1H- and 13C-NMR. Spectroscopy of the Nonadrides The 1H- and 13C-NMR. spectra of the nonadrides glaucanic acid ( 1 ), glauconic acid ( 2 ), heveadride ( 3 ), byssochlamic acid ( 4 ), scytalidin ( 5 ), rubratoxin A ( 6 ), and rubratoxin B ( 7 ) have been measured. Partial decoupling experiments permitted assignment of the majority of the signals.  相似文献

As part of our continuing studies of polymer-supported pericyclic reactions for preparing biologically interesting heterocyclic compounds, we have introduced a traceless solid-phase synthesis of hexahydrocinnolines. We developed a method in which mild reaction conditions can be used for the hetero-Diels-Alder reactions on a polymeric support. The dienoic 3-vinyl-2-cyclohexenol attached to a Wang resin through an ether linkage undergoes [4 + 2] cycloaddition reaction with several azadienophiles. The highly stereoselective Diels-Alder reaction showed preferential formation of a single cycloadduct resulting from an anti attack of the dienophile on the polymer-bound diene. Trifluoroacetic acid-mediated cleavage of the polymer-bound cycloadducts yields fused nonaromatic hexahydrocinnolines in moderate yields in three steps.  相似文献

In order to check the hypothesis that rubratoxin B ( 2 ), a C26-metabolite, is formed biogenetically by head-to-tail coupling of two identical C13-precursors derived from decanoic acid and oxaloacetic acid, two labelled forms of the postulated C13-intermediate 2-((E)-1'-octenyl)-3-[14C]methyl- and 2-((E)-1'-octenyl)-3-[13C]-methylmaleic anhydride ( 10 ), were synthesized. The labelled compounds 10 as well as a number of other 14C]- and [13C]-labelled potential precursors were administered to growing cultures of Penicillium rubrum STOLL . Significant incorporation rates of acetate (as intact units) and malonate were observed. Propionate was incorporated after decarboxylation. Succinate exhibited the highest rate of incorporation. The results are in agreement with the assumption that the C10-chain is formed by the fatty acid pathway and the C3-unit via the tricarboxylic acid cycle. After administration of 10 randomization of the label was observed. Thus the question whether compound 10 is a biogenetic intermediate remains unanswered.  相似文献

The Sonogashira coupling reaction of polymer‐supported propargylamine (=prop‐2‐yn‐1‐amine) with aryl iodides in the presence of the [Pd(PPh3)2]Cl2/CuI catalyst system produces 3‐arylprop‐1‐yn‐1‐amines smoothly at room temperature (Scheme). When propargylamine is attached on Wang resin through a carbamate linker, the common problems with amino functionality in palladium‐catalyzed couplings are overcome. The arylpropynamines are cleaved from the polymer with CF3COOH and converted into chromatographically easily separable acetamides. Our solid‐phase method opens a new pathway toward precursors of pharmacologically interesting arylpropynamines and arylpropanamines.  相似文献

Neurosteroid glucuronides were found for the first time in brain samples. The intact glucuronides were extracted from the cortex, hippocampus, hypothalamus, and mid-brain tissues of nicotine- and water-treated mice, and detected with capillary liquid chromatography-electrospray-tandem mass spectrometry (CapLC-ESI-MS/MS). The glucuronides of estradiol, cortisol, corticosterone, tetrahydrodeoxycorticosterone, pregnenolone, and isopregnanolone were identified by comparing retention times in selected reaction monitoring (SRM) chromatograms and the relative abundances of two SRM transitions of each neurosteroid glucuronide between the reference and authentic samples, thus providing reliable identification. In vitro experiments, carried out by using S9 fractions from mouse and rat brains, showed a formation of glucuronides with selected test compounds (corticosterone, pregnenolone, and dehydroepiandrosterone), suggesting that biosynthesis of neurosteroid glucuronides is possible in rodent brain.  相似文献

An ultra performance liquid chromatography–electrospray ionization-tandem mass spectrometry (UPLC–MS/MS) method was developed for the analysis of steroids and their glucuronides in urine samples. The method provides high sensitivity and fast analysis, as both steroids and their glucuronides can be analyzed directly without hydrolysis or complex sample preparation. The method was applied in profiling of targeted and nontargeted steroids and steroid glucuronides during pregnancy. The concentrations of 11 of 27 targeted steroids and steroid glucuronides and the concentrations of 25 nontargeted steroid glucuronides increased about 10–400 fold during the pregnancy. The concentrations of most of these 36 compounds began to increase in the first days of the pregnancy, increased gradually during the pregnancy, achieved a maximum in late pregnancy, and decreased sharply after delivery. Exceptionally, the concentrations of allopregnanolone and 17-hydroxypregnenolone started to increase later than those of the other steroids. Moreover, the concentrations of E2 glucuronides began to decrease one week before the delivery, in contrast to most of the steroids and steroid glucuronides, whose concentrations dropped sharply during the delivery. Concentrations of 34 compounds decreased noticeably when the subject was on sick leave owing a series of painful contractions. The results suggest that steroids and especially steroid glucuronides may provide a valuable diagnostic tool to follow the course of pregnancy.  相似文献

A miniaturized nebulizer chip for capillary liquid chromatography-atmospheric pressure chemical ionization-mass spectrometry (capillary LC-microchip APCI-MS) is presented. The APCI chip consists of two wafers, a silicon wafer and a Pyrex glass wafer. The silicon wafer has a DRIE etched through-wafer nebulizer gas inlet, an edge capillary insertion channel, a stopper, a vaporizer channel and a nozzle. The platinum heater electrode and pads for electrical connection were patterned on to the Pyrex glass wafer. The two wafers were joined by anodic bonding, creating a microchip version of an APCI-source. The sample inlet capillary from an LC column is directly connected to the vaporizer channel of the APCI chip. The etched nozzle in the microchip forms a narrow sample plume, which is ionized by an external corona needle, and the formed ions are analyzed by a mass spectrometer. The nebulizer chip enables for the first time the use of low flow rate separation techniques with APCI-MS. The performance of capillary LC-microchip APCI-MS was tested with selected neurosteroids. The capillary LC-microchip APCI-MS provides quantitative repeatability and good linearity. The limits of detection (LOD) with a signal-to-noise ratio (S/N) of 3 in MS/MS mode for the selected neurosteroids were 20-1000 fmol (10-500 nmol l(-1)). LODs (S/N = 3) with commercial macro APCI with the same compounds using the same MS were about 10 times higher. Fast heat transfer allows the use of the optimized temperature for each compound during an LC run. The microchip APCI-source provides a convenient and easy method to combine capillary LC to any API-MS equipped with an APCI source. The advantages and potentials of the microchip APCI also make it a very attractive interface in microfluidic APCI-MS.  相似文献

A two‐dimensional sparse‐data tomographic problem is studied. The target is assumed to be a homogeneous object bounded by a smooth curve. A nonuniform rational basis splines (NURBS) curve is used as a computational representation of the boundary. This approach conveniently provides the result in a format readily compatible with computer‐aided design software. However, the linear tomography task becomes a nonlinear inverse problem because of the NURBS‐based parameterization. Therefore, Bayesian inversion with Markov chain Monte Carlo sampling is used for calculating an estimate of the NURBS control points. The reconstruction method is tested with both simulated data and measured X‐ray projection data. The proposed method recovers the shape and the attenuation coefficient significantly better than the baseline algorithm (optimally thresholded total variation regularization), but at the cost of heavier computation.  相似文献

An algorithm to simultaneously increase the spatial and statistical accuracy of X‐ray Raman scattering (XRS) based tomographic images is presented. Tomography that utilizes XRS spectroscopy signals as a contrast for the images is a new and promising tool for investigating local atomic structure and chemistry in heterogeneous samples. The algorithm enables the spatial resolution to be increased based on a deconvolution of the optical response function of the spectrometer and, most importantly, it allows for the combination of data collected from multiple analyzers and thus enhances the statistical accuracy of the measured images.  相似文献

The 13C NMR spectra of nine dichlorinated bicyclo[2.2.1]heptanes (norbornanes) have been measured and assigned. The pairwise effects of chlorine substituents which cause deviations from the additivity of single-substituent effects were investigated and are discussed. The largest effect found is the high-field shift of carbons bearing vicinal cis substituents. In the case of geminal substitution deviations from additivity were found to be to low field and large in the γ, smaller in the β and negligible in the α chemical shifts. The observed deviations for 1,3-disubstituted cases vary from ?3.2 to +1.1 ppm at different carbons, allowing no simple explanation. Replacement of α-hydrogen in a diaxial 1,3-arrangement by CH3, OH or CI causes the single substituent effect, namely the γa effect, to change considerably.  相似文献
