The exceptional leaving group ability of the trimethylstannyl group in electrophilic aromatic substitutions makes possible the synthesis, in a single step, of bi- and triaroylarenes through the catalyst-free, regioselective reaction of bi- and tristannylarenes with different aroyl halides in o-dichlorobenzene as solvent. Specific di- and triketones are obtained in good to excellent yields (45-83%).  相似文献

We used correlative transmission electron microscopy (TEM) and nanoscale secondary ion mass spectrometry (NanoSIMS) imaging to quantify the contents of subvesicular compartments, and to measure the partial release fraction of 13C-dopamine in cellular nanovesicles as a function of size. Three modes of exocytosis comprise full release, kiss-and-run, and partial release. The latter has been subject to scientific debate, despite a growing amount of supporting literature. We tailored culturing procedures to alter vesicle size and definitively show no size correlation with the fraction of partial release. In NanoSIMS images, vesicle content was indicated by the presence of isotopic dopamine, while vesicles which underwent partial release were identified by the presence of an 127I-labelled drug, to which they were exposed during exocytosis allowing entry into the open vesicle prior to its closing again. Demonstration of similar partial release fractions indicates that this mode of exocytosis is predominant across a wide range of vesicle sizes.  相似文献

The formation and the effects of laser irradiation of the complex formed by protoporphyrin IX (PPIX) and tubulin was investigated. We have used tubulin as a model protein to investigate whether docked photoactive ligands can affect the structure and function of polypeptides upon exposure to visible light. We observed that laser irradiation in the Soret band prompts bleaching of the PPIX, which is accompanied by a sharp decrease in the intensity and average fluorescence lifetime of the protein (dominated by the four tryptophan residues of the tubulin monomer). The kinetics indicate non‐trivial effects and suggest that the photosensitization of the PPIX bound to tubulin prompts structural alterations of the protein. These modifications were also observed through changes in the energy transfer between Trp residues and PPIX. The results suggest that laser irradiation produces localized partial unfolding of tubulin and that the changes prompt modification of the formation of microtubules in vitro. Measurements of singlet oxygen formation were inconclusive in determining whether the changes are prompted by reactive oxygen species or other excited state mechanisms.  相似文献

The spectroscopic behavior of 2-benzyl-2-methyl-2H-benzimidazole 1,3-dioxide derivatives in solution was studied in terms of electronic and nuclear magnetic resonance ((1)H and (13)C NMR) techniques. The experimental spectra were compared to the theoretical ones, obtained at DFT level, proving that the compounds adopt in solution a bird-like conformational distribution. Also, theoretically this conformational distribution resulted the most stable in gas phase. Infrared spectroscopy was used to study solid state behavior identifying experimentally the N-O stretching near to 1380, 1365 and 1225 cm(-1) and the vibrational benzimidazole skeleton near to 1610 and 1590 cm(-1). The vibrational spectrum was satisfactorily described by DFT calculations funding the N-O stretching as a coupled vibration near to 1470, 1350 and 1285 cm(-1). The fragmentation that takes place in mass spectrometry was assigned for all of the new derivatives.  相似文献

The effect of native cyclodextrins (alpha, beta, or gammaCD with six, seven and eight glucose units, respectively), hydroxypropyl-beta-cyclodextrin (HPCD), chitosan (CHT) and glucose in water solution or water with n-propylamine (PA) as co-solvent upon the UV-vis and fluorescence properties of poorly fluorescent N-methyl carbamates pesticides (C) as bendiocarb (2,2-dimethyl-1,3-benzodioxol-4-ol methylcarbamate, BC) and promecarb (3-methyl-5-(1-methylethyl)phenol methylcarbame, PC) was examined. Fluorescent enhancement was found for both substrates with all CDs in water or PA-water except from PC with alphaCD. The addition of CHT increases the fluorescence of BC but decreases the fluorescence of PC, and glucose addition gives in both cases no spectral changes. Host-guest interaction was clearly determined by fluorescence enhancement with betaCD and HPCD with a 1:1 stoichiometry for the complexes (C:CD). The values obtained for the association constants (K(A), M(-1)) were (6+/-2)x10(2) and (2.3+/-0.3)x10(2) for BC:betaCD and BC:HPCD complexes, respectively. For PC:betaCD and PC:HPCD the values of K(A) were (19+/-2)x10(2) and (21+/-2)x10(2), respectively. The ratio of the fluorescence quantum yields for the bound and free substrates (phi(CCD)/phi(C)) was in the range 1.74-3.8. The limits of detection (L(D), microg mL(-1)) for the best conditions were (0.57+/-0.02) for BC with HPCD and (0.091+/-0.002) for PC with betaCD in water. Application to the analysis in pesticide spiked samples of tap water and fruit yields satisfactory apparent recoveries (84-114%), and for the extraction procedure in fruits and a commercial formulation, recoveries were of 81-98% and 104%, respectively. The method is rapid, simple, direct, sensitive and useful for pesticide analysis.  相似文献

The reactivity of -SC(NH)NH2 (1), MeCOS- (2), and PhCOS- (3) toward 1-naphthyl radicals was studied in DMSO. The photostimulated reaction of anions 1, 2, and 3 with 1-bromonaphthalene (4) after quenching with MeI renders 1-(methylthio)naphthalene (6) as a main product together with bis(1-naphthyl) sulfide (7) and naphthalene (5). The thioacetate ion (2) and thiobenzoate ion (3) were unreactive toward 4 as electron-donor under photostimulation; however, in the presence of potassium tert-butoxide anion (entrainment conditions), they gave the mentioned products 5, 6, and 7, after the addition of MeI. Quenching of the triplet state of 4 was assigned as the photoinduced initiation step, with a rate constant value of (4.6+/-0.5)x10(8) M-1 s-1 for tert-butoxide anion and a rough estimated value of (8+/-7)x10(7) M-1 s-1 for anion 1. By using hydrogen abstraction from DMSO as the competitive reaction, the absolute rate constants for the addition of anions 1, 2, and 3 to 1-naphthyl radicals have been determined to be 1.0x10(9), 1.2x10(9), and 3.5x10(9) M-1 s-1, respectively. This reactivity order is in agreement with the stability of the resulting radical anions (ArNu)*- (10-12)*-. The inhibition experiments of the photoinduced substitution reaction in the presence of radical scavengers and the global quantum yield higher than the unity are evidence of a radical chain mechanism for these substitution reactions by anions 1 and 2. Anion 3 adds to the 1-naphthyl radical, but is neither able to initiate nor to keep the propagation cycle. Evaluation of the electron-transfer driving forces for the reaction between (ArNu)*- and 4 together with the absence of a chain reaction for the anion 3 indicate that the propagation in the proposed mechanism is given by an acid-base reaction between the radical .C(O)Me or .C(NH)NH2 (13) and a base.  相似文献
