8-氟-5-羟基-[1,2,4]三唑并[1,5-c]嘧啶-2-磺酰胺的合成

8-氟-5-羟基-[1,2,4]三唑并[1,5-c]嘧啶-2-磺酰胺（9）为重要的医药、农药等中间体,其合成方法暂无文献报道。以2,4-二氯-5-氟嘧啶为原料,经醚化、肼解、环合、重排、磺酰胺化、脱甲基化等反应合成了9,总收率27.2%,其结构经¹H NMR,¹³C NMR和MS（ESI）确证。     

Condensed Pyridazines. Part I. Synthesis of 2-Oxo-2H-pyrano[2,3-D]-pyridazine and 2-Oxo-2H-pyrano[2,3-c] pyridazine

The reaction of 4,7-diehlorofuro[2,3-d]pyridazine (1) with potassium cyanide in DMSO gave two products, (E)-3,6-diehloro-5-(2-cyanovinyl)-4-hydroxypyridazine (II) and 5,8-dichloro-2-oxo-2H-pyrano[2,3-d]pyridazine (III) as a result of ring opening or ring expansion. A new ring system, 2-oxo-2H-pyrano[2,3-c]pyridazines (IX, XII, XIII) was obtained from 5,8-dichloro-3-methyl-2-oxo-2H-pyrano[2,3-d]pyridazine (VI).     

Structure of New 2-Oxo-2,8-dihydro-[l,2,4]oxadiazolo-[2,3-a]pyrimidinecarbamates

The structure of new 2-oxo-2,8-dihydro-[l,2,4]oxadiazolo-[2,3-a]pyrimidine-carbamates 1 was established by correlating the IR. and NMR. data with these of analogous compounds. A X-ray crystallographic analysis of 1c gave an unequivocal proof of structural assignment.     

1,2,4-Triazino[4,3-c]- and [2,3-c]quinazolines

This paper describes the reactions of 2-o-aminophenyl-1,4,5,6-tetrahydro-1-methyl-1,2,4-triazine with a variety of reagents to give substituted 1,2,4-triazino[4,3-c]quinazolines or 1,2,4-triazino[2,3-c]quinazolines. Certain representative compounds of these two new heterocyclic systems were characterized by the tlc, ir, nmr, mass spectra, and identified by unequivacol synthesis.     

Synthesis and antiviral activity of 4H-[1,2,5]oxadiazolo-[3,4-d]pyrimidine-5,7-dione 1-oxide nucleosides

A series of 4H-[1,2,5]oxadiazolo[3,4-d]pyrimidine-5,7-dione 1-oxide nucleosides was designed,synthesized and evaluated against vesicular stomatitis virus(VSV)in Wish cell.The antiviral activities of all compounds were stronger than those of acyclovir,while their toxicities were similar to those of acyclovir.

This paper describes the results of allowing a variety of reagents, such as, aldehydes, ketones, carboxylic acids, acid chlorides, ortho esters, isocyanates, S-methylisothiourea, carbon disulfide, phosgene, nitrous acid, and cyanogen bromide to react with the multifunctional compound, 3-(o-aminophenyl)-1-methyl-1,4,5,6-tetrahydro-1,2,4-triazine.     

Synthesis of 1,2,4-triazolo[1,5-a]pyrimidine-2-sulfonamides

The 1,2,4-triazolo[1,5-a]pyrimidine-2-sulfonamides are a new class of highly active herbicides. A novel cyclization method for the synthesis of these compounds is discussed.     

Synthesis of 2-aryl-5H-[1,3,4]-thiadiazolo[2,3-b]quinazolin-5-ones

3-Amino-2-thioxo-1,2,3,4-tetrahydroquinazolin-4-ones were converted into 2-aryl-5H-[1,3,4]thiadiazolo[2,3-b]quinazolin-5-ones on treatment with carboxylic acids and POCl₃. 3-Arylmethylidenamino-2-thioxo-1,2,3,4-tetrahydroquinazolin-4-ones also cyclized to 2-aryl-5H-[1,3,4]thiadiazolo[2,3-b]quinazolin-5-ones when oxidized with potassium chlorate in acetic acid, but on heating they were deaminated to give 2-thioxo-1,2,3,4-tetrahydroquinazolin-4-one and aryl nitriles. __________ Translated from Khimiya Geteotsiklicheskikh Soedinenii, No. 5, 787–791, May, 2006.     

Interaction of benzothieno[2,3-c]pyrylium salts with hydrazine. Derivatives of 5H-[2,3]benzothieno[2,3-e]diazepines

A study has been made of the reaction of 1,3-disubstituted bezothieno[2,3-c]pyrylium salts with hydrazine. It has been shown that 1,3-dialkyl-substituted benzothieno[2,3-c]pyrylium salts interact with hydrazine to give N-amino-1,3-dialkylbenzothieno[2,3-c]pyridines. The presence of a pyrylium ring on one of the positions of the phenyl group leads to a mixture of N-amino derivatives and 5H-[2,3]benzothieno[2,3-e]diazepines. In contrast, 1,3-diphenylbenzothieno[2,3-c]pyrylium perchlorate gives exclusively 5H-[2,3]benzothieno[2,3-e]diazepine.L. M. Synthesis of 2-Methyl-3-acetyl-5-oxo-5H-[1]benzopyrano[2,3-b]pyridine

For our synthetic work we needed the title compound. Literature survey revealed that 2-amino-4-oxo-4H-[1] benzopyran-3-carboxaldehyde (2-amino-3-formylchro-mone) can undergo condensation with diethyl malonate (or ethyl cyanoacetate) forming ethyl 2-hydroxy(or amino)-5-oxo-5H-[1]benzopyrano[2,3-b]pyridine-3-carboxylate¹.

The synthesis of new 2-oxo-2,8-dihydro-[l,2,4]-oxadiazoo-[2,3-a] pyrimidine-carbamates 3 described in this paper is based upon a regioselective heterocyclization of the pyrimidine-2,6-bis (alkoxycarbonylamino) N-oxides 4. Structural and mechanistic aspects of that cyclizalion are discussed.

4-Acetylamino-1-phenyl-1,4-dihydro-3(2H)-isoquinolinones and 4-amino-1-phenylisoquinolin-3-ol, prepared from the corresponding 4-hydroxyimino compounds, were converted into new isoquinolines containing fused oxazole, thiazole and 1,4-oxazine rings.     

Tricyclic heteroaromatic systems. 5H-1,2,4-triazolo[5,1-c][1,4]benzodiazepine

The synthesis of the new tricyclic heteroaromatic system 5H-1,2,4-triazolo[5,1-c][1,4]benzodiazepine, diazaanalogue of 5H-pyrrolo[2,1-c][1,4]benzodiazepine, which is the common feature of some antitumor antibiotics, is reported. The structure of the new tricyclic system and of some of its key intermediates is assigned by means of nuclear magnetic resonance studies.     

Synthesis of 5H-1, 2-dihydro-2, 5-dioxochromeno [2,3-b] pyridines

The synthesis of [1]benzothieno[2,3-c]quinolines, [1]benzothieno[2,3-c][1,2,4]triazolo[4,3-a]quinoline,and [1]benzothieno[2,3-c]tetrazolo[1,5-a]quinoline

Photocyclization of 3-chloro-N-phenylbenzo[b]thiophene-2-carboxamide 10 afforded [1]benzothieno[2,3-c]-quinolin-6(5H)-one 11 which was chlorinated to 6-chloro[1]benzothieno[2,3-c]quinoline 12 followed by dechlorination to give [1]benzothieno[2,3-c]quinoline 5 . A series of 6-substituted alkoxy and thioalkoxy[1]benzothieno[2,3-c]quinoline derivatives were prepared along with the N-methyl quaternary salt 13 of 5 . 6-Chloro[1]-benzothieno[2,3-c]quinoline 12 was converted into 6-hydrazino[1]benzothieno[2,3-c]quinoline 23 which upon treatment with formic acid yielded [1]benzothieno[2,3-c][1,2,4]triazolo[4,3-a]quinoline 6 . Treatment of 23 with nitrous acid resulted in [1]benzothieno[2,3-c]tetrazolo[1,5-a]quinoline 7 . Compounds 6 and 7 are novel heterocyclic ring systems.
