13-Hydroxyacenaphtho[1,2-b]quinolizinium bromide (13-HQBr)was selected as a fluorescence indicator to determine basic compounds in non-aqueous media. This compound possesses an acidic phenolic hydroxyl group. It presents varying absorption (ROH, 408, 430 nm; RO(-) 456, 478 nm) and excitation spectra (ROH, 425 nm; RO, 471 nm) depending on the pH of the media, but the same emission fluorescence spectrum (ROH = RO(-), 526 nm) at different pH in buffered aqueous solutions. However, in acidic non-aqueous media (acetic, formic and trifluoroacetic acids), it can be observed that the fluorescence emission spectra differ for the ionized (lambda(em) = 530 nm) and non-ionized (lambda(em) = 440, 470 nm) forms. The fluorescence intensity at the characteristic peaks depends on the acid-base equilibria in the ground and excited states. Therefore, this property could be used to evaluate the concentration of basic compounds, showing a good linearity range between fluorescence intensity and basic sample concentration.  相似文献

Conducting electroactive polymers (CPs) are materials discovered just over 20 years ago which have aroused considerable interest on account of their electronic conducting properties and unique chemical and biochemical properties. Consequently, they have numerous (bio)analytical and technological applications. CPs are easily synthesized and deposited onto the conductive surface of a given substrate from monomer solutions by electrochemical polymerization with precise electrochemical control of their formation rate and thickness. Coating electrodes with CPs under mild conditions opens up enormous possibilities for the immobilization of biomolecules and bioaffinity or biorecognizing reagents, the improvement of their electrocatalytic properties, rapid electron transfer and direct communication to produce a range of analytical signals and new analytical applications. Co-immobilization of other molecules (enzymatic co-factors or charge-transfer mediators) by entrapment within electropolymerized films or by covalent binding on these films permits straightforward fabrication of reagentless biosensors. The characteristics of CPs and their uses, mainly in amperometric biosensors, are reviewed. The most recent applications and lines of research related to CP films are summarized in the different sections of the paper, and probable future trends are discussed.  相似文献

A new reagentless system for sulphite (or sulphur dioxide) determination is reported based on the use of an organic conducting polymer, polyaniline, and its absorbance variation at 550 nm, depending on the sulphite concentration. After chemical polymerisation of aniline a very thin film of polyaniline is obtained. Although the response is not fully reversible, each film can be used for at least 10 measurements for low analyte concentrations (up to 0.5 mg l−1) and five measurements for higher sulphite concentrations. Moreover, the reproducibility, ease of preparation and low cost of the films, permit the use of a new disposable system for each measurement. When the change in absorbance at 550 nm was measured for 210 s (stabilisation time), the system showed a linear response, which ranged from 0.025 to 1.50 mg l−1 of sulphite. A theory with regard to the reaction mechanism between the polyaniline films and sulphite is also proposed. The system was applied to sulphite determination in wine samples and the results were in agreement with those obtained by the Official Method of Analysis (iodometric titration).  相似文献

Reaction of azaenolates of 2-alkyl-oxa(thia)zolines 6 with imidoyl chlorides 7 as electrophiles to furnish masked N-substituted beta-enamino acid derivatives 1-2 in 70-90% yield is described. Alternative routes are discussed. Compounds 1-2 generally appear in one tautomeric form, imino or enamino, depending on the nature of the imidoyl chloride. The configuration of the enamino moiety (Z) and the conformation (s-cis) of compounds 1-2 obtained were established by an NMR study and unequivocally set by nuclear Overhauser effect difference experiments. An X-ray structure of compound 1e is also reported, showing a strong intramolecular NH.N hydrogen bond. Ab initio calculations (HF/3-21G and HF/3-21+G) have been carried out on several representative examples (1e, 1p, and 1l) in an attempt to support and provide the correct geometry of these derivatives. Structural considerations among the possible isomers of compounds 1 are discussed. From these studies it was concluded that the theoretical calculations agree with the experimental results. In addition, a very simple one-pot procedure for the preparation of masked N-substituted alpha-alkylated beta-enamino acid derivatives 2 from 6, 7, and different alkyl halides (R(3)Y) is described.  相似文献

A new reactor and a novel in-situ sampling technique were developed for the study of the synthesis of CeO2 powders produced from dissolved cerium nitrate salts. The conical reactor minimized particle recirculation and provided a highly symmetrical and undisturbed plasma flow suitable for the analysis of the phenomena affecting the formation of CeO2 powders. Both a calorimetric study of the reactor and a thermodynamic analysis of CeO2 formation were conducted. The sampling probe is described and near-isokinetic sampling was achieved. The sampled particles were collected using a miniature wet collection system, i.e. a mist atomizer and a custom-made spray chamber. A numerical simulation of the velocity and temperature fields of the plasma gas in the reactor was done using Fluent. A comprehensive droplet-to-particle formation mechanism presented elsewhere is revisited and expanded based on calorimetry, thermodynamics of CeO2 formation, numerical simulations and collected particles. No traces of other oxidation states other than CeO2 were found.  相似文献

An FI-ICP-AES method for the determination of trace levels of mercury in biological samples has been described, which is based on the extraction of the mercury complex with 1,5-bis (di-2-pyridyl)methylene thiocarbonohydrazide (DPTH) on-line into isobuthyl-methyl ketone (IBMK). The organic phase (containing the complex) has been mixed on-line with SnCl2 in N,N-dimethylformamide. Thus, mercury vapour can be generated directly from the organic phase and separated in a gas-liquid separation device. The detection limit for mercury is 4 ng/ml and the calibration curve is linear at least from 10 to 2500 ng/ml. The relative standard deviation for 10 replicate measurements is ±1% for 100 ng/ml of Hg(II). Results from the analysis of some certified biological reference materials are given.  相似文献

We report a common HPLC method for the single or simultaneous determination of four calcium channel blockers (CCB), namely diltiazem (DTZ), verapamil (VER), nifedipine (NIF) and nitrendipine (NIT) and their active metabolites demetildiltiazem and deacetildiltiazem (MA and M1), norverapamil (NOR), and dehydronifedipine (DHN). DHN was first synthesised in our laboratory and different pH values of the mobil phase were subsequently prepared and tested for chromatographic separation. The detection system and the environmental light conditions were optimised. The best separations of all analytes were obtained using a C18 column and a mobile phase of methanol, 0.04 M ammonium acetate, acetonitrile and triethylamine (2:2:1:0.04 v/v). Quantitation was performed using imipramine (IMI) as the internal standard. For DTZ and its metabolites (M1 and MA), the wavelength chosen was 237 nm; for VER and its metabolite NOR, it was 210 nm; and, finally for NIF and its metabolite DHN and NIT it was 216 nm. When a simultaneous analysis was carried out the wavelength was of 230 nm. The optimum pH were 7.90 and 7.10 when the separation of NIT and DTZ or VER and NIF were carried out, respectively, and 7.90 when a simultaneous separation was carried out. The detection limit of the assay was less than 8 ng ml−1 for all compounds, with coefficients of variation less than 7% (for inter- and intra-day) over the concentration range of 1–1000 ng ml−1. The retention times were less than 11 min. When NIF or NIT were studied, it was necessary to use a sodium vapour lamp in order to avoid the photodegradation which takes place under daylight conditions.  相似文献

The present work examines the relationship between the antimicrobial activity of novel arginine-based cationic surfactants and the physicochemical process involved in the perturbation of the cell membrane. To this end, the interaction of these surfactants with two biomembrane models, namely, 1,2-dipalmitoyl-sn-glycero-3-phosphocholine (DPPC) multilamellar lipid vesicles (MLVs) and monolayers of DPPC, 1,2-dipalmitoyl-sn-glycero-3-[phospho-rac-(1-glycerol)] sodium salt (DPPG), and Escherichia coli total lipid extract, was investigated. For the sake of comparison, this study included two commercial antimicrobial agents, hexadecyltrimethylammonium bromide and chlorhexidine dihydrochloride. Changes in the thermotropic phase transition parameters of DPPC MLVs in the presence of the compounds were studied by differential scanning calorimetry analysis. The results show that variations in both the transition temperature (Tm) and the transition width at half-height of the heat absorption peak (deltaT1/2) were consistent with the antimicrobial activity of the compounds. Penetration kinetics and compression isotherm studies performed with DPPC, DPPG, and E. coli total lipid extract monolayers indicated that both steric hindrance effects and electrostatic forces explained the antimicrobial agent-lipid interaction. Overall, in DPPC monolayers single-chain surfactants had the highest penetration capacity, whereas gemini surfactants were the most active in DPPG systems. The compression isotherms showed an expansion of the monolayers compared with that of pure lipids, indicating an insertion of the compounds into the lipid molecules. Owing to their cationic character, they are incorporated better into the negatively charged DPPG than into zwitterionic DPPC lipid monolayers.  相似文献
