The effects of ultraviolet radiation (UV-A, 315-400 nm plus UV-B, 280-315 nm) on photosynthesis and 'light-enhanced dark respiration' (LEDR) in Euglena gracilis have been investigated by using light pulses (80 s) with increasing photon fluence rates of 59, 163, 600, 1180, 2080 and 3340 micromol m(-2) s(-1) and dark periods between the light pulses. LEDR is estimated as the maximum rate of oxygen consumption after a period of light minus the rate of oxygen consumption 30 s after the maximum rate. Without any exposure to UV radiation, the photosynthetic rate and LEDR increase with increasing photon fluence rate. After 20 and 40 min exposures to UV radiation, the photosynthetic rate and LEDR as functions of photon fluence rate are reduced. After a 20 min UV treatment respiration is greater than photosynthesis after the first light pulse of 59 micromol m(-2) s(-1) radiation, and especially at higher photon fluence rates photosynthesis is lower than the control values. The inhibitory effects of UV radiation on photosynthetic rate and LEDR are greater after a 40 min UV exposure than after a 20 min exposure. Only at 600 micromol m(-2) s(-1) is the rate of oxygen evolution greater than that of oxygen consumption after a 40 min UV treatment. Both photosynthetic rate and LEDR are inhibited by the photosynthetic inhibitor DCMU (10(-5) M) in a similar way, which indicates close regulatory interactions between photosynthesis and LEDR. Potassium cyanide (KCN) inhibits dark respiration more than it inhibits LEDR. Dark respiration is not affected to the same degree by UV radiation as are photosynthesis and LEDR.  相似文献

The alga Chlamydomonas nivalis lives in a high-light, cold environment: persistent alpine snowfields. Since the algae in snow receive light from all angles, the photon fluence rate is the critical parameter for photosynthesis, but it is rarely measured. We measured photon irradiance and photon fluence rate in the snow that contained blooms of C. nivalis. On a cloudless day the photon fluence rate at the snow surface was nearly twice the photon irradiance, and it can be many times greater than the photon irradiance when the solar angle is low or the light is diffuse. Beneath the surface the photon fluence rate can be five times the photon irradiance. Photon irradiance and photon fluence rate declined exponentially with depth, approximating the Bouguer-Lambert relationship. We used an integrating sphere to measure the spectral characteristics of a monolayer of cells and microscopic techniques to examine the spectral characteristics of individual cells. Astaxanthin blocked blue light and unknown absorbers blocked UV radiation; the penetration of these wavelengths through whole cells was negligible. We extracted astaxanthin, measured absorbance on a per-cell basis and estimated that the layer of astaxanthin within cells would allow only a small percentage of the blue light to reach the chloroplast, potentially protecting the chloroplast from excessive light.  相似文献

During adaptation of the photosynthetic apparatus of the green alga Scenedesmus obliquus to various light qualities, the accumulation of chlorophylls and pigment-protein complexes (with specific consideration of chlorophyll a/b-binding (Cab) proteins) and cab-gene expression were determined. The fluence rate dependences for chlorophyll accumulation and cab-gene expression were very different. Very low fluence rates of violet (404 nm), blue (461 nm) and red (650 nm) light below the photosynthetic threshold, i.e. between 10(-3) and 10(-1) mumol m-2 s-1, inhibited all of these reactions in cells grown under heterotrophic conditions. At elevated fluence rates (above 1 mumol m-2 s-1), red light retained its negative regulation, whereas blue light stimulated pigment accumulation. Under autotrophic conditions the pattern was more complex, because chlorophyll accumulation was unaffected by light below the photosynthetic threshold. However, the expression of cab-genes was inhibited by red light but stimulated by blue light. Cells adapted to fluence rates, which ensured photosynthetic energy supply (above 1 mumol m-2 s-1), showed an increase in chlorophyll accumulation, blue light being more effective than red light. The results confirm and extend our previous discovery of two antagonistically acting photoreceptors in Scenedesmus which mediate and coordinate the complex functional and structural changes associated with photosynthetic adaptation. One of these receptor pigments is a blue-light receptor with positive action; the other is a violet-red-light receptor which can operate far below the photosynthetic threshold and exerts a negative regulation.  相似文献

A computer-operated spectrograph was recently built at Okazaki, Japan. Different specimens can be placed on a horseshoe-shaped focal curve (10 m long) covering a wavelength range of 250 to 1000 nm so they can be irradiated simultaneously. The linear dispersion is about 0.8 nm/cm. The photon fluence rate on the focal curve is 5 x 1015. photons x cm-2 x s-1 at 300nm and 1 x 1016 photons x cm-2 x s-1 at 600 and at 900 nm. The spectral half width is 5.5 nm or less on the focal curve. The stray light content is about 10-5 of the main peak at the peak wavelength ± 100 nm. Specimens are set in microcomputer-controlled threshold boxes so that wavelengths, photon fluence rates, photon fluences and timing of irradiations are controlled automatically according to a pre-programmed schedule. An optical fiber system is also provided for remote irradiations.  相似文献

Photodynamic therapy (PDT) appears as a promising alternative in the treatment of breast cancer since it can be highly effective in curing cancer while preserving normal tissue. However, predicting outcomes in PDT still constitutes a great challenge. One of the parameters that are usually empirically determined is the rate of photon flux delivered to the tissue (light fluence rate). In the present study, we intended to understand why monolayers of human cells derived from mammary adenocarcinomas (MDA-MB-231 and MCF-7) respond quite differently to fluence rates (cells were irradiated either for 6 or for 16 min) at a fixed light dose (4.5 J cm−2) delivered with an array of LEDs in a typical methylene blue PDT protocol. While death rates of MDA-MB-231 cells were insensitive to the fluence rate, MCF-7 cells showed a quite impressive (three times) decrease in cell death levels in the shorter irradiation protocol. Independent on cell type cell death was invariably correlated with the depletion of reduced glutathione intracellular levels and consequently with widespread redox misbalance. Our data show the potential to optimize fluence rates to provide exhaustion of the cell antioxidant responses in order to circumvent therapy resistance of breast tumors.  相似文献

Isatis tinctoria L. and Isatis indigotica Fort. are biennial herbaceous plants belonging to the family of Cruciferae that are used as a source of natural indigo and show several morphological and genetic differences. Production of indigo (indigotin) precursors, indican (indoxyl beta-D glucoside) and isatan B (indoxyl ketogluconate), together with seed germination ability were compared in Isatis tinctoria and Isatis indigotica grown under six different light conditions (darkness, white, red, far red, blue, yellow light) at 25 degrees C. Light quality influenced both germination and production of indigo precursors in the two Isatis species. Different responsiveness to far red and blue light was observed. Indeed, a detrimental effect on germination by blue and far red light was found in I. tinctoria only. Different amounts of isatan B were produced under red and far red light in the two Isatis species. In I. tinctoria, the level of main indigo precursor isatan B was maximal under red light and minimal under far red light. Whereas in I. indigotica far red light promoted a large accumulation of isatan B. The photon fluence rate dependency for white and yellow light responses showed that the accumulation of indigo precursors was differently influenced in the two Isatis species. In particular, both white and yellow light enhanced above 40 micromol m(-2) s(-1) the production of isatan B in I. indigotica while only white light showed a photon fluence dependency in I. tinctoria. These results suggest a different role played by the labile and stable phytochrome species (phyA and phyB) in the isatan B production in I. tinctoria and I. indigotica. I. indigotica, whose germination percentage was not influenced by light quality, demonstrated higher germination capability compared with I. tinctoria. In fact, I. tinctoria showed high frequency of germination in darkness and under light sources that establish high phytochrome photoequilibrium (red, white and yellow light). Germination in I. tinctoria was negatively affected by far red and blue light. I. indigotica seeds appear to be indifferent to canopy-like light (far red). Our results provide further insights on the distinct behaviour of I. tinctoria and I. indigotica that belong to two different genetic clusters and different original environments.  相似文献

After a critical review of the fundamental equations describing photobiological and photochemical processes occurring in a medium exposed to a quasi‐collimated monochromatic UV light beam, the analysis in this review is extended to analogous processes driven by polychromatic UV light, such as that emitted by medium pressure mercury‐vapor arc lamps. The analysis is based on the Second Law of Photochemistry, namely that all photochemical events must be independent, and the rate of such events must be proportional to the rate of photon absorption. A consistent application of the Second Law of Photochemistry leads to a concept change; hence it is proposed herein to use photon fluence and photon fluence rate, rather than fluence (UV dose) and fluence rate, respectively, in the analysis and interpretation of photobiological and photochemical processes. As a consequence, many equations that have been used in the past must be revised, and some experimental information (e.g. action spectra) needs to be re‐analyzed.  相似文献

Abstract— Light-induced accumulations of amoebae of Dictyostelium discoideum in 'light traps' have been observed. The greatest accumulations are obtained with cell densities of about 8 × 104 cells/mm2. Accumulations are observed at incident fluence rates over about one decade both for white and for monochromatic light; higher fluence rates cause dispersal from the 'light trap'. An action spectrum for the photoaccumulation, calculated from fluence-response curves using the zero thresholds, shows a major peak between 405 and 410 nm and extends through most of the visible spectrum. This action spectrum does not coincide with any of the pigments known to be present in D. discoideum . The cellular basis for the photoaccumulation has been studied. No light effects on cell divisions or cell aggregation are observed during the 2 h duration of an accumulation experiment. Microvideographic analysis of single amoebae is consistent with the hypothesis that the amoebae are positively phototactic and move toward the light scattered from cells in the 'light trap', thus accumulating in the trap.  相似文献

Unicellular thermophilic cyanobacterium Synechococcus elongatus displayed phototaxis on agar plate at 55 degrees C. Equal-quantum action spectra for phototactic migration were determined at various fluence rates using the Okazaki Large Spectrograph as the light source. The shapes of the action spectra drastically changed depending on the fluence rate of the unilateral monochromatic irradiation: at a low fluence rate (3 mumol/m2/s), only lights in the red region had significant effect; at a medium fluence rate (10 mumol/m2/s), four major action peaks were observed at 530 nm (green), 570 nm (yellow), 640 nm (red) and 680 nm (red). At high fluence rates (30-90 mumol/m2/s), the former two peaks remained, while red peaks at 640 nm and 680 nm disappeared and, interestingly, an action peak around 700-740 nm (far-red) newly appeared. These results indicate that two or more distinct photoreceptors are involved in the phototaxis and that suitable photoreceptors are selectively active in response to the stimulus of light fluence rates. Far-red or red background lights irradiated vertically from above drastically inhibited phototaxis toward red light or far-red light, respectively. These results indicate involvement of some phytochrome(s).  相似文献

Abstract Monochromatic UV-B irradiation affects neither the absorption nor the fluorescence of the bulk pigments in the desmid Cosmarium cucumis but it impairs photomovement of these organisms at fluence rates which are not higher than the ambient level of solar UV-B irradiation. Photoaccumula-tions and phototaxis are strongly inhibited especially at wavelengths ≤ 300 nm while photodispersal at higher white light fluence rates is hardly affected by supplementary UV-B. This effect has important consequences for the growth and survival of populations in their natural environment: these photosyn-thetic organisms utilize photomovement to find and stay in areas of suitable visible light fluence rates. The UV-B component of solar irradiation both impairs the strategy of the organisms to find a suitable position and the escape mechanism by which the cells move out of areas with too strong white illuminances which photooxidize the bulk pigments and bleach the population within a few days.  相似文献

Light-induced movement responses of the heterotrichous ciliate Blepharisma japonicum were studied by physiological experiments. Two photosensory responses could be identified. A step-up photophobic response is observed as a very rapid backward movement. Microbeam irradiations of individual cells showed that only the anterior part of the ciliate is able to perceive the light stimulus that mediates the phobic reaction. The action spectrum peaks at approximately 400 nm, which indicates that a blue light receptor is involved.
Positive photokinesis of Blepharisma could be shown as a forward movement that is accelerated by increasing the applied photon fluence rate. The steady state level of the velocity depends highly on wavelength and photon fluence rate of the actinic light. After specific inhibition of the phobic reaction bv 1 m/W NH4+, photokinesis can be induced by microbeam irradiation at any part of the cell.
We isolated two main pigments by thin layer chromatography and characterized them as hypericin-like compounds: a red pigment that is obviously responsible for the red color of the ciliates (= blepharismin). and a yellow one with maximal absorption near 420 nm. The possible photoreceptor functions of these pigments are discussed.
We could not find in Blepharisma a distinct phototactic behavior which is so typical for the related ciliate Stentor.  相似文献

We investigated the role of photosynthesis in the photocontrol of extension growth of the hypocotyl of light-grown Cucumis sativus L. Previous work [Gaba and Black (1985b) Plant Physiol. 79 , 1011] demonstrated that the inhibition of cucumber hypocotyl elongation is a fluence rate dependent response in red light. However, the relative contributions of phytochrome and photosynthesis to the photon flux dependent inhibition response were not clear. Here we have shown that photoperception by the foliar cotyledons as well as the hypocotyl itself are responsible for fluence rate dependence in red light. The inhibitor of photosynthesis diuron [3-(3,4-dichlorophenyl)-1, 1-dimethylurea] reduced both the magnitude of inhibition and the fluence rate dependency in red light, indicating an involvement of photosynthesis. Furthermore, the growth of non-pigmented seedlings (treated with the herbicide norflurazon) was less inhibited by red light, with no fluence rate dependency. In particular, inhibition due to cotyledon photoperception was completely lost in non-pigmented (norflurazon-treated) plants, and much reduced by diuron treatment. Hypocotyl-perceived red light inhibition was only slightly reduced by treatment with norflurazon and diuron. Photosynthesis was compared directly to photo-inhibition of growth: the photon flux response curve of oxygen evolution of green Cucumis cotyledons was distinctly different from that of hypocotyl inhibition. In conclusion, photosynthesis is an essential requirement for the cotyledon-perceived inhibition, but the response itself is not due to photosynthesis.  相似文献

Photodynamic therapy (PDT) for actinic field cancerization is effective but painful. Pain mechanisms remain unclear but fluence rate has been shown to be a critical factor. Lower fluence rates also utilize available oxygen more efficiently. We investigated PDT effect in normal SKH1-HR mice using low and high fluence rate aminolevulinic acid (ALA) PDT and a fractionated illumination scheme. Six groups of six mice with different light treatment parameters were studied. Visual skin damage was assessed up to 7 days post-PDT. Fluorescence and reflectance spectroscopy during illuminations provided us with real-time information about protoporphyrin IX (PpIX) photobleaching. A novel dosing approach was introduced in that we used a photobleaching percentage instead of a preset fluence. Data show similar total and maximum damage scores in high and low fluence rate groups. Photobleaching of PpIX in the low fluence rate groups shows a trend toward more efficient photobleaching. Results indicate that low fluence rate PDT is as effective as and more efficient than high fluence rate PDT in normal mouse skin. Low fluence rate PDT light protocols need to be explored in human studies in search for an effective and well-tolerated treatment for actinic field cancerization.  相似文献

Chlorophyll synthesis is stimulated by red light pulses in the green alga Ulva rigida C. Aghard. Chlorophyll synthesis in darkness is greater after longer red light pulses (30 min) than after shorter red light pulses (5 min). Chlorophyll synthesis was higher after red light pulses of 14 Wm-2 fluence rate than after those of 7 Wm-2. The effect of red light showed some far-red reversibility. The reversion by far-red light was higher after red light pulses of 4 min than after those of 30 min. These results indicate the existence of a rapid induction of chlorophyll synthesis during the red light pulses and a fast escape from photoreversibility. The percentage of reversion is also affected by the fluence rate of the light pulses. The reversion was reduced by about 15% when the photon fluence rate was increased from 7 to 14 Wm-2. Reversion was also observed when red and far-red light pulses were applied successively. Thus, phytochrome or a phytochrome-like photoreceptor could be involved in the induction of chlorophyll synthesis in Ulva rigida.  相似文献

Normal Stentor, called singlets since they have a single membranellar band and oral groove surrounding their frontal field, swim away from light sources and collect in the darker areas of an unevenly illuminated container (photodispersal). Phenotypic variants, called doublets since they have 2 membranellar bands and 2 oral grooves, do not exhibit this behavior. Doublets produce photophobic responses and contractions when illuminated at the same fluence rates which produce those responses in singlets, hence their sensitivity to light is normal. Illumination of the frontal field of doublets produces a photophobic response at lower fluence rates than does illumination of their side or posterior. This directional sensitivity is quantitatively similar to that observed in singlets. However, doublets do not reorient their swimming direction after a phobic response as extensively as do singlets. This failure in reorientation is the probable reason that doublets fail to show photodispersal. These results imply that the mechanism producing photodispersal in singlets depends on photophobic responses or some other, presently undescribed, response which requires an asymmetric frontal field.  相似文献

A computer-controlled three-dimensional tracking and motion analysis system was developed to study the responses of Stentor coeruleus to short light pulses and to evaluate its distribution patterns. In addition to photokinesis and phototaxis, the step-up photophobic response was analyzed, which includes a gravity-controlled component at higher fluence rates and a light direction-dependent component at lower fluence rates.  相似文献

Abstract A method is described for predicting the effect of polychromatic irradiation upon the photo-stationary equilibrium of the plant photoreceptor phytochrome. This method follows from the rate equations for phototransformation and utilizes the in vivo action spectra for phytochrome phototransformation (Pratt and Briggs, 1966). A scanning spectroradiometer interfaced with a microcomputer is used to determine a spectral photon distribution from 360 to 800 nm. The products of the photon fluence rate and the relative quantum efficiencies at 2-nm intervals are summed over the entire visible range to yield a predicted percentage of the pigment in the Pfr form. This value was determined under eight different polychromatic light sources and was generally within 7% Pfr of the value measured in etiolated corn coleoptiles under the same light sources.  相似文献

The Bunsen-Roscoe law, also known as the reciprocity law ( E = f(F) with F = I t ) has only limited validity for monophotonic reactions of nucleic acids. Especially at low fluence rates, the extent of in vitro and in vivo photoreactions of nucleic acids in the far-UV and near-UV range is a function of the fluence and of the fluence rate ( E = f (F;I)). In vitro experiments with poly(dA)poly(dT) clearly show that the far-UV (254 nm) response, indicated by the changes of the ellipticity at 315 nm, does not obey the Bunsen-Roscoe law at low fluence rates in the range between 1 W m-2 and 20 W m-2. In vivo experiments with Escherichia coli revealed very similar anomalies. Studying the growth delay after irradiation with far-UV light at 280 nm or near-UV light at 334 nm, we have confirmed the lack of reciprocity in both spectral ranges. The failure of the Bunsen-Roscoe law for the 280 nm and 334 nm UV irradiation effect at low fluence rates was in the range O < I < 40 W m-2. In both cases reciprocity occurred at higher fluence rates (40 < I < 100 W m-2).  相似文献

Abstract— The hypothesis that phytochrome functions as a sensor of vegetational shade through the perception of the red: far-red photon fluence rate ratio requires that the mechanism of perception be compensated for wavelength-independent fluctuations in fluence rate (Smith, 1982). This paper seeks to establish the lower limit of fluence-rate compensation and to assess whether or not compensation is effective at the total fluence rates typical of herbaceous canopies. Using specially-designed cabinets, Sinapis alba L. (white mustard) seedlings were grown from germination under a range of total photosynthetically-active radiation (PAR = 400 to 700 nm) values and a range of red: far-red ratios. The data indicate that fluence-rate compensation is effective above a PAR value of ca. 60 μ.mol m2 s'. Pretreating seedlings at high red: far-red ratio and a PAR level of 300 (μmol m2S-1for increasing periods of time led to an extension of fluence rate compensation to lower fluence rates. The results are discussed in relation to the photosynthetic competence of the seedlings grown under these conditions.  相似文献
