The objective of this study is to estimate the contribution of Na+ as a counterion in the formation of H-AOT&Na-AOT-based W/O microemulsions using aqueous NaOH solution by pyranine absorbance measurements. A mixture of an aqueous NaOH solution containing pyranine/H-AOT&Na-AOT/isooctane was emulsified by changing the mixing ratio of Na-AOT (XNa-AOT = 0–1) and the mole fraction of NaOH (XNaOH = [NaOH]/the AOT concentration in the water pool = 0–1). The phase behavior of the emulsified mixture was evaluated from the absorbance of pyranine at the isosbestic point and by visual observations. W/O microelumsions are formed at the mid-range of XNa-AOT, whereas the emulsified mixture separates into two phases at lower XNa-AOT and higher XNa-AOT. The two phase boundaries shift toward lower XNa-AOT as with increasing XNaOH. The phase behavior depends on the degree of screening of electrostatic repulsions between the polar headgroups of AOT by the Na+ counterion. Interestingly, nano-sized W/O microemulsions are formed without phase separation using a highly concentrated NaOH aqueous solution when the Na-AOT mixing ratio is appropriately adjusted. The phase behavior was plotted as XNaOH versus XNa-AOT, and the correlation equations for the two phase boundaries were obtained by fitting the points. The contribution of the Na+ counterion from NaOH to W/O microemulsion formation was estimated by the correlation equations. The absorbance of pyranine and the size of W/O microemulsions, as measured by DLS, were plotted as a function of XNa+=(x[Na+   from   NaOH]+[Na+   from   Na-AOT])/[AOT], in which x is the ratio contributed by NaOH. The absorbance and size correlates well with XNa+, indicating that XNa+ is a meaningful parameter for quantitatively estimating phase behavior and size variation.  相似文献

Eight S-glycosylated 5,10,15,20-tetrakis(tetrafluorophenyl)porphyrins (1a′, 1b′, 1a and 1b (a: S-glucosylated, b: S-galactosylated)) and their 1,3-dipolar cycloadducts, i.e. chlorins 2a′, 2b′, 2a and 2b were prepared by nucleophilic substitution of the pentafluorophenyl groups with S-glycoside. These photosensitizers were characterized by 1H, 13C and 19F NMR spectroscopies and elemental analysis. The photocytotoxicity of the S-glycosylated photosensitizers and the parent porphyrin (1) and chlorin (2) was examined in HeLa cells. Photosensitizers 1, 2, 1a′, 1b′, 2a′ and 2b′ showed no significant photocytotoxicity at the concentration of 0.5 μM, while the deprotected photosensitizers 1a, 1b, 2a and 2b were photocytotoxic. The strong inhibition by sodium azide of the photocytotoxicity of these photosensitizers suggested that 1O2 is the main mediator. The S-glucosylated photosensitizers 1a and 2a showed higher photocytotoxicity than S-galactosylated 1b and 2b, respectively. The cellular uptake of 1a and 2a increased up to 24 h, while that of 1b and 2b was saturated by 12 h.  相似文献

Upon inclusion complexation with a racemic mixture of chiral fullerene C76 ((+/-)-C76), a novel cyclic host (1Rh) composed of a chiral N-methylporphyrin (PNMe) and a methylrhodium porphyrin (PRh) exhibited at 20 degrees C a 1H NMR spectral profile with diastereoisomerically split signals due to the NH group of PNMe. In contrast, because of a faster guest exchange dynamics, metal-free reference 12H, under identical conditions, did not show any signature of such diastereoisomeric splitting. Owing to the excellent resolution (15.6 Hz) of the split NH signals, the complexation with 1Rh allowed for the accurate determination of the optical purity of C76. On the basis of the 1H NMR spectral profiles of 1Rh in toluene-d8 at 20 degrees C in the presence of enantiomer mixtures of C76 of different optical purities, an absolute Deltaepsilon value of 58.5 M-1 cm-1 at 330 nm was obtained for enantiomerically pure C76.  相似文献

A semibiological molecular machine with an implemented "AND" logic gate was developed, which was capable of controlling the folding process of proteins in response to ATP and light as input stimuli. The molecular design made use of a genetically engineered chaperonin GroEL bearing, at both entrance parts of its cylindrical cavity, cysteine residues, which were functionalized by an azobenzene derivative to construct photoresponsive mechanical gates (azo-GroEL). This engineered chaperonin trapped denatured green fluorescent protein (GFP(denat)) and prohibited its refolding. However, when hosting azo-GroEL detected ATP (input stimulus 1) and UV light (input stimulus 2) at the same time, it quickly released GFP(denat) to allow its refolding. In contrast, reception of either input stimulus 1 or 2 resulted in only very slow or no substantial refolding of GFP(denat). Implementation of such "AND" logic gate mechanisms in mechanically driven biomolecular systems is an important step toward the design of secured drug delivery systems.  相似文献

In order to explore the influence of the sugar moieties of glycoconjugated chlorins on the photocytotoxicity, we studied the photochemical properties of four glycoconjugated chlorins in aqueous media such as cytoplasm and the concentration dependence of photocytotoxicity in HeLa cells. In phosphate-buffered saline, the fluorescence intensities of 5,10,15,20-tetrakis[3-(beta-D-glucopyranosyloxy)phenyl]chlorin (m-1a) and 5,10,15,20-tetrakis[3-(beta-D-galactopyranosyloxy)phenyl]chlorin (m-1b), i.e., chlorins having hexose groups, were about 2-fold greater than those of 5,10,15,20-tetrakis[3-(beta-d-xylopyranosyloxy)phenyl]chlorin (m-1c) and 5,10,15,20-tetrakis[3-(beta-d-arabinopyranosyloxy)phenyl]chlorin (m-1d), i.e., chlorins having pentose groups, owing to a sugar-dependent difference of aggregation behavior. While no cytotoxicity was found in the dark, the highest photocytotoxicity was shown by m-1a (82% inhibition) in HeLa cells. This was higher than those of m-1b, m-1c, m-1d and tetraphenylporphyrin tetrasulfonic acid. The glycoconjugated chlorins except for m-1b appeared to be distributed diffusely throughout the cytoplasm. Among the four photosensitizers, m-1a showed the highest intensity in confocal fluorescence images, in agreement with the in vitro photocytotoxicity results. For m-1c, no photocytotoxicity was found at drug concentrations from 0.2 to 0.04 microM. Hence, sugar-dependent aggregation is not the major reason for the unexpected lack of efficacy of m-1c, which is uptaken efficiently by HeLa cells. For the glycoconjugated chlorins, these results suggest the biological aspects of sugar moiety play much crucial role rather than chemical aspects.  相似文献

The Aconitum species (Ranunculaceae) are widely distributed in northern Asia and North America. Their roots are popularly used in herbal medicines in China and Japan. Many cases of accidental, suicidal and homicidal intoxication with this plant have been reported; some of these were fatal because the toxicity of Aconitum is very high. It is thus important to detect and quantify Aconitum alkaloids in body fluids, with high sensitivity. We have developed a simple and sensitive method for measuring four kinds of Aconitum alkaloids (aconitine, hypaconitine, jesaconitine and mesaconitine) by LC/electrospray (ESI)-time-of-flight (TOF)-MS. For all of them, only molecular ions were observed at an orifice voltage of 75 V; at 135 V, base peaks corresponding to [M - 60 + H]+ ions were observed. These four compounds and methyllycaconitine (internal standard) in human plasma samples were purified by solid-phase extraction. The four extracted compounds were completely separated in mass chromatograms; the calibration curves showed good linearity in the range 10-300 ng/ml, and the detection limits were estimated to be 0.2-0.5 ng/ml. Using our method, we also determined the amounts of these compounds in tuber samples. The present method is applicable in clinical and forensic toxicology.  相似文献

A phosphinite derivative that can be easily prepared in two steps from commercially available aminoindanol was found to be an effective catalyst for enantioselective acylation of diols. For the asymmetric desymmetrization of meso-1,2-diols, the corresponding monoester was obtained in up to 95% ee from the reaction in the presence of 5 mol % catalyst.  相似文献
