Gentamicin (GEN) is a kind of aminoglycoside antibiotic with the adverse effect of nephrotoxicity. Currently, no effective measures against the nephrotoxicity have been approved. In the present study, epigallocatechin gallate (EG), a useful ingredient in green tea, was used to attenuate its nephrotoxicity. EG was shown to largely attenuate the renal damage and the increase of malondialdehyde (MDA) and the decrease of glutathione (GSH) in GEN-injected rats. In NRK-52E cells, GEN increased the cellular ROS in the early treatment phase and ROS remained continuously high from 1.5 H to 24 H. Moreover, EG alleviated the increase of ROS and MDA and the decrease of GSH caused by GEN. Furthermore, EG activated the protein levels of nuclear factor erythroid 2-related factor 2 (Nrf2) and heme oxygenase-1 (HO-1). After the treatment of GEN, the protein level of cleaved-caspase-3, the flow cytometry analysis and the JC-1 staining, the protein levels of glutathione peroxidase 4 (GPX4) and SLC7A11, were greatly changed, indicating the occurrence of both apoptosis and ferroptosis, whereas EG can reduce these changes. However, when Nrf2 was knocked down by siRNA, the above protective effects of EG were weakened. In summary, EG attenuated GEN-induced nephrotoxicity by suppressing apoptosis and ferroptosis. 相似文献

Graphite anodes are well established for commercial use in lithium-ion battery systems. However, the limited capacity of graphite limits the further development of lithium-ion batteries. Hard carbon obtained from biomass is a highly promising anode material, with the advantage of enriched microcrystalline structure characteristics for better lithium storage. Tannin, a secondary product of metabolism during plant growth, has a rich source on earth. But the mechanism of hard carbon obtained from its derivation in lithium-ion batteries has been little studied. This paper successfully applied the hard carbon obtained from tannin as anode and illustrated the relationship between its structure and lithium storage performance. Meanwhile, to further enhance the performance, graphene oxide is skillfully compounded. The contact with the electrolyte and the charge transfer capability are effectively enhanced, then the capacity of PVP-HC is 255.5 mAh g−1 after 200 cycles at a current density of 400 mA g−1, with a capacity retention rate of 91.25%. Small-molecule near-infrared (NIR) imaging facilitates deep tissue penetration, low autofluorescence, non-invasive visualization, and a relatively simple operation. As such it has emerged as a popular technique for tracking biological species and events. However, the small Stokes shift of most NIR dyes often results in a low signal-to-noise ratio and self-quenching due to crosstalk between the excitation and emission spectra. With this research, we developed a NIR-based fluorescent probe WD-HOCl for hypochlorous acid (HOCl) detection using the NIR dye TJ730 as the fluorophore, which exhibits a large Stokes shift of 156 nm, with no crosstalk between the excitation and emission spectra. It contains acyl hydrazide as the responsive group and a pyridinium cation as the mitochondria-targeting group. The fluorescence intensity of WD-HOCl was enhanced by 30.1-fold after reacting with HOCl. Imaging studies performed using BV-2 cells indicated that WD-HOCl could be used for endogenous HOCl detection and imaging in living cells exposed to glucose and oxygen deprivation/reperfusion. The diphosphine complexes cis- or trans- PtCl2(P((CH2)n)3P ) (n = b/12, c/14, d/16, e/18) are demetalated by MC X nucleophiles to give the title compounds (P((CH2)n)3)P (3b–e, 91–71%). These “empty cages” react with PdCl2 or PtCl2 sources to afford trans- MCl2(P((CH2)n)3P ). Low temperature 31P NMR spectra of 3b and c show two rapidly equilibrating species (3b, 86 : 14; 3c, 97 : 3), assigned based upon computational data to in,in (major) and out,out isomers. These interconvert by homeomorphic isomerizations, akin to turning articles of clothing inside out (3b/c: ΔH‡ 7.3/8.2 kcal mol−1, ΔS‡ −19.4/−11.8 eu, minor to major). At 150 °C, 3b, c, e epimerize to (60–51) : (40–49) mixtures of (in,in/out,out) : in,out isomers, which are separated via the bis(borane) adducts 3b, c, e·2BH3. The configurational stabilities of in,out-3b, c, e preclude phosphorus inversion in the interconversion of in,in and out,out isomers. Low temperature 31P NMR spectra of in,out-3b, c reveal degenerate in,out/out,in homeomorphic isomerizations (ΔG‡Tc 12.1, 8.5 kcal mol−1). When (in,in/out,out)-3b, c, e are crystallized, out,out isomers are obtained, despite the preference for in,in isomers in solution. The lattice structures are analyzed, and the D3 symmetry of out,out-3c enables a particularly favorable packing motif. Similarly, (in,in/out,out)-3c, e·2BH3 crystallize in out,out conformations, the former with a cycloalkane solvent guest inside.It’s not a magic trick. Molecules can turn themselves inside out, just like articles of clothing or other familiar household objects. Feature selection (FS) is a vital step in data mining and machine learning, especially for analyzing the data in high-dimensional feature space. Gene expression data usually consist of a few samples characterized by high-dimensional feature space. As a result, they are not suitable to be processed by simple methods, such as the filter-based method. In this study, we propose a novel feature selection algorithm based on the Explosion Gravitation Field Algorithm, called EGFAFS. To reduce the dimensions of the feature space to acceptable dimensions, we constructed a recommended feature pool by a series of Random Forests based on the Gini index. Furthermore, by paying more attention to the features in the recommended feature pool, we can find the best subset more efficiently. To verify the performance of EGFAFS for FS, we tested EGFAFS on eight gene expression datasets compared with four heuristic-based FS methods (GA, PSO, SA, and DE) and four other FS methods (Boruta, HSICLasso, DNN-FS, and EGSG). The results show that EGFAFS has better performance for FS on gene expression data in terms of evaluation metrics, having more than the other eight FS algorithms. The genes selected by EGFAGS play an essential role in the differential co-expression network and some biological functions further demonstrate the success of EGFAFS for solving FS problems on gene expression data. 相似文献

用伊文思蓝(Evans blue, EB)作荧光探针研究了氨苄青霉素(Ampicillin, A)对牛血清白蛋白(Bovine serum albumin, BSA)的竞争反应. 伊文思蓝与牛血清白蛋白作用, 使牛血清白蛋白荧光发生猝灭, 根据Stern-Volmer方程及荧光寿命研究了荧光猝灭的类型及机理. 结果表明, 猝灭类型为静态猝灭, 即伊文思蓝和牛血清白蛋白形成了一种稳定的复合物. 伊文思蓝与牛血清白蛋白的结合常数KBSA-EB=1.122×106 L/mol, 结合点数n=0.9935, 并确定了EB和BSA之间的热力学常数及作用力类型. 当加入氨苄青霉素后, 牛血清白蛋白的相对荧光强度恢复. 这表明氨苄青霉素与伊文思蓝对牛血清白蛋白发生了竞争反应. 探讨了该竞争反应的相关机理, 求出了伊文思蓝与氨苄青霉素的结合常数为KEB-A=7.131×105 L/mol. 相似文献

The crude polysaccharide was obtained by means of the decolorization of porphyrized Cordyceps minlitaris stroma with organic solvent, extraction with hot water, precipitation in 80% ethanol, and protein removal with the Sevag method. After purification with Sephadex G-75, two of its components, CMP-1 and CMP-2, were obtained. Through the assay of gel chromatography and polarimetry, CMP-1 was identified as pure polysaccharide. The results demonstrated that CMP-1 had favorable oxidation resistance activity, which could scavenge not only oxygen-free radicals in the self-oxidation system of pyrogallic acid, but also the hydroxide-free radicals in the Fenton system. The study focused on the effects of low, medium, and high dosages of CMP-1 in rat blood serum on the proliferation of glomerular mesangial cells in vitro. Through MTT Colorimetric analysis, the activities were compared among the blank control group and the Niaoduqing positive control group CMP-1 and CMP-2. The results shows that CMP-1 was able to inhibit the proliferation of rat glomerular mesangial cells effectively. Therefore, CMP-1, one component of polysaccharides of Cordyceps minlitaris, was certainly a potential remedy for hyperplastic glomerular nephritis, whose antioxidant activity could slow down the process of chronic renal failure(CRF) to some extent. 相似文献

Scrophulariae Radix (SR) is one of the oldest and most frequently used Chinese herbs for oriental medicine in China. Before clinical use, the SR should be processed using different methods after harvest, such as steaming, “sweating”, and traditional fire-drying. In order to investigate the difference in chemical constituents using different processing methods, the two-dimensional (2D) 1H-13C heteronuclear single quantum correlation (1H-13C HSQC)-based metabolomics approach was applied to extensively characterize the difference in the chemical components in the extracts of SR processed using different processing methods. In total, 20 compounds were identified as potential chemical markers that changed significantly with different steaming durations. Seven compounds can be used as potential chemical markers to differentiate processing by sweating, hot-air drying, and steaming for 4 h. These findings could elucidate the change of chemical constituents of the processed SR and provide a guide for the processing. In addition, our protocol may represent a general approach to characterizing chemical compounds of traditional Chinese medicine (TCM) and therefore might be considered as a promising approach to exploring the scientific basis of traditional processing of TCM. 相似文献
