Prenylated xanthones, isolated from the fruit hulls and the edible arils and seeds of Garcinia mangostana, were tested for their antituberculosis potential. Alpha- and beta-mangostins and garcinone B exhibited strong inhibitory effect against Mycobacterium tuberculosis with the minimum inhibitory concentration (MIC) value of 6.25 microg/ml. Tri- and tetra-oxygenated xanthones with di-C5 units or with a C5 and a modified C5 groups are essential for high activities. Substitution in the A and C rings has been shown to modify the bioactivity of the compounds.  相似文献

Two new xanthones, designated garcimangosxanthone F (1) and garcimangosxanthone G (2), were isolated from the EtOAc-soluble fraction of ethanolic extract from the pericarp of Garcinia mangostana. Their structures were established as 1,6,7-trihydroxy-5-(3-methylbut-2-enyl)-8-(3-hydroxy-3-methylbutyl)-6′,6′-dimethylpyrano[2′,3′:3,2]xanthone and 1,6,7-trihydroxy-5-(3-methylbut-2-enyl)-8-(3-hydroxy-3-methylbutyl)-6′,6′-dimethyl-4′,5′-dihydropyrano[2′,3′:3,2]xanthone, respectively, on the basis of their 1D, 2D NMR and MS data interpretation.  相似文献

Two new xanthones, 3-hydroxy-6-methoxy-5'-isopropyl-4',5'-dihydrofuro[2',3'?:?7, 8]-6″,6″-dimethyl-4″,5″-dihydropyrano[2″,3″?:?1,2]xanthone (1) and 1,6-dihydroxy-7-methoxy-8-(3-methylbut-3-enyl)-6',6'-dimethyl-4',5'-dihydropyrano[2'3'?:?3,2]xanthone (2), were isolated from the pericarp of Garcinia mangostana. A new prenylated xanthone, 5-O-methylcelebixanthone (1), together with six known compounds; celebixanthone (2), 1,3,7-trihydroxy-2,4-di(3-methylbut-2-enyl)xanthone (3), cochinchinone A (4), alpha-mangostin (5), beta-mangostin (6) and cochinchinone C (7) were isolated from roots of Cratoxylum cochinchinense. Their structures were elucidated by spectroscopic methods. Compounds 2 and 4-7 showed cytotoxic activity against the human lung cancer cell line (NCI-H187) with IC(50) values ranging from 0.65 to 5.2 microg/ml. Compounds 1, 2, 6 and 7 also showed antimalarial activity against Plasmodium falciparum with IC(50) values of 3.2, 4.9, 7.2 and 2.6 microg/ml, respectively.  相似文献

Thirteen xanthones (1-13) were isolated from the resin of Garcinia hanburyi. Among them, two new compounds (namely gaudichaudic acid, and isogambogenic acid, 1, 2), and one new natural product (deoxygaudichaudione A, 3) were identified on the basis of extensive spectral evidence including detailed 2D NMR data. Ten of these xanthones were tested for their cytotoxicities against human leukemia K562 (K562/S) and doxorubicin-resistant K562 (K562/R) cell lines, and showed similar inhibitory effects on both cell lines, suggesting that this group of polyprenylated xanthones might not be multidrug resistance (MDR) substrates.  相似文献

From the CHCl3-soluble fraction of the fruits of Garcinia mangostana (Clusiaceae), six xanthone derivatives, alpha-mangostin (1), gamma-mangostin (2), gartanin (3), deoxygartanin (4), 1-isomangstanin (5) and garcinone E (6), were isolated. All these compounds significantly inhibited HSC-T6 viability as assessed by employing HSC-T6 hepatic stellate cells as an in vitro assay system. Among them, compounds 1 and 2, the most potent and major constituents of G. mangostana, inhibited HSC-T6 viability in dose- and time-dependent manners. In addition, compounds 1 and 2 significantly reduced collagen content, a pathological characteristic of liver fibrosis. Taken together, G. mangostana and its constituents might be beneficial for the treatment of liver fibrosis.  相似文献

A new prenylated xanthone, 1,3,5,6-tetrahydroxy-4,7,8-tri(3-methyl-2-butenyl)xanthone (1), was isolated from the wood of Garcinia xanthochymus together with a known xanthone, garciniaxanthone E (2). Their structures were determined by spectroscopic analysis. Compounds 1 (3 microM) and 2 (10 microM) elicited marked enhancement of nerve growth factor-mediated neurite outgrowth in PC12D cells.  相似文献

Two new prenylated xanthones, afzeliixanthones A (1) and B (2), together with three known xanthones (3-5) and two phytosterols, beta-sitosterol and stigmasterol, were isolated from the CH2Cl2/MeOH (1:1) extract of the stem bark of Garcinia afzelii ENGL. collected in the South West Province of Cameroon. Structures were mainly established using one and two-dimensional NMR and mass spectroscopies. Studies on the stem of Garcinia mangostana have led to the isolation of one new xanthone mangosharin (1) (2,6-dihydroxy-8-methoxy-5-(3-methylbut-2-enyl)-xanthone) and six other prenylated xanthones, alpha-mangostin (2), beta-mangostin (3), garcinone D (4), 1,6-dihydroxy-3,7-dimethoxy-2-(3-methylbut-2-enyl)-xanthone (5), mangostanol (6) and 5,9-dihydroxy-8- methoxy-2,2-dimethyl-7-(3-methylbut-2-enyl)-2H,6H-pyrano-[3,2-b]-xanthene-6-one (7). The structures of these compounds were determined by spectroscopic methods such as 1H NMR, 13C NMR, mass spectrometry (MS) and by comparison with previous studies. All the crude extracts when screened for their larvicidal activities indicated very good toxicity against the larvae of Aedes aegypti. This article reports the isolation and identification of the above compounds as well as bioassay data for the crude extracts. These bioassay data have not been reported before.  相似文献

The fruit rinds of mangosteen (Garcinia mangostana Linn.) have long been used as traditional medicines for treatment of skin infections, wounds, and diarrhea. A simple thin-layer chromatography (TLC)-densitometric method has been developed for the simultaneous quantification of alpha-mangostin in the extracts from unripe and ripe fruit rinds of G. mangostana. It was found in the ranges of 10.48 +/- 0.83 and 16.65 +/- 0.38% (w/w) in the dried unripe and ripe fruit rinds, respectively. The method was validated for linearity, precision, accuracy, limit of detection (LOD), and limit of quantitation (LOQ). The linearity was found over the range of 100-500 ng/spot with regression coefficient 0.999. Intraday and interday precision studies showed the relative standard deviation was <2%. Accuracy of the method was determined by a recovery study conducted at 3 different levels, and the average recovery was 99.49%. The LOD and LOQ were 40 and 100 ng, respectively. The proposed TLC-densitometric method was found to be simple, precise, specific, sensitive, and accurate. This method can be used for routine quality control of raw material of G. mangostana fruit rind, extract, and its products. It also can be applied in quantifying this marker compound in other drugs.  相似文献

A new caged-tetraprenylated xanthone, hanburinone (1), was isolated from the fresh fruits of Garcinia hanburyi together with four known caged-tetraprenylated xanthones; isomoreollin B (2), morellin (3), moreollic acid (4) and morellic acid (5). Their structures were elucidated by analysis of spectroscopic data and comparison of the NMR data with those reported previously. Compounds 4 and 5 showed moderately antibacterial activity against methicillin-resistant Staphylococcus aureus with a MIC value of 25 microg/ml.  相似文献

The air-dried fruit hulls of Garcinia mangostana Linn. were extracted with 85% ethanol. Furthermore, a new xanthone, 1,3,6-trihydroxy-2,5-bis(3-methylbut-2-enyl)-6',6'-dimethyl-4',5'-dihydropyrano[2',3':7,8]xanthone, along with five known xanthones related to their antioxidant activity was purified by silica gel column chromatography and then identified using spectroscopic methods (1D and 2D NMR, MS). The antioxidant activities were evaluated using 1,1-diphenyl-2-picrylhydrazyl (DPPH) radical-scavenging capability. An activity-guided isolation and purification process were used to identify the components, showing the strong DPPH radical-scavenging activity of G. mangostana.  相似文献

Inspired by the combination of unique structure and potent bioactivities exhibited by several family members of the caged Garcinia xanthones, we developed a synthesis of simplified analogues that maintain the overall caged motif. The caged structure of these compounds was constructed via a site-selective Claisen/Diels-Alder reaction cascade. We found that the fully substituted caged structure, in which are included the C18 and C23 geminal methyl groups, is necessary to maintain bioactivity. Analogue had comparable activity to the natural products of this family, such as gambogic acid. These compounds exhibit cytotoxicity in a variety of tumor cell lines at low micromolar concentrations and were found to induce apoptosis in HUVE cells. In addition, studies with HL-60 and HL-60/ADR cells indicate that these compounds are not affected by the mechanisms of multidrug resistance, conferred by P glycoprotein expression, typical of relapsed cancers and thus represent a new and potent pharmacophore.  相似文献

Two new prenylated xanthones (=9H‐xanthen‐9‐ones), garcimangosxanthones D ( 1 ) and E ( 2 ), together with the six known xanthones 3 – 8 , were isolated from the pericarp of Garcinia mangostana. Their structures were determined by analysis of their spectroscopic data. All of the isolated compounds were biologically evaluated for their in vitro cytotoxic activity against A549, Hep‐G2, and MCF‐7 human‐cancer cell lines and antioxidant activity. Compound 1 exhibited moderate cytotoxicity against Hep‐G2 (IC50=19.2 μM ) and weak cytotoxicity against MCF‐7 (IC50=62.8 μM ) cell lines, and compound 2 showed moderate cytotoxicity against A549, Hep‐G2, and MCF‐7 cell lines with IC50 values of 12.5–20.0 μM (Table 2). Both compounds 1 and 2 demonstrated a weak antioxidant activity with ferric reducing antioxidant power (FRAP) values of 41±7 and 130±4 μmol/g, respectively (Table 3).  相似文献

A megastigmane sulphoglycoside together with three phenolic compounds were isolated from the water-soluble fraction of the pericarps of Garcinia mangostana. The structure of the new compound was determined as 4-O-sulpho-β-d-glucopyranosyl abscisate (1) by spectroscopic data. Proanthocyanidin A2 (2) showed potent α-glucosidase inhibitory and DPPH scavenging activities with IC50 values of 3.46 and 11.6 μM, respectively.  相似文献
