NanoFAST: structure-based design of a small fluorogen-activating protein with only 98 amino acids

One of the essential characteristics of any tag used in bioscience and medical applications is its size. The larger the label, the more it may affect the studied object, and the more it may distort its behavior. In this paper, using NMR spectroscopy and X-ray crystallography, we have studied the structure of fluorogen-activating protein FAST both in the apo form and in complex with the fluorogen. We showed that significant change in the protein occurs upon interaction with the ligand. While the protein is completely ordered in the complex, its apo form is characterized by higher mobility and disordering of its N-terminus. We used structural information to design the shortened FAST (which we named nanoFAST) by truncating 26 N-terminal residues. Thus, we created the shortest genetically encoded tag among all known fluorescent and fluorogen-activating proteins, which is composed of only 98 amino acids.

We solved the structure of a fluorogen-activating protein FAST and synthesized the library of potential fluorogens. Using these data, we designed the shortest genetically encoded fluorescent tag among all known.     

Higher order bose-einstein correlations in π+p and K+p collisions at 250 GeV/c

Bose-Einstein correlations up to fourth order are presented at $\sqrt s = 22GeV$ . Genuine third-order correlations are observed. The experimental data are compared with predictions from a quantum statistical approach of radiation from a partially coherent source and with the FRITIOF model.     

Unexpected stereoselective sodium acetate catalyzed multicomponent cyclization of aryl aldehydes, malononitrile and acetone into cis-4-dicyanomethylene-2,6-diarylcyclohexane-1,1-dicarbonitriles

A new type of multicomponent reaction is described in which five organic molecules form a cyclohexane ring. Aryl aldehydes, malononitrile and acetone in the presence of a catalytic amount of sodium acetate are stereoselectively cyclized into cis-4-dicyanomethylene-2,6-diarylcyclohexane-1,1-dicarbonitriles in 30-60% yields.     

Classes of tuples of commuting contractions satisfying the multivariable von Neumann inequality

We obtain a decomposition for multivariable Schur-class functions on the unit polydisk which, to a certain extent, is analogous to Agler's decomposition for functions from the Schur-Agler class. As a consequence, we show that d-tuples of commuting strict contractions obeying an additional positivity constraint satisfy the d-variable von Neumann inequality for an arbitrary operator-valued bounded analytic function on the polydisk. Also, this decomposition yields a necessary condition for solvability of the finite data Nevanlinna-Pick interpolation problem in the Schur class on the unit polydisk.     

Moderate deviations for queues in critical loading

We establish logarithmic asymptotics of moderate deviations for queue-length and waiting-time processes in single server queues and open queueing networks in critical loading. One-Circuit Stabilization of Millimeter-Wave Solid-State Oscillator by Quasi-Optical Resonator

The construction and operating characteristics of the highly stabilized millimeter wave IMPATT oscillator are described. The frequency stability is 8.1 × 10^-7 at 115 GHz. The output power is no less than 15 mW. The sphere-corner-echelette open resonator have been used for oscillator stabilization. This approach is also adaptable to the Gunn oscillator.     

Radiographic analysis of the structures of films obtained by vacuum sputtering of some copper-based low alloys

Diffractometric phase analysis of the structures of alloys on copper base, containing small concentrations of Mn, Pd, Al, Ti, Zr, Ce, and Ni, and of films on their base, was carried out. Thegeometry of particles in the external field yielding a givenminimum energy configuration is studied. As an application,the wandering vectors of the shift operator in the Dirichletspaces associated with finitely atomic measures are also studied.In particular, the zero locus of a wandering vector is discussed.
