Bicyclams inhibit HIV replication by binding to the CXCR4 chemokine receptor, which is the main coreceptor for gp120 used by X4, T‐tropic strains of HIV for membrane fusion and cell entry. Bicyclam AMD3100 mainly interacts with the aspartic acid residues namely Asp171 and Asp262, which are located at the extracellular ends in the CXCR4 coreceptor. Incorporation of some metal ions by the macrocyclic rings of bicyclam enhances its binding affinity to the CXCR4 receptor and enhances their anti‐HIV activity because the acetate can make a strong coordination bond to the metal and one weaker hydrogen bond to nitrogen in the cyclam ring. The interaction energy (Eint) between 150 metal–bicyclam complexes and aspartic acid has been evaluated. The metal–bicyclam complexes are obtained by the incorporation of six metal ions namely Fe3+, Co3+, Ni2+, Cu2+, Zn2+, and Pd2+ in 25 well‐known bicyclams including AMD3100. In most of the cases, Fe and Co–bicyclam complexes interact best with aspartic acid. The anti‐HIV activity of metal–bicyclam complexes can be predicted on the basis of interaction energy before the synthesis of the metal–bicyclam complex. Trietazine was selectively separated from aqueous solution containing the competitor molecule cyanazine, which is similar in size and shape to the template molecule. Structural features of the molecularly imprinted column were figured out by SEM. The influence of the mobile‐phase composition, applied electrical field, and pH of the mobile phase on the recognition of trietazine by the imprinted monolithic polymer has been evaluated, and the imprint effect in the trietazine‐imprinted monolithic polymer was demonstrated by an imprinting factor. The optimized monolithic column resulted in separation of trietazine from a structurally related competitor molecule, cyanazine. In addition, fast separation was obtained within 6 min by applying higher electrical field, with the electrophoretic mobility of 2.97 × 10?8 m2V?1s?1 at pH 11.0.  相似文献

The 4-(o-propargyloxy)styrylcoumarins are prepared by the condensation of O-propargylated salicylaldehyde with substituted coumarin-4-acetic acids. The intramolecular Diels–Alder reaction of 4-(o-propargyloxy)styrylcoumarins, without any catalyst gives fused-ring coumarins. The reaction in boiling nitrobenzene leads to aromatization of the initial Diels–Alder adduct and these aromatized products are highly fluorescent.  相似文献

The photochemistry of anti-inflammatory drug desonide (De, 1) was studied in aerobic as well as in anaerobic condition with different irradiation wavelengths (254, 310 nm) in acetonitrile and 2-propanol. All photoproducts obtained were isolated and characterized on the basis of IR, (1)H-, (13)C-NMR spectroscopy and elemental analysis study. The products were: 11beta,21-dihydroxy-16alpha,17alpha-(1-methylethylidenedioxy)-1,5-cyclopregn-3-ene-2,20-dione 2 (254 nm), 11beta-hydroxy-16alpha,17alpha-(1-methylethylidenedioxy)androsta-1,4-diene-3-one 3 (310 nm/2-propanol), 17beta-hydroperoxy-11beta-hydroxy-16alpha,17alpha-(1-methylethylidenedioxy)androsta-1,4-diene-3-one 4 (310 nm/O(2)/2-propanol). Cyclohexadienone moiety in ring A and keto group at C(17) were found to be deeply modified by UV light therefore, loss of biological activity both during storage and in vivo can not be ruled out.  相似文献

Silver chloride was found to be stable even after calcination at 650 °C for 10 h. SEM studies revealed the morphology of silver chloride as hexagonal particles. TEM studies show the size of silver chloride particles to have an average size of 6–7 nm. Thermal studies suggest that silver chloride nanoparticles behave like ionic liquid or molten salt in the range of 455–650 °C.  相似文献

Immunoaffinity adsorbent for transferrin (Tf) purification was prepared by immobilizing anti‐transferrin (Anti‐Tf) antibody on magnetic monosizepoly(glycidyl methacrylate) beads, which were synthesized by dispersion polymerization technique in the presence of Fe3O4nanopowder and obtained with an average size of 2.0 μm. The magnetic poly(glycidyl methacrylate) (mPGMA) beads were characterized by Fourier transform infrared spectroscopy, swelling tests, scanning electron microscopy, electron spin resonance spectroscopy, thermogravimetric analysis and zeta sizing analysis. The density and swelling ratio of the beads were 1.08 g/cm3 and 52%, respectively. Anti‐Tf molecules were covalently coupled through epoxy groups of mPGMA. Optimum binding of anti‐Tf was 2.0 mg/g. Optimum Tf binding from aqueous Tf solutions was determined as 1.65 mg/g at pH 6.0 and initial Tf concentration of 1.0 mg/mL. There was no remarkable loss in the Tf adsorption capacity of immunoaffinity beads after five adsorption–desorption cycles. Tf adsorption from artificial plasma was also investigated and the purity of the Tf molecules was shown with gel electrophoresis studies.  相似文献

Thirteen phases are now evidenced in the composition space diagram of the Al(OH)3tren–HF–ethanol system at 190 °C. Solvothermal syntheses are performed under microwave heating. Six new organic–inorganic fluorides crystallise and their structures are determined: (H3O)·[H4tren]2·(AlF6)3·6H2O (P-1, Z = 2), [H3tren]2·(AlF5(H2O))3·8H2O (C2/c, Z = 8), [H3tren]4·(AlF6)2·(Al2F11)·(F)·10H2O (P21/n, Z = 2), [H3tren]2·(Al4F18)·3.5H2O (P63, Z = 2), [H3tren]2·(Al4F18) (P-1, Z = 1), and [H3tren]4·(Al8F35)·(OH)·H2O (P-1, Z = 1). The existence domains are approximately located for all phases. Tren amine is tetraprotonated at high HF concentration, otherwise it is triprotonated. A protonated water cluster, H3O+(H2O)6, appears in (H3O)·[H4tren]2·(AlF6)3·6H2O while a new Al4F18 unit, found in [H3tren]2·(Al4F18), is evidenced; it results from corner and edge sharing association of four AlF6 octahedra. Finally, the structure of [H3tren]4·(Al8F35)·(OH)·H2O revealed the largest known fluoroaluminate polyanion, built from eight vertex sharing AlF6 octahedra, (Al8F35)11−.  相似文献

Reaction of N,N′‐(cyclohexane‐1,2‐diylidene)bis(4‐fluorobenzohydrazide), C20H18F2N4O2, ( LF ), with zinc chloride and mercury(II) chloride produced different types and shapes of neutral coordination complexes, namely, dichlorido[N,N′‐(cyclohexane‐1,2‐diylidene)bis(4‐fluorobenzohydrazide)‐κ2N,O]zinc(II), [ZnCl2(C20H18F2N4O2)], ( 1 ), and dichlorido[N,N′‐(cyclohexane‐1,2‐diylidene)bis(4‐fluorobenzohydrazide)‐κ4O,N,N′,O′]mercury(II), [HgCl2(C20H18F2N4O2)], ( 2 ). The organic ligand and its metal complexes are characterized using various techniques: IR, UV–Vis and nuclear magnetic resonance (NMR) spectroscopies, in addition to powder X‐ray diffraction (PXRD), single‐crystal X‐ray crystallography and microelemental analysis. Depending upon the data from these analyses and measurements, a typical tetrahedral geometry was confirmed for zinc complex ( 1 ), in which the ZnII atom is located outside the bis(benzhydrazone) core. The HgII atom in ( 2 ) is found within the core and has a common octahedral structure. The in vitro antibacterial activities of the prepared compounds were evaluated against two different bacterial strains, i.e. gram positive Bacillus subtilis and gram negative Pseudomonas aeruginosa bacteria. The prepared compounds exhibited differentiated growth‐inhibitory activities against these two bacterial strains based on the difference in their lipophilic nature and structural features.  相似文献

Solid-state NMR (ssNMR) spectroscopy facilitates the non-destructive characterization of structurally heterogeneous biomolecules in their native setting, for example, comprising proteins, lipids and polysaccharides. Here we demonstrate the utility of high and ultra-high field 1H-detected fast MAS ssNMR spectroscopy, which exhibits increased sensitivity and spectral resolution, to further elucidate the atomic-level composition and structural arrangement of the cell wall of Schizophyllum commune, a mushroom-forming fungus from the Basidiomycota phylum. These advancements allowed us to reveal that Cu(II) ions and the antifungal peptide Cathelicidin-2 mainly bind to cell wall proteins at low concentrations while glucans are targeted at high metal ion concentrations. In addition, our data suggest the presence of polysaccharides containing N-acetyl galactosamine (GalNAc) and proteins, including the hydrophobin proteins SC3, shedding more light on the molecular make-up of cells wall as well as the positioning of the polypeptide layer. Obtaining such information may be of critical relevance for future research into fungi in material science and biomedical contexts.  相似文献
