Cardamonin is a polyphenolic natural product that has been shown to possess cytotoxic activity against a variety of cancer cell lines. We previously reported the semi-synthesis of a novel Cu (II)–cardamonin complex (19) that demonstrated potent antitumour activity. In this study, we further investigated the bioactivity of 19 against MDA-MB-468 and PANC-1 cancer cells in an attempt to discover an effective treatment for triple-negative breast cancer (TNBC) and pancreatic cancer, respectively. Results revealed that 19 abolished the formation of MDA-MB-468 and PANC-1 colonies, exerted growth-inhibitory activity, and inhibited cancer cell migration. Further mechanistic studies showed that 19 induced DNA damage resulting in gap 2 (G2)/mitosis (M) phase arrest and microtubule network disruption. Moreover, 19 generated reactive oxygen species (ROS) that may contribute to induction of apoptosis, corroborated by activation of caspase-3/7, PARP cleavage, and downregulation of Mcl-1. Complex 19 also decreased the expression levels of p-Akt and p-4EBP1, which indicates that the compound exerts its activity, at least in part, via inhibition of Akt signalling. Furthermore, 19 decreased the expression of c-Myc in PANC-1 cells only, which suggests that it may exert its bioactivity via multiple mechanisms of action. These results demonstrate the potential of 19 as a therapeutic agent for TNBC and pancreatic cancer. 相似文献

Encapsulation of nickel oxide (NiO) particles is of great interest to the researchers as such modification produces remarkable improvement in properties and versatility in application potential. In this investigation, nanosized NiO particles were first prepared by calcination of nickel hydroxide precursor obtained using a simple liquid‐phase process. The produced NiO particles were stabilized with oleic acid and then treated with tetraethylorthosilicate to produce NiO/SiO2 composite seed particles. Finally tri‐layered inorganic/organic composite particles were prepared by seeded copolymerization of styrene and 2‐hydroxyethyl methacrylate (HEMA) in the presence of NiO/SiO2 composite seed particles. The produced composite particles named as NiO/SiO2/P(S‐HEMA) were colloidally stable, and the obtained particles were characterized by Fourier transform infrared spectroscopy, scanning electron microscopy, X‐ray diffraction, X‐ray photoelectron spectroscopy and thermogravimetric analyses. To describe the flows of fluids over a wide range of pressures, it is necessary to take into account the fact that the viscosity
of the fluid depends on the pressure. That the viscosity depends on the pressure has been verified by numerous careful experiments.
While the existence of solutions local-in-time to the equations governing the flows of such fluids are available for small,
special data and rather unrealistic dependence of the viscosity on the pressure, no global existence results are in place.
Our interest here is to establish the existence of weak solutions for spatially periodic three-dimensional flows that are
global in time, for a large class of physically meaningful viscosity-pressure relationships.

The use of infrared laser-assisted fluorination to release oxygen from milligram quantities of silicates or other oxide mineral grains is a well-established technique. However, relatively few studies have reported the optimisation of this procedure for oxygen-17 isotope measurements. We describe here details of an analytical system using infrared (10 μm) laser-assisted fluorination, in conjunction with a dual inlet mass spectrometer of high resolving power ( approximately 250) to provide (17)O and (18)O oxygen isotope measurements from 0.5-2 mg of silicates or other oxide mineral grains. Respective precisions (1) of typically 0.08 and 0.04 per thousand are obtained for the complete analytical procedure. Departures from the mass-dependent oxygen isotope fractionation line are quantified by Delta(17)O; our precision (1) of such measurements on individual samples is shown to be +/-0.024 per thousand. In turn, this permits the offset between parallel, mass-dependent fractionation lines to be characterised to substantially greater precision than has been possible hitherto. Application of this system to investigate the (17)O versus (18)O relationship for numerous terrestrial whole-rock and mineral samples, of diverse geological origins and age, indicates that the complete data set may be described by a single, mass-dependent fractionation line of slope 0.5244+/- 0.00038 (standard error). A waveguide in integrated optics is defined by its refractiveindex. The guide is assumed to be invariant in the propagationdirection while in the transverse direction it is supposed tobe a compact perturbation of an unbounded stratified medium.We are interested in the modes guided by this device, whichare waves with a transverse energy confined in a neighbourhoodof the perturbation. Our goal is to analyse the existence of such guided modes. Underthe assumptions of weak guidance the problem reduces to a two-dimensionaleigenvalue problem for a scalar field. The associated operatoris unbounded, selfadjoint, and bounded from below. Its spectrumconsists of the discrete spectrum corresponding to the guidedmodes and of the essential spectrum corresponding to the radiationmodes. We present existence results of guided modes and an asymptoticstudy at high frequencies, which shows that contrarily to thecase of optical fibers, the number of guided modes can remainbounded. The major tools are the min-max principle and comparisonof results between different eigenvalue problems. The originalityof the present study lies in the stratified character of theunbounded reference medium. 相似文献

Background
Opioid agonist drugs produce analgesia. However, long-term exposure to opioid agonists may lead to opioid dependence. The analgesic and addictive properties of opioid agonist drugs are mediated primarily via the mu-opioid receptor (MOR). Opioid agonists appear to alter neuronal morphology in key brain regions implicated in the development of opioid dependence. However, the precise role of the MOR in the development of these neuronal alterations remains elusive. We hypothesize that identifying and characterizing novel MOR interacting proteins (MORIPs) may help to elucidate the underlying mechanisms involved in the development of opioid dependence. 相似文献7.

A direct spectrophotometric method for the determination of aluminium with morin has been developed. Morin reacts in slightly acidic 50% ethanolic media (0.0001-0.0015 M H(2)SO(4)) with Al to give a deep-yellow chelate which has an absorption maximum at 421 nm. The average molar absorptivity and Sandell's sensitivity were found to be 5.3 x 10(3) l mol(-1) cm(-1) and 5 ng of Al cm(-2), respectively. The reaction is instantaneous and absorbance remains stable for 48 h. The colour system obeys Beer's law from 10 ng ml(-1) to 5.0 mug ml(-1) of Al; the stoichiometric composition of the chelate is 2:3 (Al:morin). The interference from over 50 cations, anions and complexing agents has been studied at 0.1 mug ml(-1) of Al. The method was applied successfully to some certified reference material samples (alloys and steels), environmental waters (inland and surface), biological samples (human blood, urine and gallstone), soils and complex synthetic mixtures. 相似文献

Electron donor acceptor complexes (EDA) of the 1,4,7,10,13,16-hexaoxacyclooctadecane (18-crown-6) as a rich donor were spectrophotometrically discussed and synthesized in solid form according the interactions with different nine of usual π-acceptors like 2,3,5,6-tetrachlorocyclohexa-2,5-diene-1,4-dione (p-chloranil; p-CHL), tetrachloro-1,2-benzoquinone (o-chloranil; o-CHL), 2,3-dichloro-5,6-dicyano-1,4-benzoquinone (DDQ), tetracyanoquinodimethane (TCNQ), 2,6-dichloroquinone-4-chloroimide (DCQ), 2,6-dibromoquinone-4-chloroimide (DBQ), 2,5-dichloro-3,6-dihydroxy-1,4-benzoquinone (chloranilic acid; CLA), N-bromosuccinimide (NBS), 2,4,6-trinitrophenol (picric acid; PA). Spectroscopic and physical data such as formation constant (K(CT)), molar extinction coefficient (?(CT)), standard free energy (ΔG°), oscillator strength (f), transition dipole moment (μ), resonance energy (R(N)) and ionization potential (I(p)) were estimated in chloroform or methanol at 25°C. Based on the elemental analysis and photometric titrations the CT-complexes were formed indicated the formation of 1:1 charge-transfer complexes for the o-CHL, TCNQ, DCQ, DBQ and NBS acceptors but 1:3 ratio for p-CHL, DDQ, CLA and PA, respectively. The charge-transfer interactions were interpretative according to the formation of dative ion pairs [18C6(?+), A(?-)], where A is acceptor. All of the resulting charge transfer complexes were isolated in amorphous form and the complexes formations on IR and (1)H NMR spectra were discussed. 相似文献
