Gold nanoparticles (Au NPs), which are extremely useful materials for imaging and photothermal therapy, typically require a drug delivery system to transport them to the affected tissue and into the cells. Since liposomes are approved as drug carriers, complexes of liposomes with Au NPs were considered ideal solutions to deliver Au NPs to the target site in vivo. In this study, we prepared complexes of various liposomes with Au NPs via physical absorption and characterized them. The time dependency of the surface plasmon resonance of this complex, which is a unique property of Au NPs, shows that the liposomes promote the formation of stable dispersions of Au NPs under isotonic conditions, even though intact Au NPs aggregate immediately. From a release assay of calcein from liposomes and transmission electron microscopy analysis, the Au NPs were complexed with liposomes without membrane disruption. These complexes could be formed by using cationic liposomes and polyethylene glycol-modified liposomes, as well as by using phosphatidylcholine liposomes, which are useful for drug and gene delivery. We proposed this kind of complex as a nanomedicine with diagnostic and therapeutic ability.  相似文献

Ampholytic polymers comprised of 2-(diethylamino)ethyl methacrylate (DEAEMA) and acrylic acid (AA) in varying molar ratios were used to modify egg yolk phosphatidylcholine (EYPC) liposomes previously encapsulated with pyranine. The modified EYPC liposomes were found to be pH responsive. The critical pH values at which the pyranine release from these liposomes at 37 °C were observed to decrease with increasing DEAEMA content in the polymers. Explanations were given for these phenomena.  相似文献

We have so far developed a new type of cationic lipid that consists of an amine-terminated poly(amidoamine) dendron and two long alkyl chains. In this study, we designed a dendron-bearing lipid (DL) modified with sugar moieties as the ligand at the terminal groups of the dendron to improve the transfection activities. We combined lactobionic acid at the end of every dendritic branch of the DL to produce galactose moieties-attached DL (Gal-DL). The lipoplexes containing Gal-DL exhibited enhancement of transfection of HepG2 cells. Moreover, Gal-DL significantly promoted the transfection activity of the lipoplexes including poly(ethylene glycol)-attached dendron-bearing lipid (PEG-DL).  相似文献

An in situ nitric acid analyzer has been designed for sensing in the remote atmosphere. Its operation is based on measuring the concentration difference between the total odd nitrogen species (NO(y)) and HNO(3)-scrubbed NO(y). NO(y) was measured by an NO-ozone chemiluminescent detector equipped with a molybdenum converter. A temperature-controlled annular denuder coated with NaCl was used as a scrubber of gaseous HNO(3). The current detection limit of the HNO(3) analyzer was estimated to be 158 and 71 pptv (parts per trillion by volume) with an integration time of 2 and 10 min (2sigma), respectively. The analyzer was calibrated using standard gaseous HNO(3) to verify its performance. The HNO(3) concentrations measured by the scrubber difference and the NO-ozone chemiluminescence methods (SDCL method) were in agreement with those obtained by the denuder method within the experimental uncertainties (more than +/-4%). We conducted limited field observations of NO(y) and HNO(3) in the remote atmosphere using the newly designed SDCL method at Cape Hedo, Okinawa, Japan.  相似文献

The first preparation of acridin‐9(10H)‐ones carrying a tertiary thiocarbamoyl group at C(10), i.e., N,N‐dialkyl‐9‐oxoacridine‐10(9H)‐carbothioamides 9 , is described. The method is based on the reaction of (2‐halophenyl)(2‐isothiocyanatophenyl)methanones 7 , prepared from (2‐aminophenyl)(2‐halophenyl)methanones 5 by a convenient three‐step sequence, with secondary amines in DMF at room temperature to generate the corresponding thiourea derivatives 8 in situ, which are treated with NaH at 100–120° to provide the desired products in one‐pot reactions in generally good yields.  相似文献

Series of double quantum dots each with a size around 400 × 400nm2 have been realized by delineating a 2DEG in modulation-doped AlGaAs/GaAs with 100 nm wide Schottky split gates fabricated by an electron-beam lithography and a lift-off technique. The split gate in the middle of the double dot allows us to control interdot coupling widely. The charging diagram obtained from linear transports in the Coulomb blockade regime shows that the isolated dots merge into a single composite dot with increase of interdot coupling. A clear Coulomb staircase has been observed in the double-dot system at a limited high-bias condition.  相似文献
