Total synthesis of (+)-anamarine

An enantiospecific total synthesis of polyhydroxy δ-pyrone natural product (+)-anamarine is accomplished. The main features of the synthesis include the stereoselective reduction of the ketone obtained by the desymmetrization of the bis-dimethyl amide of tartaric acid and further elaboration involving asymmetric Brown's allylation and ring closing metathesis.     

Regioselective synthesis of thiophene fused sultam derivatives via iodocyclization approach and their application towards triazole linker

An efficient regioselective synthesis of 4-iodo-2,3-disubstituted-2H-thieno[3,2-e][1,2]thiazine-1,1-dioxide derivatives via iodocyclization approach using iodine under mild reaction condition described herein. This coupling–iodocyclization strategy tolerated a variety of functional groups such as alkyl, cycloalkyl, phenyl producing the six-membered heterocyclic ring selectively. The resulting 4-iodo-2,3-disubstituted-2H-thieno[3,2-e][1,2]thiazine-1,1-dioxide was coupled with a variety of boronic acids (Suzuki coupling) and activated alkenes (Heck coupling). The iodo group was utilized for Sonogashira coupling followed by efficient transformation to azido precursor, which was used for the synthesis of various thieno-sultam linked with triazole.     

An efficient one pot method for synthesis of carboxylic acids from nitriles using recyclable ionic liquid [bmim]HSO4

Environmentally benign ionic liquid [bmim]HSO₄ was found suitable for conversion of nitriles into carboxylic acids under mild conditions with excellent purity.     

Vitamin B6 cofactor derived chemosensor for the selective colorimetric detection of acetate anions

A novel vitamin B₆ cofactor derived anion sensor L for the selective colorimetric detection of acetate has been developed by the condensation of pyridoxal and 2-aminothiophenol. The sensor L showed a noteworthy change in the visible region of the spectrum and was detected by the ‘naked-eye’ for both acetate and fluoride anions in DMSO but selectively for acetate in DMSO/H₂O (88:12, v/v). The anion recognition ability of L was investigated by spectroscopic (UV–vis and ¹H NMR) and DFT methods.     

Zinc triflate catalyzed acylation of alcohols,phenols, and thiophenols

An expedient procedure for the acylation of alcohols, phenols, and thiophenols using catalytic amount of Zn(OTf)₂ is described. This procedure is highly suitable for industrial application due to use of less toxic metal as a part of catalyst, short reaction time at ambient temperature, without any racemization of chiral alcohols.     

Cu-promoted single-pot intramolecular esterification of C-3 functionalized azetidin-2-one: an efficient diastereoselective access to azido-/amino-aza-lactones

A facile, single pot diastereoselective access to seven and eight membered aza-heterocycles was developed by using β-lactam-synthon approach. The developed protocol does not involve the typical intricacies viz. the use of expensive transition metal catalysts and high boiling solvents, associated with the convenient protocols.     

Novel domino reactions for the efficient synthesis of 5,6-dihydro-1,4,2-oxathiazines

A facile efficient synthesis of novel 3-aryl-5,6-dihydro-1,4,2-oxathiazin-6-ols from the reaction of (E)-N-hydroxyarylimidoyl chlorides and 1,4-dithiane-2,5-diol in the presence of triethylamine is described. This transformation presumably proceeds via in situ generation of 2-mercaptoacetaldehyde and nitrile oxide and their concomitant [3+3] annulation.     

Base mediated synthesis of α-aminated aroyl/acetylnaphthalenes through [4+2] annulations

We have developed a base promoted simple, efficient and alternative approach for the synthesis of 4-amino-3-aroyl//heteroaroyl/acetyl-2-methylsulfanyl-naphthalene-1-carbonitriles by reaction of easily accessible 3,3-bis(methylthio)-1-aryl/heteroaryl/acetylprop-2-en-1-one and 2-cyanomethyl-benzonitrile. Reaction of 1-(2-halo/methoxy-phenyl)-3,3-bis(methylthio)prop-2-en-1-one and 2-cyanomethyl-benzonitrile under basic conditions also afforded 6-(methylthio)-7-oxo-7,12-dihydrobenzo[c]acridine-5-carbonitrile along with usual product. Structure of the synthesized product has been confirmed by single X-ray crystallography.     

Cu(OTf)2-promoted efficient synthetic route towards glycospiro-pyrrolo[2,1-a]isoquinolines

A simple, convenient and efficient protocol for the construction of an array of glycospiro-pyrroloisoquinolines using isoquinolinium ylide and a carbohydrate-derived exocyclic olefin in the presence of Cu(OTf)₂–Et₃N catalytic system is described. Isoquinoline and alkylbromoacetates/2-bromoacetophenones were employed to generate the azomethine ylides in the presence of Et₃N in refluxing toluene and subsequent exposure to the olefin led to the desired isoquinoline derivatives.     

rac-BINAP-PdCl2 catalyzed Heck reactions of 3-formylquinolin-2-yl chlorides with methyl acrylate: synthesis of methyl 3-(3-formylquinolin-2-yl) acrylates

rac-BINAP-PdCl₂ catalytic system catalyzed Heck reaction of 3-formylquinolin-2-yl chlorides with methyl acrylate in DMA is described to the synthesis of methyl 3-(3-formyl-quinolin-2-yl)-acrylates, in good to excellent yields. The reaction could be also extended with other activated alkenes to afford Heck products. Fused-benzene ring in heterocyclic and carbocyclic moieties was found to enhance the yields.
