Higher animal's lipases are well characterized; however, much less is known about lipases from mollusks. A lipolytic activity was located in the land snail (Eobania vermiculata) digestive glands (hepatopancreas), from which a snail digestive lipase (SnDL) was purified. Pure SnDL has a molecular mass of 60 kDa; it does not present the interfacial activation phenomenon. It was found to be more active on short-chain triacylglycerols than on long-chain triacylglycerols. The NH2-terminal sequence of the SnDL shows 66% of identity with the 17 NH2-terminal amino acids of a putative lipase from sea urchin (Strongylocentrotus purpuratus). No sequence identity was found with known lipases. Interestingly, neither colipase nor bile salts were detected in the snail hepatopancreas. This suggests that colipase evolved in vertebrates simultaneously with the appearance of an exocrine pancreas and a true liver which produces bile salts. Altogether, these results suggest that SnDL is a member of a new group of digestive lipases belonging to invertebrates.  相似文献

A cellulose triacetate (CTA) membrane containing trioctylphosphine oxyde (TOPO) as carrier and 2-nitrophenyloctyl ether (NPOE) as a plasticizer was prepared. The membrane CTA + NPOE + TOPO was characterised using chemical techniques as well as Fourier Transform InfraRed (FTIR) spectroscopy, X-ray diffraction and Scanning Electron Microscopy (SEM). The CTA membrane is characterised by well-defined pores; these pores are completely filled with the NPOE and carrier. Surfaces of membranes with TOPO are smooth. The systems constituted by the mixture of CTA + NPOE, CTA + NPOE + TOPO do not give any diffraction. This can be due to the absence of crystallization within the membrane. On the other hand, this result should be attributable to the amorphous state of the structure, which permits us to eliminate the mechanism of transfer of the ions by electron jump. A comparative study of transport across a polymer inclusion membrane (PIM) and a supported liquid membrane (SLM) containing the same carrier in chloroform has shown that uranium or molybdenum transport efficiency was increased using PIM instead of SLM. PIM showed higher stability than SLM, the flux of transport remain constant in the former case after 2 weeks.  相似文献

Natural herbal medicines are an important source of enzyme inhibitors for the discovery of new drugs. A number of natural extracts such as green tea have been used in prevention and treatment of diseases due to their low‐cost, low toxicity and good performance. The present study reports an online assay of the activity and inhibition of the green tea extract of the Glucose 6‐phosphate dehydrogenase (G6PDH) enzyme using multilayer capillary electrophoresis based immobilized enzyme microreactors (CE‐IMERs). The multilayer CE‐IMERs were produced with layer‐by‐layer electrostatic assembly, which can easily enhance the enzyme loading capacity of the microreactor. The activity of the G6PDH enzyme was determined and the enzyme inhibition by the inhibitors from green tea extract was investigated using online assay of the multilayer CE‐IMERs. The Michaelis constant (Km) of the enzyme, the IC50 and Ki values of the inhibitors were achieved and found to agree with those obtained using offline assays. The results show a competitive inhibition of green tea extract on the G6PDH enzyme. The present study provides an efficient and easy‐to‐operate approach for determining G6PDH enzyme reaction and the inhibition of green tea extract, which may be beneficial in research and the development of natural herbal medicines. The electrochemical behaviors of native and thermally denatured fish DNA was investigated using boron-doped diamond (BDD) film electrode by cyclic voltammetry. The BDD electrode afforded us to measure weak current less than muA for the DNA solution in 100 microl. The mixture of acetic acid and sodium acetate solution (0.2 M) was used as a supporting electrolyte. Two oxidation peaks were observed at about +1.1 V and +1.3 V at pH 4.6 for thermally denatured fish DNA. This is due to the oxidation of guanine and adenine in the denatured fish DNA, respectively. In contrast, the native fish DNA showed ill-defined peaks at +1.1 V. Furthermore, the electrochemical behaviors of thermally denatured fish DNA were studied in the presence of cytosine, cytidine, cytidine-5-monophosphate, tetrakis(1-methypyridinium-4-yl)porphyrin (H(2)(TMPyP)(4+)) and Ru(II)(TMPyP)(4+). The oxidation peak intensity at +1.1 V gradually decreased with the increase of the concentrations of the above compounds. Based on the above studies, electrochemical behaviors of the thermally denatured fish DNA at BDD electrode is discussed.  相似文献

We propose a three-field formulation for efficiently solving a two-dimensional Stokes problem in the case of nonstandard boundary conditions. More specifically, we consider the case where the pressure and either normal or tangential components of the velocity are prescribed at some given parts of the boundary. The proposed computational methodology consists in reformulating the considered boundary value problem via a mixed-type formulation where the pressure and the vorticity are the principal unknowns while the velocity is the Lagrange multiplier. The obtained formulation is then discretized and a convergence analysis is performed. A priori error estimates are established, and some numerical results are presented to highlight the perfomance of the proposed computational methodology.

A commercial cation exchange resin Amberlite 200 has been modified after immersion in solutions of polyethyleneimine (PEI). The kinetic of fixation of the metallic ions have been determined. The modification of the surface of the resin deals with a change in the order of the affinities of the resins towards cations. The retention is the function of the formation and the stability of the complex. The conditions of modification (pH, PEI concentration and time of immersion) have been examined and the modification was confirmed by the determination of the exchange capacities, the distribution coefficient (P) and the selectivity factors (S). The obtained results revealed the effect of PEI on the exchange properties of the resin. The pH range selected (6-8) permitted a good adherence of PEI onto the resin surface. The quantity of the adsorbed PEI was increased by raising the initial concentration and the immersion period. The exchange capacity for copper ion passed from 2.6 mmol g−1, in the case of unmodified resin, to 3.9 mmol g−1 for the modified one.  相似文献

The crystal structure of tripotassium pentairon hexaphosphate has been determined by single‐crystal X‐ray diffraction. The structure contains one Fe atom on a center of symmetry, one K, two Fe and two P atoms on twofold axes, and one Fe, two P and one K atom in general positions. The K3Fe5(PO4)6 structure consists of a complex three‐dimensional framework of corner‐sharing between iron polyhedra, and corner‐ and edge‐sharing between PO4 tetrahedra and iron polyhedra (FeO5 and FeO6). This linkage between iron and phosphorus forms intersecting channels containing the K atoms.  相似文献

A novel, highly selective and sensitive paper-based colorimetric sensor for trace determination of copper (Cu2+) ions was developed. The measurement is based on the catalytic etching of silver nanoplates (AgNPls) by thiosulfate (S2O32−). Upon the addition of Cu2+ to the ammonium buffer at pH 11, the absorption peak intensity of AuNPls/S2O32− at 522 nm decreased and the pinkish violet AuNPls became clear in color as visible to the naked eye. This assay provides highly sensitive and selective detection of Cu2+ over other metal ions (K+, Cr3+, Cd2+, Zn2+, As3+, Mn2+, Co2+, Pb2+, Al3+, Ni2+, Fe3+, Mg2+, Hg2+ and Bi3+). A paper-based colorimetric sensor was then developed for the simple and rapid determination of Cu2+ using the catalytic etching of AgNPls. Under optimized conditions, the modified AgNPls coated at the test zone of the devices immediately changes in color in the presence of Cu2+. The limit of detection (LOD) was found to be 1.0 ng mL−1 by visual detection. For semi-quantitative measurement with image processing, the method detected Cu2+ in the range of 0.5–200 ng mL−1(R2 = 0.9974) with an LOD of 0.3 ng mL−1. The proposed method was successfully applied to detect Cu2+ in the wide range of real samples including water, food, and blood. The results were in good agreement according to a paired t-test with results from inductively coupled plasma-optical emission spectrometry (ICP-OES).  相似文献
