(R)-[1-(Dimethylamino)ethyl]benzene reacts with nBuLi in a 1:1 molar ratio in pentane to quantitatively yield a unique hetero-aggregate (2 a) containing the lithiated arene, unreacted nBuLi, and the complexed parent arene in a 1:1:1 ratio. As a model compound, [Li(4)(C(6)H(4)CH(Me)NMe(2)-2)(2)(nBu)(2)] (2 b) was prepared from the quantitative redistribution reaction of the parent lithiated arene Li(C(6)H(4)CH(Me)NMe(2)-2) with nBuLi in a 1:1 molar ratio. The mono-Et(2)O adduct [Li(4)(C(6)H(4)CH(Me)NMe(2)-2)(2)(nBu)(2)(OEt(2))] (2 c) and the bis-Et(2)O adduct [Li(4)(C(6)H(4)CH(Me)NMe(2)-2)(2)(nBu)(2)(OEt(2))(2)] (2 d) were obtained by re-crystallization of 2 b from pentane/Et(2)O and pure Et(2)O, respectively. The single-crystal X-ray structure determinations of 2 b-d show that the overall structural motifs of all three derivatives are closely related. They are all tetranuclear Li aggregates in which the four Li atoms are arranged in an almost regular tetrahedron. These structures can be described as consisting of two linked dimeric units: one Li(2)Ar(2) dimer and a hypothetical Li(2)nBu(2) dimer. The stereochemical aspects of the chiral Li(2)Ar(2) fragment are discussed. The structures as observed in the solid state are apparently retained in solution as revealed by a combination of cryoscopy and (1)H, (13)C, and (6)Li NMR spectroscopy. 相似文献

This article describes the development and full characterization of a microfluidic chip for electrofusion of human peripheral blood B-cells and mouse myeloma (NS-1) cells to generate hybridomas. The chip consists of an array of 783 traps, with dimensions that were optimized to obtain a final cell pairing efficiency of 33±6%. B cells were stained with a cytoplasmic stain CFDA to assess the different stages of cell fusion, i.e. dye transfer to NS-1 cells (initiating fusion) and membrane reorganization (advanced fusion). Six DC pulses of 100 μs (2.5 kV/cm) combined with an AC field (30 s, 2 MHz, 500 V/cm) and pronase treatment resulted in the highest electrofusion efficiency of paired cells (51±11%). Hybridoma formation, with a yield of 0.33 and 1.2%, was observed after culturing the fused cells for 14 days in conditioned medium. This work provides valuable leads to improve the current electrofusion protocols for the production of human antibodies for diagnostic and therapeutic applications. 相似文献

The synthesis and mesomorphic properties of 30 bent-core compounds with a fluorine substituent in one of the outer rings are reported. The banana-shaped compounds are all derived from resorcinol and contain esters as linking groups between the five aromatic rings. The different mesophases have been characterized by polarizing optical microscopy, differential scanning calorimetry, X-ray diffraction studies and electro-optical investigations. The compounds with the longer terminal chains exhibit an antiferroelectric SmCP phase. Upon introduction of a fluorine substituent the layer spacing increases, as compared with the corresponding unsubstituted compound. The introduction of one terminal vinyl group in the mono-substituted bent-core mesogens has no significant influence on the liquid crystalline properties and the switching remains antiferroelectric. Due to the introduction of the terminal double bond, these banana-shaped compounds are suitable for the preparation of siloxane polymers or attachment to a hydrogen-terminated silicon surface. 相似文献

A new three‐component reductive arylation of amides with stable reactants (iPrOH and arylboronate esters), making use of a 2‐pyridinyl (Py) directing group, is described. The N‐Py‐amide substrates are readily prepared from carboxylic acids and PyNH2, and the resulting N‐Py‐1‐arylalkanamine reaction products are easily transformed into the corresponding chlorides by substitution of the HN‐Py group with HCl. The 1‐aryl‐1‐chloroalkane products allow substitution and cross‐coupling reactions. Therefore, a general protocol for the transformation of carboxylic acids into a variety of functionalities is obtained. The Py‐NH2 by‐product can be recycled. 相似文献

This study presents the development of microreactor protocols for the successful continuous flow end group modification of atom transfer radical polymerization precursor polymers into azide end‐capped materials and the subsequent copper‐catalyzed azide alkyne click reactions with alkyne polymers, in flow. By using a microreactor, the reaction speed of the azidation of poly(butyl acrylate), poly(methyl acrylate), and polystyrene can be accelerated from hours to seconds and full end group conversion is obtained. Subsequently, copper‐catalyzed click reactions are executed in a flow reactor at 80 °C. Good coupling efficiencies are observed and various block copolymer combinations are prepared. Furthermore, the flow reaction can be carried out in only 40 min, while a batch procedure takes several hours to reach completion. The results indicate that the use of a continuous flow reactor for end group modifications as well as click reactions has clear benefits towards the development and improvement of well‐defined polymer materials. © 2014 Wiley Periodicals, Inc. Reaction of aryllithium reagents LiR (R = C(6)H(4)((R)-CH(Me)NMe(2))-2 (1a), C(6)H(3)(CH(2)NMe(2))(2)-2,6 (1b), C(6)H(4)(CH(2)N(Me)CH(2)CH(2)OMe)-2 (1c)) with 1 equiv of sulfur (1/8 S(8)) results in the quantitative formation of the corresponding lithium arenethiolates [Li{SC(6)H(4)((R)-CH(Me)NMe(2))-2}](6) (3), [Li{SC(6)H(3)(CH(2)NMe(2))(2)-2,6}](6) (4), and [Li{SC(6)H(4)(CH(2)N(Me)CH(2)CH(2)OMe)-2}](2) (5). Alternatively, 3 can be prepared by reacting the corresponding arenethiol HSC(6)H(4)((R)-CH(Me)NMe(2))-2 (2) with (n)BuLi. X-ray crystal structures of lithium arenethiolates 3 and 4, reported in abbreviated form, show them to have hexanuclear prismatic and hexanuclear planar structures, respectively, that are unprecedented in lithium thiolate chemistry. The lithium arenethiolate [Li{SC(6)H(4)(CH(2)N(Me)CH(2)CH(2)OMe)-2}](2) (5) is dimeric in the solid state and in solution, and crystals of 5 are monoclinic, space group P2(1)/c, with a = 17.7963(9) ?, b = 8.1281(7) ?, c = 17.1340(10) ?, beta = 108.288(5) degrees, Z = 4, and final R = 0.047 for 4051 reflections with F > 4sigma(F). Hexameric 4 reacts with 1 equiv of lithium iodide and 2 equiv of tetrahydrofuran to form the dinuclear adduct [Li(2)(SAr)(I)(THF)(2)] (6). Crystals of 6 are monoclinic, space group P2(1)/c, with a = 13.0346(10) ?, b = 11.523(3) ?, c = 16.127(3) ?, beta = 94.682(10) degrees, Z = 4, and final R = 0.059 for 3190 reflections with F > 4sigma(F). 相似文献
