A simple and sensitive fluorometric high-performance liquid chromatographic method was developed for the determination of midodrine in human plasma. After liquid-liquid extraction from plasma, the drug and 2-phenylglycinol (internal standard) were convened into the corresponding fluorescent derivatives by reaction with 3,4-dihydro-6,7-dimethoxy-4-methyl-3-oxoquinoxaline-2-carbonyl chloride, a fluorescence derivatization reagent for amines. The derivatives were separated within 30 min on a reversed-phase column using isocratic elution with acetonitrile-methanol-water (10:30:60, v/v) and were detected spectrofluorometrically at 485 nm with excitation at 400 nm. In this paper, we discuss two key aspects of magnetisation reversal in magnetic thin films with perpendicular anisotropy. Firstly, a study has been made of the additional field required to erase data written perpendicular to a thin film recording disk as the linear data density is increased. It has been found that an increase in data density from 40 to 360 kfci results in an increase of 1.25 kOe in the field required to erase the data. Secondly, the effect of varying the level of exchange coupling by co-sputtering CoCrPt samples with SiO2 has been studied using a characterisation technique that is independent of the self-demagnetising field. It has been found that the samples are fully exchange de-coupled when the film contains >9.8% SiO2 and the activation volume of reversal remains constant for higher levels of SiO2. 相似文献

An ammonium-sulfate-precipitable (33–70%) fraction in extracts from eggs of silkworm Bombyx mori contains photoreactivating enzyme that reactivates the transforming activity of UV inactivated Hemophilus influenzae DNA. The action spectrum for in vitro photoreactivation with the enzyme has a broad peak around 365–385 nm, with a shoulder extending to 460 nm. This relatively higher photoreactivation efficiency at wavelengths longer than 450 nm seems to be a unique feature of DNA photoreactivating enzyme of silkworm. Using gel filtration, a mol wt of 42,000 was estimated for the enzyme. Optimum and isoionic pH of the enzyme were 7.2 and 5.4, respectively. These properties of silkworm enzyme are within the range of variations in reported biochemical characteristics of photoreactivating enzymes from different species. 相似文献

Modified 3-aminopropylsilyl silica stationary phases with 1,8-naphthalic anhydrides [3-(1,8-naphthalimido)propyl (NAIP), 3-(4-amino-1,8-naphthalimido)propyl (4A-NAIP), 3-(3-nitro-1,8-naphthalimido)propyl (3N-NAIP), and 3-(4-nitro-1,8-naphthalimido)propyl modified silyl silica gel (4N-NAIP)] are prepared in a single reaction. Chromatographic properties of these phases are characterized using various solutes including monosubstituted benzenes, alkyl benzenes, and polycyclic aromatic hydrocarbons (PAHs) in high-performance liquid chromatography. Even though the retention in these NAIPs is mainly based on the reversed-phase mode, 3N- and 4N-NAIP show a remarkable recognition toward aromatic hydrocarbons, which is attributed to their strong pi-pi interaction. Furthermore, the recognition ability is according to the molecular planarity and non-linearity of PAHs. Subsequently, 3N- and 4N-NAIP has a distinct selectivity from octadecyl silica and, therefore, will be a valuable alternative for a separation or a solid-phase extraction of aromatic compounds. 相似文献

Three new pseudoguaiane-type sesquiterpenes, dichrocepholides A-C, and two new pseudoguaiane-type sesquiterpene dimers, dichrocepholides D and E, were isolated from the aerial part of Dichrocephala integrifolia. Their stereostructures were determined on the basis of chemical and physicochemical evidence. The reaction of 1,2-dimethoxytetramethyldisilane with styrene and α-methylstyrene in the presence of NaOMe catalyst in tetrahydrofuran (THF) gave the new silacyclopentanes 1,1-dimethyl-2,4-diphenyl-1-silacyclopentane (IIIa) and 1,1,2,4-tetramethyl-2,4-diphenyl-1-silacyclopentane (IIIb), respectively. These silacyclopentanes were found to exist as cis-trans mixtures. The use of sodium metal in place of NaOMe afforded similar results. Reactions of a polysilane mixture, MeO-(SiMe2)nOMe (n ≧ 3), with the styrenes also gave similar results. In some cases, polysilacycloalkanes such as 1,2,3-trisilacyclopentanes (IV) and 1,2,3,4-tetrasilacyclohexanes (V) were obtained as by-products. A mechanism for the formation of the silacyclopentanes and polysilacycloalkanes is presented. It was found that electron impact decomposition of silacyclopentanes IIIa and IIIb, trisilacycloalkane IV and tetrasilacycloalkane V gave molecular ions corresponding to the silacyclopropane, cyclotrisilane and cyclotetrasilane systems. 相似文献

A fluorimetric liquid chromatographic method for the determination of 5-hydroxyindoles based on the benzylamine derivatization process mediated through an online photocatalytic oxidation has been developed. In this study, we used a photocatalytic column comprising tefzel tubing packed with TiO2-coated glass beads, as a pre-column derivatization reactor. The fluorescence derivatization of 5-hydroxyindoles using benzylamine proceeded during their passage through the reaction column under near-UV irradiation. The 5-hydroxyindole derivatives were separated continuously on a reversed-phase liquid chromatography within 50 min, using 100 mM acetate buffer (pH 4.6)-acetonitrile (72:28, v/v; isocratic elution) containing 3 mM sodium octanesulfonate; the samples were detected fluorimetrically at 465 nm upon excitation at 350 nm. The detection limits (signal-to-noise ratio = 3) of the 5-hydroxyindoles were in the range from 160 to 360 fmol per 5 μL injection. We have applied this method, which requires minimal sample pre-treatment, to the determination of 5-hydroxyindole-3-acetic acid in human urine. 相似文献

6-Rhodamine B amine functions as a highly sensitive fluorescence derivatization reagent for mono- and oligosaccharides; it reacts with the reducing end of saccharides under acidic conditions. The fluorescent derivatives of five monosaccharides can be separated within 25 min by reversed-phase liquid chromatography with isocratic elution. The detection limits (S/N = 3) for mono-, di-, and oligosaccharides are 7-51, 13, and 9-35 fmol/20 microl injection, which correspond to analyte concentrations of 35-255, 65, 45-175 nM, respectively. We have applied this derivatization method successfully to the analysis of the components of oligosaccharides in glycoproteins (ribonuclease B and fetuin) following their acidic or enzymatic hydrolysis. The results from these analyses are in good agreements with the reported values established previously. 相似文献
