Participation d'un groupe amide a la formation de sels de dioxolanne-1,3 ylium-2; Modele biomimetique de reaction de peptidation

An amide group is shown to be capable of intramolecular participation in the formation of an 1,3-dioxolan 2-ylium cation giving a tricyclic organic cation of a new type $\text{5a}$,$\text{b}$. This cation may be considered as a model of an electrophilic intermediate which might be formed from a peptidyl t-RNA during protein biosynthesis.The tricyclic salt $\text{5a}$,$\text{b}$ reacts with water and methanol like a classical dioxolenium salt. An amide acetal $\text{18}$ can be obtained from dimethylamine and 5a; hydrolysis of $\text{18}$ does not lead to the corresponding amide $\text{20}$ but to the cleavage of the CN. bond.     

Single nucleotide polymorphism analysis by allele-specific primer extension with real-time bioluminescence detection in a microfluidic device

A microfluidic approach for rapid bioluminescent real-time detection of single nucleotide polymorphism (SNP) is presented. The method is based on single-step primer extension using pyrosequencing chemistry to monitor nucleotide incorporations in real-time. The method takes advantage of the fact that the reaction kinetics differ between matched and mismatched primer-template configurations. We show here that monitoring the initial reaction in real time accurately scores SNPs by comparing the initial reaction kinetics between matched and mismatched configurations. Thus, no additional treatment is required to improve the sequence specificity of the extension, which has been the case for many allele-specific extension assays. The microfluidic approach was evaluated using four SNPs. Three of the SNPs included primer-template configurations that have been previously reported to be difficult to resolve by allele-specific primer extension. All SNPs investigated were successfully scored. Using the microfluidic device, the volume for the bioluminescent assay was reduced dramatically, thus offering a cost-effective and fast SNP analysis method.     

Discriminant of a Germ {Phi}: (C2, 0)->(C2, 0) and Seifert Fibred Manifolds

Let f and g be two analytic function germs without common branches.We define the Jacobian quotients of (g, f), which are ‘firstorder invariants’ of the discriminant curve of (g, f),and we prove that they only depend on the topological type of(g, f). We compute them with the help of the topology of (g,f). If g is a linear form transverse to f, the Jacobian quotientsare exactly the polar quotients of f and we affirm the resultsof D. T. Lê, F. Michel and C. Weber.     

Isomerization of a peptidic fragment studied theoretically in vacuum and in explicit water solvent at finite temperature

The cis-trans isomerization of N-methylacetamide, a molecular model of the polypeptide chain, is examined via umbrella sampling Car-Parrinello MD and classical MD, in both gas and solution phases at 300 K. A new analysis of the C(O)-N bond interconversion and a full examination of the solvent shell structure are presented.
