Anisotropic friction model of DNA electrophoresis in polymer solutions: Comparison with direct observations

The electrophoresis of DNA chains in uncrosslinked polymer solutions with a Brownian dynamics simulation with an anisotropic friction tensor was analyzed. According to the degree of anisotropy, three types of migration behavior are obtained: fluctuation without or with periodicity between U‐shaped and compact conformations, or migration with linear conformation. We found good agreement between our simulation results and the direct observations of DNA by fluorescence microscopy. © 2003 Wiley Periodicals, Inc. Mutation detection in the drug-resistant hepatitis B virus polymerase gene using nanostructured reverse micelles.

The emergence of drug-resistant hepatitis B virus (HBV) has been reported in patients with prolonged administration of lamivudine, which is a potent drug for the prevention of HBV infection. Lamivudine-resistant HBV has several types of mutations at the YMDD motif of its DNA polymerase. We successfully demonstrated that monitoring the hybridization behavior in nanostructured reverse micelles enables us to detect single nucleotide polymorphisms (SNPs). With the aid of reverse micelles, a model 40-mer oligonucleotide containing a single-base substitution was clearly distinguished from the normal, complementary oligonucleotide. In addition, we extended this technique to a high-throughput analysis. The results obtained with a 96-well micro-plate reader indicated the possibility of SNPs detection toward multiple samples of patients.     

Addition–fragmentation chain transfer: Molecular weight distributions and retardation in the system methyl methacrylate/methyl α‐(bromomethyl)acrylate

A detailed investigation of addition–fragmentation chain transfer (AFCT) in the free‐radical polymerization of methyl methacrylate (MMA) in the presence of methyl α‐(bromomethyl)acrylate (MBMA) was carried out to elucidate mechanistic details with efficient macromonomer synthesis as an underlying goal. Advanced modeling techniques were used in connection with the experimental work. Curve fitting of simulated and experimental molecular weight distributions with respect to the rate coefficient for addition of propagating radicals to MBMA (k_add) over 60–120 °C resulted in E_add = 21.7 kJ mol^?1 and A_add = 2.18 × 10⁶ M^?1 s^?1 and a very weak temperature dependence of the chain‐transfer constant (E_add ≈ E_p). The rate coefficient for fragmentation of adduct radicals at 60 °C was estimated as k_f ≈ 39 s^?1 on the basis of experimental data of the MMA conversion and the concentration of 2‐carbomethoxy‐2‐propenyl end groups. The approach developed is generic and can be applied to any AFCT system in which copolymerization does not occur and in which the resulting unsaturated end groups do not undergo further reactions. © 2004 Wiley Periodicals, Inc. The crystal structure and absolute configuration of the antitumor platinum complex trans(OH)-Pt(OH)2(malonato)(1R,2R-cyclohexanediamine).

The absolute configuration of the anti-tumor complex trans(OH)-Pt(OH)2(malonato)(1R,2R-cyclohexanediamine) was determined by X-ray anomalous scattering technique. The final unit cell was monoclinic, space group P2(1) with a = 9.142 A, b = 7.788 A, c = 11.946 A, beta = 96.48 degrees, Z = 2. The crystal structure was determined by direct method and difference Fourier synthesis, and refined to R = 0.025 and Rw = 0.033 based on 2768 independent reflections. The platinum atom has roughly octahedral coordination. The cyclohexane ring has the expected chair configuration, with two amino groups in equatorial positions while the malonato ligand, in contrast, shows a boat conformation for six membered Pt O-C-C-C-O ring.     

Studies on steroids. CCLIII. Capillary gas chromatographic behaviour of diethylhydrogensilyl-diethylsilylene derivatives of stereoisomeric bile acids.

The capillary gas chromatographic behaviour of diethylhydrogensilyl (DEHS) ethers and/or diethylsilylene (DES) derivatives of fifty bile acids including 4- and 6-hydroxylated compounds is described. The methylene unit (MU) values of methyl and pentafluorobenzyl esters of bile acids were determined as their trimethylsilyl (TMS), dimethylethylsilyl (DMES) ethers and DEHS-DES derivatives. The differences in methylene unit values between the corresponding TMS ethers and DMES ethers or DEHS-DES derivatives were used for estimating the number and stereochemistry of hydroxyl groups on the steroid nucleus. On treatment with the silylating agent N,O-bis (diethylhydrogensilyl)trifluoroacetamide, bile acids possessing isolated hydroxyl in addition to diaxial trans-glycol groups were easily converted into the DEHS ehters, whereas those having a vicinal glycol group except for the diaxial group were converted into cyclic DES derivatives. The mass spectrometric properties obtained with negative-ion chemical ionization detection are discussed.     

Synthesis of peptides and oligosaccharides by using a recyclable fluorous tag

A hexakis(fluorous chain)-type alcohol was used in the synthesis of oligosaccharides and peptides through connection with a linker suitable for the particular type of target compound. After the preparation of the desired compound, the fluorous alcohol was easily recovered in good yields under basic conditions. It appears that the fluorous alcohol can be recovered, recycled, and reused.     

Determination of some rare-earth elements by plasma-jet emission spectrometry

The effects of a magnetic field on spectral intensities in plasma-jet spectrometry were examined, and detection limits for rare-earth elements were calculated. Plasma-jet emission spectrometry was applied to the determination of lanthanum, yttrium and gadolinium in a monazite sample from which thorium had been separated. A standard addition method was used in order to improve accuracy, and the internal standard and background compensation method was applied to measurements of spectral line intensities to achieve good precision. The coefficient of variation was 1.51% for 50 μg La ml^?1.     

Separation of N2-ethyl-2'-deoxyguanosine-5'-monophosphate and four native deoxyribonucleoside monophosphates using capillary zone electrophoresis with polyethylene glycol as buffer additive

We investigated the separation of five deoxyribonucleoside monophosphates: 2'-deoxyguanosine-5'-monophosphate (dGMP), 2'-deoxyadenosine-5'-monophosphate (dAMP), 2'-deoxycytosine-5'-monophosphate (dCMP), 2'-deoxythymidine-5'-monophosphate (dTMP) and a dGMP adduct possessing N2-ethyl-guanine, which has been noted in relation to mutagenesis of alcohol, using capillary zone electrophoresis (CZE). The concentration of polyethylene glycol (PEG) as a modifier and the pH of the running solutions can efficiently control the observed separation. Interaction of PEG with analytes was quantitatively evaluated. PEG worked effectively as a hydrophobic selector in these separations. The values of pKa of the acidic-NH-groups in the base moieties of dGMP, dTMP, and the dGMP adduct are close to that of boric acid used as buffer of the running solutions. The control of their charge was facilitated, enabling improved separations. A more sufficient and fast separation was achieved by both optimization of pH of the running solutions and PEG concentration compared with that obtained by pH control alone. On-line concentration using a stacking method followed by the PEG-assisted CZE was briefly studied.     

Total synthesis of slow reacting substances (SRS's): 6-epi-leukotriene C and 6-epi-leukotriene D

6-$\text{Epi}$-leukotrienes C and D (3 and 4) have been synthesized unambiguously via the 5($\text{S}$), 6($\text{R}$)-epoxide (5,6-$\text{cis}$) which is isomeric with leukotriene A. These 6-$\text{epi}$-leukotrienes are less active (especially 4) than leukotrienes C and D (1 and 2) and have not been found in substantial quantity in natural SRS sources.
