Chemotherapy is an important therapeutic approach for the treatment of cancer. Currently, many anticancer drugs are available in the market that plays an important role in cancer treatment, but concerns such as, drug resistance and side effects create an urgent need for the development of new anti-tumor drugs with high potency and less side effects. Heterocycles are of great interest due to their fascinating anticancer activity. Among them, 1,3,4-oxadiazoles showed attracting anti-tumor activity and its derivatives are under clinical trials for the treatment of cancer. Hybridization of 1,3,4-oxadiazole moiety with other heterocyclic pharmacophoresis a promising approach to overcome various disadvantages of current anticancer drugs such as drug resistance, toxicity, and other side effects. Thus, 1,3,4-oxadiazole-heterocycle hybrids occupy a significant position in the discovery of anti-tumor drugs. Among the reported oxadiazole-based hybrids reviewed here, compounds 45i, 59j, and 62x showed the highest anticancer activity with IC50 values in the nanomolar range. This review summarizes the recent developments in the anticancer potential, structure–activity relationships, and mechanisms of actions of 1,3,4-oxadiazole-heterocycle hybrids. 相似文献

Sorafenib is the first drug approved to treat advanced hepatocellular carcinoma (HCC) and continues as the gold-standard therapy against HCC. However, acquired drug resistance represents a main concern about sorafenib therapy. The flavanol quercetin found in plants has shown great anti-cancer and anti-inflammatory properties. In this work, quercetin was used as a therapeutic agent alone or in combination with a sorafenib chemotherapy drug to improve the routine HCC treatment with sorafenib. The in vitro and in vivo results presented here confirm that quercetin alone or in combination with sorafenib significantly inhibited HCC growth, induced cell cycle arrest and induced apoptosis and necrosis. Further molecular data shown in this report demonstrate that quercetin alone or combined with sorafenib downregulated key inflammatory, proliferative and angiogenesis-related genes (TNF-α, VEGF, P53 and NF-κB). Combined quercetin/sorafenib treatment markedly improved the morphology of the induced liver damage and showed significant antioxidant and anti-tumor effects. The advantage of combined treatment efficacy reported here can be attributed to quercetin’s prominent effects in modulating cell cycle arrest, apoptosis, oxidative stress and inflammation. 相似文献

The single‐electron transfer living radical polymerization (SET‐LRP) method in the presence of chain transfer agent was used to synthesize poly(N‐isopropylacrylamide) [poly(NIPAM)] with a low molecular weight and a low polydispersity index. This was achieved using Cu(I)/2,2′‐bipyridine as the catalyst, 2‐bromopropionyl bromide as the initiator, 2‐mercaptoethanol as the chain transfer agent (TH), and N,N‐dimethylformamide (DMF) as the solvent at 90 °C. The copper nanoparticles with diameters of 16 ± 3 nm were obtained in situ by the disproportionation of Cu(I) to Cu(0) and Cu(II) species in DMF at 22 °C for 24 h. The molecular weights of poly(NIPAM) produced were significantly higher than the theoretical values, and the polydispersities were less than 1.18. The chain transfer constant (Ctr) was found to be 0.051. Although the kinetic analysis of SET‐LRP in the presence of TH corroborated the characteristics of controlled/living polymerization with pseudo‐first‐order kinetic behavior, the polymerization also exhibited a retardation period (k > ktr). The influence of molecular weight on lower critical solution temperature (LCST) was investigated by refractometry. Our experimental results explicitly elucidate that the LCST values increase slightly with decreasing molecular weight. Reversibility of solubility and collapse in response to temperature well correlated with increased molecular weight of poly(NIPAM). © 2011 Wiley Periodicals, Inc. Novel hexa‐armed and pyrene (Pyr) end‐capped phosphazene dendrimer [N3P3‐(Pyr)6] and star polymer with poly(ε‐caprolactone) (PCL) arms [N3P3‐(PCL‐Pyr)6] were prepared via two series of reactions. In these series, core‐first approach was used starting from a hexa‐hydroxy functional phosphazene derivative (N3P3‐(OH)6). It was used as an initiator in the ring‐opening polymerization of ε‐caprolactone to prepare a hexa‐armed PCL star polymer (N3P3‐(PCL‐OH)6). Hydroxyl functionalities of N3P3‐(OH)6 and N3P3‐(PCL‐OH)6 were then successfully converted into bromide and azide, in turn. Further end‐group modifications of azide functional dendrimer precursor (N3P3‐(N3)6) and star polymer (N3P3‐(PCL‐N3)6) were achieved quantitatively via the Cu(I) catalyzed click reaction between azide functional groups and 1‐ethynyl pyrene in the final step. A reversible addition‐fragmentation chain transfer (RAFT) agent was directly anchored onto superparamagnetic Fe3O4 nanoparticles (SPNPs) in a simple procedure using a ligand exchange reaction of 2‐[(dodecylsulfanylcarbonylthiolsulfanyl) propionic acid] (DCPA) with oleic acid initially present on the surface of Fe3O4 nanoparticles. The DCPA‐modified SPNPs were then used for the surface‐mediated RAFT polymerization of di(ethylene glycol) ethyl ether acrylate and (oligoethylene glycol) methyl ether acrylate to fabricate structurally well‐defined hybrid SPNPs with temperature‐responsive poly[di(ethylene glycol) ethyl ether acrylate‐co‐(oligoethylene glycol) methyl ether acrylate] shell and magnetic Fe3O4 core. Evidence of a well‐controlled surface‐mediated RAFT polymerization was gained from a linear increase of number‐average molecular weight with overall monomer conversions and relatively narrow polydispersity indices of the copolymers grown from the SPNPs. The resultant hybrid nanoparticles exhibited superparamagnetic property with a saturation magnetization of 55.1–19.4 emu/g and showed a temperature‐responsive phenomenon as the temperature changed between 25 and 40 °C. © 2013 Wiley Periodicals, Inc. In this paper, the ($G′/G$)-expansion method is suggested to establish new
exact solutions for fractional differential-difference equations in the sense of modified
Riemann–Liouville derivative. The fractional complex transform is proposed to convert
a fractional partial differential difference equation into its differential difference
equation of integer order. With the aid of symbolic computation, we choose nonlinear
lattice equations to illustrate the validity and advantages of the algorithm. It is shown
that the proposed algorithm is effective and can be used for many other nonlinear

Focusing properties of an acoustic lens based on a modified triangular sonic crystal slab whose columns are aligned on concentric arcs of equal radial distance are investigated. Capability of focusing normally-incident plane waves is demonstrated by means of Finite Element Method. Focusing mechanisms are discussed on the basis of band structures and equifrequency contours considering a model where triangular lattice is elongated along ΓK direction. Focusing behavior of the proposed lens is argued to arise from negative refractions at the air-lens interfaces accompanied by index guiding in its interiors. Wavelength-order confinement in the transverse direction is observed. Double focusing is attributed to geometrical effects and contribution of self guiding is discussed. Possibility of occurrence of birefraction at the input face of the lens together with positive refraction at the output face is also discussed. 相似文献

In the present study, spiro (1a), dispiro (1b, 2, 3), per-substituted spermine-bridged (6–9) and dispiroansa spermine (10) derivatives of cyclotriphosphazene have been synthesized. The structures of the novel compounds (1b, 6–10) have been characterized by elemental analysis, FTIR, mass spectrometry, 1H and 31P NMR spectroscopy. The molecular structures of 1b, 2, 8, and 10 were determined by single crystal X-ray crystallography. In order to investigate the anti-tumour properties of the newly synthesized cyclotriphosphazene derivatives, in vitro cytotoxic activity test (MTT assay) has been performed using HT-29 (human colon adenocarcinoma) and Hep2 (human epidermoid larynx carcinoma) cell lines. The result of the MTT assay showed that while compound 1a has cytotoxic effect on both Hep2 and HT-29 cell lines, compound 3 has only cytotoxic effect towards the Hep 2 cells. 相似文献

Nucleophilic substitution reactions of cyclotriphosphazene derivatives having five-membered spiro rings, N(3)P(3)Cl(4)[O(CH(2))(2)X] (X = NH or O) with alkoxides (of tetraethylene glycol and some mono-functional alcohols) give unexpected rearrangements to form stable seven-membered ring ansa compounds, even though crystallographic evidence shows ring distortion and compression of the cyclophosphazene ring. With weaker nucleophiles such as sodium phenoxide and pyrrolidine substitution at a PCl2 group is preferred and no rearrangement takes place. In contrast, reactions of the analogous phosphazenes containing six-membered spiro rings, N(3)P(3)Cl(4)[O(CH(2))(3)X], with all of the above reagents give only normal substitution reactions at the PCl2 moieties and no rearrangement products. The spiro to ansa rearrangements in cyclophosphazenes are remarkable as the reported primary reaction products with the same difunctional reagents HO(CH(2))(2)XH are predominantly spiro, with some dangling and bridging derivatives, but no ansa compounds. 相似文献
