Enzymes that degrade pectin are called pectinases. Pectinases of microbial origin are used in juice clarification as the process is cost-effective. This study screened a pectinase-producing bacterium isolated from soil and identified as Bacillus subtilis 15A B-92 based on the 16S rRNA molecular technique. The purified pectinase from the isolate showed 99.6 U/mg specific activity and 11.6-fold purity. The molecular weight of the purified bacterial pectinase was 14.41 ± 1 kD. Optimum pectinase activity was found at pH 4.5 and 50 °C, and the enzyme was 100% stable for 3.5 h in these conditions. No enzymatic inhibition or activation effect was seen with Fe2+, Ca2+, or Mg2+. However, a slight inhibition was seen with Cu2+, Mn2+, and Zn2+. Tween 20 and 80 slightly inhibited the pectinase, whereas iodoacetic acid (IAA), ethylenediaminetetraacetate (EDTA), urea, and sodium dodecyl sulfate (SDS) showed potent inhibition. The bacterial pectinase degraded citrus pectin (100%); however, it was inactive in the presence of galactose. With citrus pectin as the substrate, the Km and Vmax were calculated as 1.72 mg/mL and 1609 U/g, respectively. The high affinity of pectinase for its substrate makes the process cost-effective when utilized in food industries. Nanocrystalline thin films of copper selenide have been grown on glass and tin doped-indium oxide substrates using chemical method. At ambient temperature, golden films have been synthesized and annealed at 200 °C for 1 h and were examined for their structural, surface morphological and optical properties by means of X-ray diffraction (XRD), scanning electron microscopy and UV-vis spectrophotometry techniques, respectively. Cu2−xSe phase was confirmed by XRD pattern and spherical grains of 30 ± 4 - 40 ± 4 nm in size aggregated over about 130 ± 10 nm islands were seen by SEM images. Effect of annealing on crystallinity improvement, band edge shift and photoelectrochemical performance (under 80 mW/cm2 light intensity and in lithium iodide electrolyte) has been studied and reported. Observed p-type electrical conductivity in copper selenide thin films make it a suitable candidate for heterojunction solar cells. 相似文献

An infinitely extending homogenous partially ionized plasma endowed with several physical mechanisms and permeated by a variable magnetic field is considered. The combined effect of these parameters, namely, Hall currents, finite conductivity, ion viscosity, collision with neutrals and thermal conductivity on the gravitational instability of the plasma is studied. It is found that the several mechanisms play different physical roles in the perturbed problem. Jeans' Criterion is analyzed in the framework of Tsallis' statistics for possible modifications due to the presence of nonextensive effects. The powder diffractometer and Hi-Q diffractometer at Dhruva reactor make use of five identical 1-D position sensitive detectors (PSDs) to scan scattering angles in the range 3° to 140°. In order to improve the overall throughput of these spectrometers, it is planned to install a bank of 15 high-efficiency and high-resolution PSDs arranged in three layers with five PSDs in each layer. With each high pressure PSD (3He 10 bar + Kr 2 bar) showing the efficiency gain of 1.8 at 1.2 Å, detector bank is expected to show overall gain of 5.5 times the present detection efficiency and reduction in data collection time by equivalent factor. The 1-D PSDs are developed in batches of five, and are characterized so that all PSDs operate at uniform parameters such as position resolution, uniformity of efficiency and linearity of response. Position spectrum indicates the differential position resolution to be ~1 mm and integral position resolution to be 3–4 mm. Broadening of position spectrum at the extreme end of sensitive length of PSD is analysed using fine shift of the beam. Dependence of position resolution and dynamic range of output pulse on the input impedance of pre-amplifier is also presented. 相似文献

We develop a nonlinear two-dimensional fluid model of whistler turbulence that includes effect of electron fluid density perturbations. The latter is coupled nonlinearly with wave magnetic field. This coupling leads essentially to finite compressibility effects in whistler turbulence model. We find from our simulations that despite strong compressibility effects, the density fluctuations follow the evolution of the wave magnetic field fluctuations. In a characteristic regime where large scale whistlers are predominant, the coupled density fluctuations are found to follow a Kolmogorov-like phenomenology in the inertial range turbulence. Consequently, the turbulent energy is dominated by the large scale (compared to electron inertial length) eddies and it follows a Kolmogorov-like k−7/3 spectrum, where k is a characteristic wavenumber. 相似文献

the need of other divisions in BARC, Department of Atomic Energy units and some universities and research institutes in India
and abroad for a variety of applications. The NR facility set up by SSPD at Apsara reactor has been used for a variety of
applications in nuclear, aerospace, defense and metallurgical industries. The work done in the development of neutron detectors
and neutron radiography is reported in this article.

In the present work emission and absorption spectroscopy have been used to determine the plasma parameters of neon in a hollow cathode discharge lamp. The excitation temperature is determined using the intensity ratio method and Boltzmann's plot method whereas the electron density is determined from the Stark broadening of the spectral lines. The behavior of the optogalvanic signal as a function of laser energy has been studied for three transitions from the 2p53s [1/2]2 metastable state following ΔJ=ΔK=0, ±1 dipole selection rules.
