Nucleophilic attack of azide on 2-bromo-3-fluoro-3-(1-trityl-1H-imidazol-4-yl)-propan-1-ol (1a) in aprotic solvent occurs on the 2-position to give the 2-azido derivative (2a). Reduction of azide and removal of the trityl group produces β-fluorohistidinol (6a). Elimination of HBr from 1a followed by “FBr” addition to the resulting double bond gives 2-bromo-3,3-difluoro-3-(1-trityl-1H-imidazol-4-yl)-propan-1-ol (1b). Nucleophilic attack of azide followed by reduction and removal of the trityl group, as for the preparation of 6a, gives β,β-difluorohistidinol (6b). Initial attempts, under a variety of conditions, to oxidize the fluorinated histidinol precursors to carboxylic acids have not been successful.  相似文献

The synthesis of β-thiolactone and β-lactam analogs of tetrahydrolipstatin is described from a common late-stage β-lactone derivative. These analogs, and a cis-disubstituted β-lactone analog of tetrahydrolipstatin, were screened for activity against porcine pancreatic lipase and for inhibition of cell growth of a panel of four human cancer lines.  相似文献

A short and efficient synthesis for β-acyl and β-alkoxycarbonyl heterocyclic sulfones containing benzothiazol (BT) and phenyltetrazol (PT) heterocyclic core is presented here. The objective of this research was to improve the aqueous solubility, dissolution rate and, consequently, bioavailability of diacerein, along with avoiding its side effect of diarrhea, by complexation with β-cyclodextrin (β-CD) and HP-β-cyclodextrin (HP-β-CD). Phase solubility curve was classified as an AN type for both the CDs, which indicated formation of complex of diacerein with β-CD and HP-β-CD in 1:1 stoichiometry and demonstrating that both CDs are proportionally less effective at higher concentrations. The complexes were prepared by kneading method and were evaluated to study the effect of complexation on aqueous solubility and rate of dissolution in phosphate buffer (pH 6.8). Based on the dissolution profile HP-β-CD was selected for preparing fast disintegrating tablet of diacerein which was compared with marketed formulation (MF-J). The HP-β-CD complex was probed for Fourier transform infrared spectroscopy, differential scanning calorimetry, and powder X-ray diffraction studies which evidenced stable complex formation and increase in amorphousness of diacerein in complex. In brief, the characterization studies confirmed the inclusion of diacerein within the non-polar cavity of HP-β-CD. HP-β-CD complex showed improved in vitro drug release profile compared to pure drug and similar to that of marketed formulation respectively.  相似文献

Novel tetranuclear copper complexes, Cu4(OH)2(ClO4)3 (HA)·H2O (1) and Cu4(ClO4)5(H3B)·3H2O (2), were synthesized by reacting 1,5-bis(1-phenyl-3-methyl-5-pyrazolone-4)-1 ,5-pentanedione with 1,3-propanediamine and 2-hydroxyl-1,3-propanediamine in the presence of a template reagent copper ion. New [2+2] type open cyclic multidentate ligands are also obtained from the reaction (H4A and H6B stand for new compounds from 1,3-propanediamine and 2-hydroxyl-1,3-propanediamine, respectively). They each contain five C = O, three C = N and one NH2 groups. The complexes were characterized by elemental analyses, conductivity, FT-i.r. (micro-i.r., deconvolution technique), FAB-MS, e.s.r., electronic spectra and extended X-ray absorption fine structure (EXAFS). Copper ions in (1) are basically four coordinate with tetragonal geometry. The average coordination bond distances of Cu–N and Cu–O are 1.91 Å and 2.05 Å. In (2), copper ions are primarily five coordinate with square-based pyramidal geometry. The average coordination bond distances of Cu–N and Cu–O are 1.93Å and 2.08Å. Four copper atoms in molecules may be arranged tetragonally. Both the ligand field and the coordination bonds in complex (1) are stronger than those in (2). Investigations on variable temperature susceptibilities show that some antiferromagnetic exchange interaction exist in the complexes. The plots of –1 versus T obey the Curie-Weiss law only at low temperature. Preliminary results of a bioassay indicate that the two complexes have some antitumour activity in vitro.  相似文献

An investigation into the comparative reactivity of simple β-lactones and β-thiolactones toward a thiol and a primary amine is reported. A simple 3-mercaptomethyl-2-oxetanone is found to undergo rearrangement in the presence of aqueous base to give the corresponding thietane-3-carboxylic acid rather than the 3-hydroxymethyl-2-thietanone. Implications for the use of β-thiolactones in bioorganic and medicinal chemistry are discussed.  相似文献

The syntheses, structures, and magnetic properties of a series of di- and trivalent hydridotris(3,5-dimethylpyrazol-1-yl)borate (Tp*) cyanomanganates are described. Treatment of tris(acetylacetonate)manganese(III) [Mn(acac)(3)] with KTp* and tetra(ethyl)ammonium cyanide affords [NEt(4)][(Tp*)Mn(II)(κ(2)-acac)(CN)] (1), as the first monocyanomanganate(II) complex; attempted oxidation of 1 with iodine affords {(Tp*)Mn(II)(κ(2)-acac(3-CN))}(n) (2) as a one-dimensional chain and bimetallic {[NEt(4)][(Tp*)Mn(II)(κ(2)-acac(3-CN))](2)(μ-CN) (3) as the major and minor products, respectively. A fourth complex, [NEt(4)][(Tp*)Mn(II)(η(2)-acac(3-CN))(η(1)-NC-acac)] (4), is obtained via treatment of Mn(acac(3-CN))(3) with KTp* and [NEt(4)]CN, while [NEt(4)](2)[Mn(II)(CN)(4)] (5) was prepared from manganese(II) trifluoromethanesulfonate and excess [NEt(4)]CN. Tricyanomanganate(III) complexes, [cat][(Tp*)Mn(III)(CN)(3)] [cat = NEt(4)(+), 7; PPN(+), 8], are prepared via sequential treatment of Mn(acac(3-CN))(3) with KTp*, followed by [NEt(4)]CN, or [cat](3)[Mn(III)(CN)(6)] with (Tp*)SnBu(2)Cl. Magnetic measurements indicate that 1, 2, and 4 contain isotropic Mn(II) (S = (5)/(2); g = 2.00) centers, and no long-range magnetic ordering is found above 1.8 K. Compounds 7 and 8 contain S = 1 Mn(III) centers that adopt singly degenerate spin ground states without orbital contributions to their magnetic moments.  相似文献

Ring opening of oxazolines, prepared from l-serinates, with trimethylsilyl halides (TMSX) led to β-halogeno-N-benzoyl-α-amino esters in good to excellent yields. Quaternization of triphenylphosphine by the β-bromo or -iodo amino esters gave the corresponding β-phosphonium salts in overall yields of up to 93% and with e.e. >96%. Hydrolysis of the ester function afforded the phosphonium salt bearing an N-benzoyl-α-amino acid substituent, with partial racemization. However, the reaction of the TMSX with the carboxylic salt, prepared by saponification of the starting oxazoline ester, furnished the corresponding β-halogeno-N-benzoyl-α-amino acids in 70–95% yields. Quaternization of triphenylphosphine by the bromo or iodo derivatives led to the phosphonium salts bearing a free acid function in 95% yield, without racemization. The efficiency of this synthesis was demonstrated by the preparation of these phosphonium salts in excellent overall yields, by a one-pot procedure starting from the oxazoline.  相似文献

The title compound β-sitosterol(C29H50O), an active phytosterol in many medicinal and edible plants, was characterized by X-ray diffraction analysis and extensive nuclear magnetic resonance(NMR) data. It crystallizes in monoclinic system, space group P21 with C29H50O·1/2H2O, a = 9.4226(7), b = 7.4824(9), c = 36.889(3) , V = 2597.0(4) 3, Z = 4, Dx = 1.084 g/cm3, Mr = 423.70, F(000) = 948, and μ = 0.064 mm-1. The final R = 0.0886 and wR = 0.2234 for 10157 observed reflections(I 2σ(I)). The molecular crystal structure of β-sitosterol shows relative stereochemistry of 24R-ethylcholest-5-en-3β-ol. The molecule is composed of one steroid nucleus(3 six-membered rings and 1 five-membered ring) and one sidechain of 10 carbons. There are two C29H50O molecules and one H2O molecule in a symmetrical unit, and the title compound is stacked into a special laminated structure through hydrogen bonds and van der Waal forces. The standard (p° = 0.1 MPa) molar enthalpy of formation β-tetralone was measured, at T = 298.15 K, by static bomb calorimetry and the standard molar enthalpy of vaporization, at T = 298.15 K, was obtained using Calvet microcalorimetry.These values were used to derive the standard molar enthalpy of formation of the compound in the gaseous phase, at T = 298.15 K, ?(75.2 ± 2.5) kJ · mol?1.Additionally, high-level density functional theory calculations using the B3LYP hybrid exchange-correlation energy function with extended basis sets and more accurate correlated computational techniques of the MCCM/3 suite have been performed.  相似文献

Background

The defect structure of organic materials is important as it plays a major role in their crystal growth properties. It also can play a subcritical role in “hot-spot” detonation processes of energetics and one such energetic is cyclotetramethylene-tetranitramine, in the commonly used beta form (β-HMX).

Results

The as-grown crystals grown by evaporation from acetone show prismatic, tabular and columnar habits, all with {011}, {110}, (010) and (101) faces. Etching on (010) surfaces revealed three different types of etch pits, two of which could be identified with either pure screw or pure edge dislocations, the third is shown to be an artifact of the twinning process that this material undergoes. Examination of the {011} and {110} surfaces show only one type of etch pit on each surface; however their natural asymmetry precludes the easy identification of their Burgers vector or dislocation type. Etching of cleaved {011} surfaces demonstrates that the etch pits can be associated with line dislocations. All dislocations appear randomly on the crystal surfaces and do not form alignments characteristic of mechanical deformation by dislocation slip.

Conclusions

Crystals of β-HMX grown from acetone show good morphological agreement with that predicted by modelling, with three distinct crystal habits observed depending upon the supersaturation of the growth solution. Prismatic habit was favoured at low supersaturation, while tabular and columnar crystals were predominant at higher super saturations. The twin plane in β-HMX was identified as a (101) reflection plane. The low plasticity of β-HMX is shown by the lack of etch pit alignments corresponding to mechanically induced dislocation arrays. On untwinned {010} faces, two types of dislocations exist, pure edge dislocations with b?=?[010] and pure screw dislocations with b?=?[010]. On twinned (010) faces, a third dislocation type exists and it is proposed that these pits are associated with pure screw dislocations with b?=?[010].

A polycyclic caged compound with high strain—hexanitrohexaazaisowurtzitane (HNIW)—has been synthesized via a three-step reaction: condensation, hydrogenolysis debenzylation and nitrolysis, starting with benzylamine and glyoxal. HNIW is the most powerful high energy density compound (HEDC) ever tested. β-HNIW possesses a caged structure consisting of two five-membered rings and one six-membered ring with a nitro group attached to each of the six bridging nitrogens. The nitro group lies basically within a plane. The lengths of C—C bonds of β-HNIW range from 0. 156 nm to 0.159 nm, 0.002–0.005 nm longer than the sp3 C-C bond. The β-HNIW’s crystal belongs to orthorhombic system and space groupPca21 with parameters:a = 0.9670 (2),b = 1.1616 (2),c = 1.3032 (3) nm;V = 1.4638(5) nm3,Z = 4; Dc = 1.989 g/cm3 and Dm = 1.982 g/cm3. The possibility of coupling of d-glucose and d-galactose with 4-bromo-3-methylaniline, 2,4,6-tribromoaniline, and 2-amino-5-bromopyridine was studied. The substituent in the aromatic ring was found to influence the conditions and possibility of the reaction. The yields of β-d-glucopyranosyl- and β-d-galactopyranosylamines from 4-bromo-3-methylaniline and 2-amino-5-bromopyridine were 50–65%; 2,4,6-tribromoaniline did not react at all.  相似文献

A combined amino- and N-heterocyclic carbene (NHC)-catalyzed one-pot reaction sequence for the synthesis of simple enantioenriched β-hydroxy and β-amino esters using commercially available catalysts at low catalyst loadings has been developed. The desired products were obtained in high yield and excellent enantiopurity. The generation of quaternary stereocenters and application in gram-scale synthesis were also realized, with no requirements of inert or anhydrous reaction conditions, thus making this transformation a highly practical protocol.  相似文献
