Cryogenic storage of cell suspensions allows long-term maintenance of cultures. The main purpose of the study was to develop a successful cryogenic protocol for 10-year-old embryogenic cell suspensions of G. tibetica. We examined three techniques of freezing: (I) controlled-rate cooling with various cryoprotectants (0.1-0.5 M DMSO, 0.5-1.0 M sucrose, 0.5-1.0 M glycerol, 0.25-1.0 M proline) or preculture with 0.4 M sorbitol and cryoprotectants (0.065-0.1 M DMSO, 0.2-0.8 M proline), (II) vitrification (PVS2) and (III) encapsulation. Cell viability was assessed by the TTC test and biomass increase. After controlled-rate cooling the majority of cells were lethally damaged, with only 3% viability observed. Vitrification and encapsulation approaches were more effective, assuring high levels of post-thaw viability ca. 85% and 7%, respectively. The encapsulation procedure gave faster recovery of the culture suspension than did vitrification, and ensured culture homogeneity and embryogenic competence.  相似文献

'Herbal highs' have been advertised as legal and natural substitutes to cannabis, but a detailed examination of these products has revealed that the herbal matrix is laced with synthetic substances that mimic the effects of marijuana. Producers select the ingredients based on the results of scientific studies on the affinities of different chemicals to cannabinoid receptors. Naphthoylindoles have turned out to be the most popular class of substances identified in the products. Legal actions taken in order to tackle the problem of uncontrolled access to one substance have usually resulted in the marketing of derivatives or analogues. In the study, the mass spectral behavior of twelve synthetic cannabinoids from the naphthoylindole family under electrospray ionization (ESI) was investigated. LC-QTOFMS experiments were performed in three modes (low fragmentor voltage, high fragmentor voltage with/without collision energy), and they enabled the identification of protonated molecules and main ions. A general fragmentation pattern under this ionization method was proposed, and mechanisms of ion formation were discussed. The developed procedure allowed the determination of substituent groups of the core naphthoylindole structure and distinction between positional isomers. The obtained results were used for the prediction of the ESI-MS spectra for many naphthoylindoles with a high affinity to cannabinoid receptors. Similarities and differences between ESI-MS and electron impact-MS spectra of naphthoylindoles were discussed. The developed identification process was presented on an example of an analysis of an unknown herbal material, in which JWH-007 was finally identified. Knowledge of the fragmentation mechanisms of naphthoylindoles could also be used by other researchers for identification of unknown substances in this chemical family.  相似文献

The dynamical coupling between density gradients and particle transport has been investigated using similar experimental tools in the plasma boundary of different tokamak (JET, ISTTOK) and stellarator (TJ-II) devices, showing that the size of turbulent events is minimum in the proximity of the most probable density gradient. Experimental results were found to be consistent with results from two very different models of plasma turbulence and transport. The present findings, common to several plasma devices, suggest the importance of self-regulation mechanisms between plasma transport and gradients in fusion devices.  相似文献

This paper deals with the photo-induced fluorimetric determination of the herbicide Fluometuron with the aid of a continuous-flow assembly of the emergent and new methodology known as Multicommutation which was provided with an on-line photoreactor. Maximum fluorescence intensity was observed at basic pH solutions, 1×10−4 mol l−1 NaOH, after 1.4 min of irradiation and being the maximum at λexc 247.0 nm and λem 325.0 nm. The influence of different experimental parameters either chemical (pH, surfactants presence, solvent polarity and temperature) or hydrodynamic (time of photo-degradation, size and number of different segments and flow-rate) was tested. The linear dynamic range was from 0.01 to 4.0 mg l−1 of Fluometuron; the inter-day reproducibility (as R.S.D.) of the slope was 0.001% and 1.7% from the peaks intra-day reproducibility. A large series of potential interferents was studied and finally the method was applied to human urine, soil, formulation and water samples.  相似文献

We experimentally demonstrate the first integrated temporal Fourier transformer based on a linearly chirped Bragg grating waveguide written in silica glass with a femtosecond laser. The operation is based on mapping the energy spectrum of the input optical signal to the output temporal waveform by making use of first-order chromatic dispersion. The device operates in reflection, has a bandwidth of 10 nm, and can be used for incident temporal waveforms as long as 20 ps. Experimental results, obtained through both temporal oscilloscope traces and Fourier transform spectral interferometry, display a successful Fourier transformation of in-phase and out-of-phase pairs of input optical pulses, and demonstrate the correct functionality of the device for both amplitude and phase of the temporal output.  相似文献

Spontaneous formation of solitary wave structures has been observed in Earth's magnetopause, and is shown to be caused by the breakup of a zonal flow by the action of drift wave turbulence. Here we show matched observations and modeling of coherent, large-scale solitary electrostatic structures, generated during the interaction of short-scale drift wave turbulence and zonal flows at the Earth's magnetopause. The observations were made by the Cluster spacecraft and the numerical modeling was performed using the wave-kinetic approach to drift wave-zonal flow interactions. Good agreement between observations and simulations has been found, thus explaining the emergence of the observed solitary structures as well as confirming earlier theoretical predictions of their existence.  相似文献

We show that, in a nanometric size stable electrodeposited Ni contact, it is possible to modify the magnetoresistance by applying current pulses and external magnetic fields whereby the same current path is used for detection and modification. We can pass from positive to negative magnetoresistance with values as large as 25% at room temperature, all in the same contact. We propose that the effect may be due to switching and moving domain walls in the contact region under the combination of current effects and external fields.  相似文献
