The effects of the insertion of a spin-labeled molecule (10-doxyl-stearic acid) on the structure and dynamics of model lipid bilayers in gel-like as well as in liquid-ordered-like phases are studied using molecular dynamic simulations. The perturbing effects of the labeled molecule on the structure of the bilayers are analyzed. We have also studied the relationship between the structural and dynamic properties of the bilayer phase and those of the labeled molecule. We found that the insertion of the labeled molecule in the bilayer at the concentration considered here (1:70) produces local and global perturbations in the gel-like phase. There is an increase of the area associated with the lipid molecules that produces a larger tilting angle of this condensed phase. In this gel-like phase, we also found that the z component of the order parameter of the labeled molecule associated with the electron paramagnetic resonance (EPR) spectra has the same temperature dependence as the axial correlation times of the lipid molecules. The mechanism by which the doxyl reorientation senses the dynamics of the layers is determined by the correlation between the gauche defect transitions of the labeled alkyl chain and its environment. For the liquid-ordered-like phase, we found that cholesterol molecules play the role of wedges that open free spaces in the lipid structure below the ring position and order the alkyl chains at the depths of the rings, leading to small inclination angles. The doxyl ring of the labeled molecule is located just below the cholesterol ring moiety, having fewer gauche defects than in the case of the gel-like phase. The change in depth of the doxyl ring causes a reorientation of this group that leads to an increase of the order parameter as the temperature rises.  相似文献

A microcontrolled, portable and inexpensive photometer is proposed. It uses a near infrared light emitting diode (NIR LED) as radiation source, a PbSe photoresistor as infrared detector and a programmable interrupt controller (PIC) microcontroller as control unit. The detector system presents a thermoresistor and a thermoelectric cooling to control the detector temperature and keep the noise at low levels. The microcontroller incorporated total autonomy on the proposed photometer. As its components are inexpensive and of easy acquisition, the proposed NIR LED-photometer is an economical alternative for chemical analyses in small routine, research and/or teaching laboratories. By being portable and microcontrolled, it also allows carrying out field chemical analyses. The instrument was successfully applied on the screening analysis to verify adulteration in gasoline samples.  相似文献

Composite electrodes were prepared using graphite powder and silicone rubber in different compositions. The use of such hydrophopic materials interned to diminish the swallowing observed in other cases when the electrodes are used in aqueous solutions for a long time. The composite was characterized for the response reproducibility, ohmic resistance, thermal behavior and active area. The voltammetric response in relation to analytes with known voltammetric behavior was also evaluated, always in comparison with the glassy carbon. The 70% (graphite, w/w) composite electrode was used in the quantitative determination of hydroquinone (HQ) in a DPV procedure in which a detection limit of 5.1x10(-8)molL(-1) was observed. HQ was determined in a photographic developer sample with errors lower then 1% in relation to the label value.  相似文献

The determination of fluoride ions in water samples is accomplished by using a tubular flow through detector constructed by drilling a channel through a commercially available LaF(3) crystal electrode in such a way that the original contacts of the non-modified unit are maintained. Its performance when incorporated in both FIA and SIA systems was evaluated and the results show that the tubular unit retains the characteristics of the non-modified electrode. In SIA conditions an extended linear range of response and lower detection limit were achieved when compared with the electrode performance in FIA conditions. These aspects together with the additional advantage of low sample and reagent consumptions in SIA when compared to FIA, makes the incorporation of the proposed tubular ISE in a SIA system the preferred approach for on line determination and monitoring of fluoride content in natural water samples.  相似文献

A DFT study on site selectivity in successive 1,3-dipolar cycloadditions of meso-tetraarylporphyrins with azomethine ylide and N-methylnitrone has been carried out. The calculation of the thermodynamic stability of both ylide and nitrone-derived adducts reveals that bacteriochlorins are more stable and have stronger aromatic character than isobacteriochlorins. Calculations of whole reaction pathways show that cycloadditions of azomethine ylide on porphyrin and its derived chlorin are irreversible and hence kinetically controlled. Solvent influence on the site selectivity of this reaction has also been considered, and appears to be decisive in controlling the site selectivity. In contrast, cycloadditions of nitrone over porphyrin and chlorin are clearly reversible, pointing to a thermodynamic control of these reactions.  相似文献

This study investigated the representativeness of samples for assessing chemical elements in milk bulk tanks. Milk samples were collected from a closed tank in a dairy plant and from an open top tank in a dairy farm. Samples were analyzed for chemical elements by instrumental neutron activation analysis (INAA). For both experiments, Br, Ca, Cs, K, Na, Rb and Zn did not present significant differences between samples thereby indicating the appropriateness of the sampling procedure adopted to evaluate the analytes of interest.  相似文献

The present work reports a simple and quick strategy for simultaneous determination of paracetamol (PC) and ascorbic acid (AA) in pharmaceutical formulations using flow injection method with multiple pulse amperometric detection. The method allows the resolution of the mixture without chemical pretreatment of the sample or electrode modification or the use of chemometric techniques for data analysis. The compounds are detected by applying four sequential pulses (waveform) in function of time to a three‐electrode amperometric system that uses a wall‐jet cell with gold as working electrode. AA is direct detected at +0.40 V and PC is indirectly detected at 0.0 V by the reduction (desorption) of the oxidation product (N‐acetyl‐p‐benzoquinoneimine) electrochemically generated at +0.65 V. The fourth potential pulse (?0.05 V) is applied for the complete regeneration (cleaning) of the gold electrode surface. The linear response range was optimized between 5 and 24 mg L?1 for AA and 50 and 240 mg L?1 for PC. The difference between the two responses ranges (10‐fold) present correlation with the concentration of these compounds in two different pharmaceutical formulations available in the Brazilian market. The analytical frequency was calculated in 60 injections per hour. The use of the proposed methodology for PC quantification in the presence of higher AA concentrations was also carried out. Using the standard addition method, it was possible to detect PC in trace levels (LD=0.2 mg L?1) in the presence of 880‐fold more of AA (176 mg L?1).  相似文献

The electrocatalysis of the oxygen reduction reaction by lanthanum-strontium manganate La0.5Sr0.5MnO3 (LSM) has been studied by cyclic voltammetry using the rotating ring-disc electrode technique (RRDE) in alkaline medium. From the ring-disc data and other kinetic parameters it was concluded that the oxygen reduction occurs by dissociative chemisorption at low overpotentials. At higher overpotentials, the formation of hydrogen peroxide (HO2 in this case) on the electrocatalyst has been observed. The apparent exchange current density for oxygen reduction on LSM has been found to be 2 × 10−7 A cm−2, while the corresponding Tafel slope is 0.100 V per decade. The possible reaction mechanism for electroreduction of oxygen on this oxide catalyst has been discussed. Published in Russian in Elektrokhimiya, 2008, Vol. 44, No. 7, pp. 919–923.
