Atomically precise gold nanoclusters are a fascinating class of nanomaterials that exhibit molecule-like properties and have outstanding photoluminescence (PL). Their ultrasmall size, molecular chemistry, and biocompatibility make them extremely appealing for selective biomolecule labeling in investigations of biological mechanisms at the cellular and anatomical levels. In this work, we report a simple route to incorporate a preformed Au25 nanocluster into a model bovine serum albumin (BSA) protein. A new approach combining small-angle X-ray scattering and molecular modeling provides a clear localization of a single Au25 within the protein to a cysteine residue on the gold nanocluster surface. Attaching Au25 to BSA strikingly modifies the PL properties with enhancement and a redshift in the second near-infrared (NIR-II) window. This study paves the way to conrol the design of selective sensitive probes in biomolecules through a ligand-based strategy to enable the optical detection of biomolecules in a cellular environment by live imaging. 相似文献

Transfer Factors (Fv) of 238U, 226Ra, 234Th, 210Po and 210Pb from five different agricultural soils in semi-arid region (Syria) to four different barley genotypes were studied in an agricultural potted experiment. The geometric mean of the Fv values were (0.08) for 210Pb, and (0.02) for 210Po, while it ranged from 0.18 to 0.42 ,from 0.08 to 0.15 and from 0.22 to 0.4 for 238U, 234Th and 226Ra, respectevily. The Fv values of 238U and 226Ra were within the recommended global medians, while the Fv values of 234Th, 210Pb and 210Po were higher. There is no clear relationship between the soil properties and Fv of all studied radionuclides to barley genotypes. Moreover, the expression of glutathione (GSH) gene, which is belived to be involoved in heavy metal removal was generally low in all studied varieties grown in all soil types.
BackgroundAcinetobacter baumannii is a highly antimicrobial resistant nosocomial pathogen. Resistance to currently used antibiotics has limited effective drugs against this bacterium. This study aimed to propose a rational inhibitor design against the LpxA protein of A. baumannii using a virtual screening method based on a similar structure of ligands.MethodsIn this study, we targeted LpxA protein, which is involved in the early stage of LPS biosynthesis. In the next step, we used Peptide920 and 1,2- Ethanediol as templates to find similar compounds using Drugbank and Zinc15 webservers, respectively. Subsequently, molecular dynamics (MD) simulations were carried out for LpxA protein and two complexes of ZINC895081 and Macrolactam-1 which represented the highest binding affinity and best conformation. Finally, ADMET properties, water solubility and drug-likeness of the desired compounds were evaluated using SwissADME and DruLiTo softwares.ResultsAccording to considered criteria, Drugbank suggested 5 compunds including Ilomastat, Macrolactam-1, Macrolactam-2, Macimorelin, and Oglufanide. On the other hand, Zinc15 webserver suggested 4 compunds including ZINC895048, ZINC895081, ZINC901061 and ZINC1531008. The result of the HDOCK server and Molegro virtual docker (MVD) showed that Macrolactam-1 and ZINC895081 (Citrate) had the highest docking score. In addition, MD simulations showed that ZINC895081 and Macrolactam-1 ligands have the stable binding to the LpxA protein. According to Lipinski's rule, these two compounds are non-carcinogenic, non-toxic and promising inhibitors against LpxA of A. baumannii.ConclusionIt seems that Macrolactam-1 and ZINC895081 (Citrate) are two valuable promising inhibitors against the LpxA protein of A. baumannii. Further in vitro and in vivo experiments are needed to confirm the capabilities of these proposed compounds against A. baumannii.相似文献
We consider the reconstruction of a heat equation modeling the temperature distribution of a rod from readings at one end only. We assume that the rod, whose length is unknown, is totally buried except for a neighborhood of one of its ends, which is accessible for temperature measurements. This work is the first ever attempt to measure the interfacial tension (IFT) in a gas–gas system, the interfacial tension of supercritical carbon dioxide (SCO2–methane) was measured at temperatures of 368.15 and 433.15 K and pressures between 6.9 and 41.37 MPa, using a modified pendant drop method. A highly sensitive, selective and rapid method for the determination μg L(-1) level of Au(III) based on the rapid reaction of Au(III) with 2,3-dichloro-6-(3-carboxy-2-hydroxy-1-naphthylazo)quinoxaline (DCHNAQ) and the solid phase extraction of the colored complex with a reversed phase polymer-based C18 cartridge have been developed. The DCHNAQ reacted with Au(III) to form a violet complex of a molar ratio 3:1 [DCHNAQ to Au(III)] in the presence of 5.0 M of phosphoric acid solution and Triton X-100 medium. This complex was enriched by the solid phase extraction with a polymer-based C18 cartridge. The enrichment factor of 100 was achieved. The molar absorptivity of the complex is 2.73×10(5) l mol(-1) cm(-1) at 633 nm in the measured solution. The system obeys Beer's law in the range of 0.02-1.30 μg ml(-1), whereas the optimum concentration ranges obtained from Ringbom plot was 0.08-1.24 μg ml(-1). The relative standard deviation for ten replicates sample of 0.6 μg ml(-1) level is 1.28%. The detection and quantification limits, are 6.1 and 19.5 ng ml(-1) in the original sample. This method was applied to the determination of gold in water, jewel and ore samples with good results comparing to the GFAAS method. 相似文献
