Antibiotic resistance has increased over the past two decades. New approaches for the discovery of novel antibacterials are required and innovative strategies will be necessary to identify novel and effective candidates. Related to this problem, the exploration of bacterial targets that remain unexploited by the current antibiotics in clinical use is required. One of such targets is the \(\beta \) -ketoacyl-acyl carrier protein synthase III (FabH). Here, we report a ligand-based modeling methodology for the virtual-screening of large collections of chemical compounds in the search of potential FabH inhibitors. QSAR models are developed for a diverse dataset of 296 FabH inhibitors using an in-house modeling framework. All models showed high fitting, robustness, and generalization capabilities. We further investigated the performance of the developed models in a virtual screening scenario. To carry out this investigation, we implemented a desirability-based algorithm for decoys selection that was shown effective in the selection of high quality decoys sets. Once the QSAR models were validated in the context of a virtual screening experiment their limitations arise. For this reason, we explored the potential of ensemble modeling to overcome the limitations associated to the use of single classifiers. Through a detailed evaluation of the virtual screening performance of ensemble models it was evidenced, for the first time to our knowledge, the benefits of this approach in a virtual screening scenario. From all the obtained results, we could arrive to a significant main conclusion: at least for FabH inhibitors, virtual screening performance is not guaranteed by predictive QSAR models. 相似文献

Let V be a module with . V is called a quasi-Baer module if for each ideal J of S, for some . On the other hand, V is called a Rickart module if for each , for some . For a module N, the quasi-Baer module hull (resp., the Rickart module hull ) of N, if it exists, is the smallest quasi-Baer (resp., Rickart) overmodule, in a fixed injective hull of N. In this paper, we initiate the study of quasi-Baer and Rickart module hulls. When a ring R is semiprime and ideal intrinsic over its center, it is shown that every finitely generated projective R-module has a quasi-Baer hull. Let R be a Dedekind domain with F its field of fractions and let be any set of R-submodules of . For an R-module with , we show that has a quasi-Baer module hull if and only if is semisimple. This quasi-Baer hull is explicitly described. An example such that has no Rickart module hull is constructed. If N is a module over a Dedekind domain for which is projective and , where is the torsion submodule of N, we show that the quasi-Baer hull of N exists if and only if is semisimple. We prove that the Rickart module hull also exists for such modules N. Furthermore, we provide explicit constructions of and and show that in this situation these two hulls coincide. Among applications, it is shown that if N is a finitely generated module over a Dedekind domain, then N is quasi-Baer if and only if N is Rickart if and only if N is Baer if and only if N is semisimple or torsion-free. For a direct sum of finitely generated modules, where R is a Dedekind domain, we show that N is quasi-Baer if and only if N is Rickart if and only if N is semisimple or torsion-free. Examples exhibiting differences between the notions of a Baer hull, a quasi-Baer hull, and a Rickart hull of a module are presented. It is well known that the Rickart property of rings is not a left-right symmetric property. We extend the notion of the left Rickart property of rings to a general module theoretic setting and define 𝔏-Rickart modules. We study this notion for a right R-module M R where R is any ring and obtain its basic properties. While it is known that the endomorphism ring of a Rickart module is a right Rickart ring, we show that the endomorphism ring of an 𝔏-Rickart module is not a left Rickart ring in general. If M R is a finitely generated 𝔏-Rickart module, we prove that End R (M) is a left Rickart ring. We prove that an 𝔏-Rickart module with no set of infinitely many nonzero orthogonal idempotents in its endomorphism ring is a Baer module. 𝔏-Rickart modules are shown to satisfy a certain kind of nonsingularity which we term “endo-nonsingularity.” Among other results, we prove that M is endo-nonsingular and End R (M) is a left extending ring iff M is a Baer module and End R (M) is left cononsingular. 相似文献

We present a parallelization of the revised simplex method for large extensive forms of two-stage stochastic linear programming (LP) problems. These problems have been considered too large to solve with the simplex method; instead, decomposition approaches based on Benders decomposition or, more recently, interior-point methods are generally used. However, these approaches do not provide optimal basic solutions, which allow for efficient hot-starts (e.g., in a branch-and-bound context) and can provide important sensitivity information. Our approach exploits the dual block-angular structure of these problems inside the linear algebra of the revised simplex method in a manner suitable for high-performance distributed-memory clusters or supercomputers. While this paper focuses on stochastic LPs, the work is applicable to all problems with a dual block-angular structure. Our implementation is competitive in serial with highly efficient sparsity-exploiting simplex codes and achieves significant relative speed-ups when run in parallel. Additionally, very large problems with hundreds of millions of variables have been successfully solved to optimality. This is the largest-scale parallel sparsity-exploiting revised simplex implementation that has been developed to date and the first truly distributed solver. It is built on novel analysis of the linear algebra for dual block-angular LP problems when solved by using the revised simplex method and a novel parallel scheme for applying product-form updates. 相似文献

Deterministic sample average approximations of stochastic programming problems with recourse are suitable for a scenario-based parallelization. In this paper the parallelization is obtained by using an interior-point method and a Schur complement mechanism for the interior-point linear systems. However, the direct linear solves involving the dense Schur complement matrix are expensive, and adversely affect the scalability of this approach. We address this issue by proposing a stochastic preconditioner for the Schur complement matrix and by using Krylov iterative methods for the solution of the dense linear systems. The stochastic preconditioner is built based on a subset of existing scenarios and can be assembled and factorized on a separate process before the computation of the Schur complement matrix finishes on the remaining processes. The expensive factorization of the Schur complement is removed from the parallel execution flow and the scaling of the optimization solver is considerably improved with this approach. The spectral analysis indicates an exponentially fast convergence in probability to 1 of the eigenvalues of the preconditioned matrix with the number of scenarios incorporated in the preconditioner. Numerical experiments performed on the relaxation of a unit commitment problem show good performance, in terms of both the accuracy of the solution and the execution time. 相似文献

Investigations performed on silicon nanowires of different lengths by scanning electron microscopy revealed coalescence processes in longer nanowires. Using X‐ray diffraction (XRD), it was found that the shape of the pole figure in reciprocal space is ellipsoidal. This is the signature of lattice defects generated by the relaxation of the strain concentrated in the coalescence regions. This observation is strengthened by the deviation of the XRD peaks from Gaussianity and the appearance of the acoustic phonon mode in the Raman spectrum. It implies that bending, torsion and structural defects coexist in the longer nanowires. To separate these effects, a grazing‐incidence XRD technique was conceived which allows the nanowire to be scanned along its entire length. Both ω and ϕ rocking curves were recorded, and their shapes were used to extract the bending and torsion profiles, respectively, along the nanowire length. Dips were found in both profiles of longer nanowires, while they are absent from shorter ones, and these dips correspond to the regions where both bending and torsion relax. The energy dissipated in the nanowires, which tracks the bending and torsion profiles, has been used to estimate the emergent dislocation density in nanowire arrays. 相似文献

Bovine serum albumin (BSA)–squaraine supramolecular adducts with aggregation‐induced emission (AIE) properties were prepared and characterized by spectroscopic methods. While squaraine dyes showed a very low fluorescence quantum yield in water, a great enhancement in the fluorescence of the aggregated BSA adducts was achieved due to the abnormal aggregation‐induced emission properties of squaraines. The adducts formation was studied from a kinetic point of view showing unexpected structure–properties relationships. 相似文献
