Validation of a method for predicting the precision, limit of detection and range of quantitation in competitive ELISA.

The mathematical model for predicting the precision, limit of detection (LOD) and range of quantitation (ROQ) in a competitive enzyme-linked immunosorbent assay (ELISA) proposed by Hayashi et al. (Anal. Chem., 2004, 76, 1295) was validated. The model describes the relative standard deviation (RSD) of concentration estimates by the RSDs of pipetting volumes of analyte, enzyme-conjugated antigen, antibody and substrate solutions, and the standard deviation (SD) of inherent absorbances between the wells in an ELISA plate. For 6 kinds of direct competitive ELISA kits, the LOD and ROQ predicted by the model agreed well with those obtained by experiments with real samples. It was also confirmed that the model is applicable to the prediction of uncertainty that depends on the pipetting error of the viscous antiserum solution. The model was demonstrated to be useful for estimating the LOD and ROQ of competitive ELISA.     

Electrode reaction of zinc ions at a dropping mercury electrode in ammonium sulphate solutions

The electrode reaction of Zn(II) at a DME in aqueous solutions of (NH₄)₂SO₄ has been studied by d.c. and square-wave polarography at 25.0±0.1°C. The electrochemical kinetic parameters for the reaction are determined. The appearance of the second wave in the square-wave polarogram has been attributed to the low values of the rate parameter and the transfer coefficient.     

Voltammetry at partially covered electrodes: Part I. Chronopotentiometry and chronoamperometry at model electrodes

Equations for chronopotentiometry and chronoamperometry at partially covered electrodes have been derived using a model of hexagonal array of cylidrical spaces terminated, at the electrode surface, by concentric active and inactive regions. The boundary value problem was shown to be analogous to that for a charge transfer preceded by a chemical reaction. Experiments with the reduction of ferricyanide on gold model electrodes partially covered with photoresist layer showed excellent agreement with the theory. Application of the equations to estimation of coverage and size of active sites distributed on a electrode surface is discussed.     

Mutagenesis approaches to deduce structure-function relationships in terpene synthases

This review highlights mutagenesis studies of terpene synthases, specifically sesquiterpene synthases and oxidosqualene cyclases. Mutagenesis studies of these enzymes have provided mechanistic insights, structure-function relationships for specific enzymatic residues, novel terpene structures and enzymes with novel activities. The literature through 2002 is reviewed and 113 references cited.

Synthesis of polyamides by the polyaddition of bisimidazoline with dicarboxylic acids

N,N′-Dipropionylethylenediamine was synthesized by the ring-opening addition reaction of 2-ethyl-2-imidazoline with propionic acid at 220°C. By applying this reaction to polymerization, polyamides were synthesized by the ring-opening polyaddition reaction at 220°C. of 1,4-bis(imidazoline-2-yl)butane with adipic acid, succinic acid, sebacic acid, and terephthalic acid. The reaction product of 1,4-bis(imidazoline-2-yl)butane with adipic acid, which was proposed to be nylon 26, was compared with an authentic sample of nylon 26 and shown to possess a very similar infrared spectrum and melting point.
