5,6-Dimethylbenz[a]phenazine ( 4 ), an aza analogue of the carcinogenic 5,6-dimethylbenz-[c]acridine has been obtained by a 1,1-dehydration-rearrangement (Wagner-Meerwin type) from 5,5-dimethyl-6-hydroxy-5,6-dihydrobenz[a]phenazine ( 3 ). Polyesters and poly(ester carbonates) were synthesized via ring‐opening polymerization with new tin(II) macroinitiator adducts containing oligomeric L ‐lactide (LLA), rac‐lactide (rac‐LA), and ?‐caprolactone (CL). The novel initiating species were synthesized by the reaction of LLA, rac‐LA, or CL with Sn(OEt)2 (monomer concentration/initiator concentration ≤20) and then were dissolved in methylene chloride or toluene and stored in a stoppered flask for the subsequent ring‐opening polymerization of cyclic esters and carbonates. The soluble tin alkoxide macroinitiators yielded predictable and quantitative initiation of polymerization for up to 1 month of storage time at room temperature. The resulting polymers displayed low polydispersity (≤1.5), and a high monomer conversion (>95%) was obtained within relatively short polymerization times (≤2 h). Adjusting the monomer/macroinitiator ratio effectively controlled the molecular weights of the polymers. NMR was used to characterize the initiating species and polymer microstructure, and size exclusion chromatography was used to determine the molecular weight properties of the polymers. © 2002 Wiley Periodicals, Inc. Poly(amidoamine) (PAMAM) dendrimers of different generations with carboxyl, acetyl, and hydroxyl terminal groups and a folic acid (FA)-dendrimer conjugate were separated and analyzed using reverse-phase high performance liquid chromatography (HPLC). Analysis of both the individual PAMAM derivatives and the separation of mixed generations can be achieved using a linear gradient 0-50% acetonitrile (ACN) (balance water) within 40 min. We also show that PAMAMs with defined acetylation and carboxylation degrees can be analyzed using HPLC. Furthermore, a generation 5 dendrimer-FA conjugate (G5.75Ac-FA4; Ac denotes acetyl) was analyzed and its specific binding with a bovine folic acid binding protein (FBP) was monitored. The HPLC and sodium dodecyl sulfate-polyacrylamide gel electrophoresis (SDS-PAGE) results indicate the formation of three complexes after the binding of G5.75Ac-FA4 with FBP. Dendrimers with FA moieties show much higher specific binding capability with FBP than those without FA moieties. Findings from this study indicate that HPLC is an effective technique not only for characterization and separation of functionalized PAMAM dendrimers and conjugates but also for investigation of the interaction between dendrimers and biomolecules. 相似文献
