Is the A-ring of sparteine essential for high enantioselectivity in the asymmetric lithiation-substitution of N-Boc-pyrrolidine?

The simplest chiral portion of sparteine, N,N'-dimethyl-2-endo-methylbispidine, was prepared and evaluated in the asymmetric lithiation-substitution of N-Boc-pyrrolidine. The results indicate that the complete A-ring of sparteine is essential for high levels of asymmetric induction. DFT-QSSR analyses of the diamine/Li(+) complexes and DFT calculations of the pertinent i-PrLi/diamine/N-Boc-pyrrolidine complexes are predictive and provide complementary pictures of the stereochemical features critical to this transformation.     

Environmentally Sustainable Achiral and Chiral Chromatographic Analysis of Amino Acids in Food Supplements

Two LC methods were developed for the achiral and chiral reversed-phase (RP) analysis of an amino acid (AA) pool in a food supplement, in compliance with the main paradigms of Green Chromatography. A direct achiral ion-pairing RP-HPLC method was optimized under gradient conditions with a water-ethanol (EtOH) eluent containing heptafluorobutyric acid (0.1%, v/v), to quantify the eight essential AAs (Ile, Leu, Lys, Met, Phe, Thr, Trp, and Val) contained in the food supplement. Thus, the usually employed acetonitrile was profitably substituted with the less toxic and more benign EtOH. The method was validated for Leu and Phe. The chiral LC method performed with a teicoplanin chiral stationary phase was developed with a water-EtOH (60:40, v/v) eluent with 0.1%, v/v acetic acid. The enantioselective analysis was carried out without any prior derivatization step. Both developed methods performed highly for all eight AAs and revealed that: (i) the content of six out of eight AAs was consistent with the manufacturer declaration; (ii) only L-AAs were present. Furthermore, it was demonstrated that a two-dimensional achiral–chiral configuration is possible in practice, making it even more environmentally sustainable. A molecular modelling investigation revealed interesting insights into the enantiorecognition mechanism of Lys.

Synthesis of pyrazino[1,2-a]benzimidazol-1(2H)ones via a microwave assisted Buchwald-Hartwig type reaction

A convenient synthesis of substituted pyrazino[1,2-a]benzimidazol-1(2H)ones starting from 3,5-dichloropyrazinones has been accomplished. Various 3-anilino-pyrazinones were easily converted to the desired tricyclic structures by applying a microwave assisted Buchwald-Hartwig type cyclization.

Ultra-thin-layer chromatography mass spectrometry and thin-layer chromatography mass spectrometry of single peptides of angiotensin-converting enzyme inhibitors

The separation of structurally related angiotensin-converting enzyme (ACE) inhibitors lisinopril, cilazapril, ramipril and quinapril and their corresponding active diacid forms (prilates) by conventional TLC silica gel 60 plates was contrasted with that afforded by monolithic ultra-thin-layer chromatographic (UTLC) plates. For the use of UTLC plates technical modifications of the commercially available equipments for the sample application, development and detection were made. Plates were developed in modified horizontal developing chamber using ethyl acetate-acetone-acetic acid-water (4:1:0.25:0.5, v/v). Detection of the separated compounds was performed densitometrically in absorption/reflectance mode at 220 nm and after exposure to iodine also by image analysis. The obtained results showed that monolithic layer is more efficient for the separation of structurally similar polar compounds, such as prilates than conventional silica layers. Identification of the compounds was confirmed by ESI-MS after their on-line extraction from the UTLC and TLC plates by means of Camag TLC-MS interface.     

Prescribed Gauss curvature problem on singular surfaces

We study the existence of at least one conformal metric of prescribed Gaussian curvature on a closed surface \(\Sigma \) admitting conical singularities of orders \(\alpha _i\)’s at points \(p_i\)’s. In particular, we are concerned with the case where the prescribed Gaussian curvature is sign-changing. Such a geometrical problem reduces to solving a singular Liouville equation. By employing a min–max scheme jointly with a finite dimensional reduction method, we deduce new perturbative results providing existence when the quantity \(\chi (\Sigma )+\sum _i \alpha _i\) approaches a positive even integer, where \(\chi (\Sigma )\) is the Euler characteristic of the surface \(\Sigma \).     

Map operations and k-orbit maps

A k-orbit map is a map with k flag-orbits under the action of its automorphism group. We give a basic theory of k-orbit maps and classify them up to k?4. “Hurwitz-like” upper bounds for the cardinality of the automorphism groups of 2-orbit and 3-orbit maps on surfaces are given. Furthermore, we consider effects of operations like medial and truncation on k-orbit maps and use them in classifying 2-orbit and 3-orbit maps on surfaces of small genus.
