Ethanol extract of the aerial parts of Bergenia ligulata was subjected to solvent–solvent separation followed by various chromatographic techniques that lead to isolation of bergenine (1), p-hydroxybenzoyl bergenin (2), 11-O-galloylbergenin (3) and methyl gallate (4) as major constituents. Ethyl acetate fraction showed a dose-dependent urease inhibitory pattern with IC50 value of 54μg/mL. Structures of compounds 1 and 3 were established by XRD and 2, 4 by NMR. All these compounds were subjected to DPPH scavenging activity, reducing power assay and urease inhibitory activity. The EC50 7.45 ± 0.2 μg/mL and 5.39 ± 0.28 μg/mL values in terms of antioxidant and reducing power, respectively, were less for 3. Compounds 1–3 showed moderate to significant urease inhibitory potential with IC50 57.1 ± 0.7, IC50 48.4 ± 0.3 and 38.6 ± 1.5. Antioxidant activities and urease inhibitory potential were investigated and compound 3 was found to be the most active. 相似文献

In recent years, nanomaterials have made their way into hundreds of biomedical, life-sciences and technological applications. One such nanomaterial of extreme importance is nanoalumina (Al2O3 nanoparticles). This nanomaterial is an epitome of diversity with applications exhibited in the fields of catalysis, cosmetics, theranostics, energy generation, biosensors, drug-delivery, tumor-regression, etc. However, problems persist in terms of biocompatibility, cost-effectiveness, reproducibility and mass-production of nanoalumina by the presently existent physical, chemical and biological methodologies. Herein, we for the first time are presenting a top-down biofabrication method by which size reduction of commercial bulk alumina/aluminum oxide (5 µm) into nanoalumina (5–25 nm) is carried out by a thermophilic fungus Humicola sp. within 96 h of reaction at just 50 °C. The so-formed nanoalumina is highly stable, water dispersible, fluorescent and natural protein capped; characterization engaged standard techniques. These nanoparticles exhibit anti-bacterial properties against Gram-positive Bacillus subtilis strain and may serve as broad spectrum bactericidal agents. We believe that our novel top-down approach may be extensively used in the facile, inexpensive, eco-friendly and reliable fabrication of abundant quantities of nanomaterials of different chemical compositions, sizes and shapes with better control and predictability over the properties as derived from their substrates. A full field solution, based on small deformation, three-dimensional elastic–plastic finite element analysis of the centrally cracked thin disk under mode I loading has been performed. The solution for the stresses under small-scale yielding and lo!cally fully plastic state has been compared with the HRR plane stress solution. At the outside of the 3D zone, within a distance of rσo/J=18, HRR dominance is maintained in the presence of a significant amount of compressive stress along the crack flanks. Ahead of this region, the HRR field overestimate the stresses. These results demonstrate a completely reversed state of stress in the near crack front compared to that in the plane strain case. The combined effect of geometry and finite thickness of the specimen on elastic–plastic crack tip stress field has been explored. To the best of our knowledge, such an attempt in the published literature has not been made yet. For the qualitative assessment of the results some of the field parameters have been compared to the available experimental results of K, gives a fair estimate of the crack opening stress near the crack front at a distance of order 10−2 in. On the basis of this analysis, the Linear Elastic Fracture Mechanics approach has been adopted in analyzing the fatigue crack extension experiments performed in the disk (Part II). 相似文献

A method is described for the simultaneous determination of paracetamol, tizanidine, and diclofenac in mixtures. The method was based on HPLC separation of the three drugs followed by UV detection at 254 nm. The separation was carried out on a Hypersil ODS, C18 (250 x 4.6 mm id, 10 microm particle size) column using the mobile phase aqueous 0.2% ammonium carbonate-methanol (60 + 40, v/v) at a flow rate of 1 mL/min. The linear regression analysis data were used for the regression curve in the range of 170-10 000 ng/mL for paracetamol, 120-10 000 ng/mL for tizanidine, and 20-10 000 ng/mL for diclofenac. No chromatographic interference from tablet excipients was found. In order to check the selectivity of the proposed method, degradation studies were carried out using hydrolysis (acid, basic, and neutral), thermolysis, and oxidation. The developed method, after being validated in terms of precision, robustness, recovery, LOD, and LOQ, was successively applied to the analysis of pharmaceutical formulations and human serum. 相似文献

Okadaic acid (OA), responsible for gastrointestinal problems, inhibits protein phosphatase 2A (PP2A). Therefore, the inhibition exerted by the toxin on PP2A could be used to detect the presence of OA in aqueous solution and in shellfish sample.In this work, two commercial PP2As (from ZEU Immunotec and Millipore) and one produced by molecular engineering (from GTP Technology) were tested. Enzymes were used in solution and also immobilized within a polymeric gel. In solution, best performances were obtained using PP2A purchased from ZEU Immunotec (Spain). OA was detected in aqueous solution in concentration as low as 0.0124 μg L−1 using the enzyme from ZEU Immunotec whereas the detection limits were 0.47 μg L−1 and 0.123 μg L−1 with PP2As from Millipore and GTP Technology, respectively. Considering that the immobilization step contributes to stabilize the PP2A activity, enzymes were entrapped within a photopolymer and an agarose gel. These different polymeric matrices were optimized, tested and compared. Agarose gel seems to be a good alternative to the photopolymer largely used in our group. For instance, the IC50 value obtained with the test based on PP2A from ZEU Immunotec immobilized within an agarose gel was 1.98 μg L−1. This value was 1.8-fold lower than those obtained with the photopolymer test using the same enzyme. The proposed test is sensitive, fast and does not require expensive equipment. To evaluate the efficiency of the assay, PP inhibition tests based on PP2A from ZEU Immunotec in solution or immobilized within a gel were used for OA detection in contaminated shellfish. 相似文献

The change in the morphology of a series of dicationic gemini surfactants C(14)H(29)(CH(3))(2)N(+)-(CH(2))(s)-N(+)(CH(3))(2)C(14)H(29), 2Br(-) (14-s-14; s=4-6) on their interaction with inorganic (KBr, KNO(3), KSCN) and organic salts (NaBenz, NaSal) have been thoroughly investigated by means of (1)H NMR spectral analysis and the results are well supported by viscosity measurements. The presence of salt counterions results in structural transition (spherical to nonspherical) of gemini micelles in aqueous solution. With an increase in salt concentration all the three gemini surfactants showed changes in their aggregate morphology. This change is dependent on the nature and size of the added counterion. The effect of inorganic counterions on the micellar growth is observed to follow the Hofmeister series (Br(-) < NO(3)(-) < SCN(-)). The roles of organic counterions are discussed on the basis of probable solubilization sites of the substrate molecule in the gemini micelles, showing more growth in case of Sal(-) than Benz(-). The results are confirmed in terms of the obtained values of chemical shift (δ), line width at half height (lw), and relative viscosity (η(r)). Also, the growth of micelles was most pronounced for the gemini surfactant with the shortest spacer (s=4). This was attributed to the unique molecular structure of gemini surfactant micelles having flexible polymethylene spacer chain linking the twin polar headgroups. 相似文献
