numerous well-characterized families of glycosidases, family 4 appears to be the anomaly, requiring both catalytic NAD+ and a divalent metal for activity. The unusual cofactor requirement prompted the proposal of a mechanism involving key NAD+-mediated redox steps as well as elimination of the glycosidic oxygen. Primary kinetic isotope effects for the 2- and 3-deutero substrate analogues, isotopic exchange with solvent, and structural analysis of a 6-phospho-beta-glucosidase, BglT (E.C. 3.2.1.6), provided evidence in support of the proposed mechanism, which has striking resemblances to that of the sugar dehydratases. Furthermore, analysis of the stereochemical outcome indicated that family 4 enzymes are retaining glycosidases.  相似文献

Extracts from the seeds of Annona muricata yielded three new Annonaceous acetogenins: muricatetrocin A (= (5S)-3-{(2R)-2-hydroxy-9-{(2R,5S)-tetrahydro-5-[(1S,4S,5S)-1,4,5-trihydroxyheptadecyl]furan-2-yl}nonyl}-5-methylfuran-2(5H)-one; 1 ), muricatetrocin B (= (5S)-{(2R)-2-hydroxy-9-{(2S,5S)-tetrahydro-5-[(1S,4S,5S)-1,4,5-trihydroxyheptadecyl]furan-2-yl}nonyl}-5-methylfuran-2(5H)-one; 2 ), and gigantetrocin B (= (5S)-3-{(2R)-2-hydroxy-7-{(2S,5S)-tetrahydro-5-[(1S,4R,5R)-1,4,5-trihydroxynonadecyl]furan-2-yl}heptyl}-5-methyl-furan-2(5H)-one; 3 ). Their C-skeletons were deduced by mass spectrometry. Configurations were determined by 1H-NMR of ketal derivatives and 2D-NMR experiments utilizing Mosher esters. A previously described compound, gigantetrocin A (= (5S)-3-{(2R)-2-hydroxy-7-{(2S,5S)-tetrahydro-5-[(1S,4S,5S)-1,4,5-trihydroxynonadecyl]furan-2-yl}heptyl}-5-methylfuran-2-(5H)one; 4 ), was also isolated and is new to this species. Compounds 1–4 were all selectively cytotoxic for the HT-29 human colon-tumor cell line with potencies at least 10 times that of adriamycin.  相似文献

We describe a fully automated high performance liquid chromatography 9.4 tesla Fourier transform ion resonance cyclotron (FTICR) mass spectrometer system designed for proteomics research. A synergistic suite of ion introduction and manipulation technologies were developed and integrated as a high-performance front-end to a commercial Bruker Daltonics FTICR instrument. The developments incorporated included a dual-ESI-emitter ion source; a dual-channel electrodynamic ion funnel; tandem quadrupoles for collisional cooling and focusing, ion selection, and ion accumulation, and served to significantly improve the sensitivity, dynamic range, and mass measurement accuracy of the mass spectrometer. In addition, a novel technique for accumulating ions in the ICR cell was developed that improved both resolution and mass measurement accuracy. A new calibration methodology is also described where calibrant ions are introduced and controlled via a separate channel of the dual-channel ion funnel, allowing calibrant species to be introduced to sample spectra on a real-time basis, if needed. We also report on overall instrument automation developments that facilitate high-throughput and unattended operation. These included an automated version of the previously reported very high resolution, high pressure reversed phase gradient capillary liquid chromatography (LC) system as the separations component. A commercial autosampler was integrated to facilitate 24 h/day operation. Unattended operation of the instrument revealed exceptional overall performance: Reproducibility (1-5% deviation in uncorrected elution times), repeatability (<20% deviation in detected abundances for more abundant peptides from the same aliquot analyzed a few weeks apart), and robustness (high-throughput operation for 5 months without significant downtime). When combined with modulated-ion-energy gated trapping, the dynamic calibration of FTICR mass spectra provided decreased mass measurement errors for peptide identifications in conjunction with high resolution capillary LC separations over a dynamic range of peptide peak intensities for each spectrum of 10(3), and >10(5) for peptide abundances in the overall separation.  相似文献
