Chloramine-T (N-sodium-N-chloro-p-toluene-sulfonamide) is a candidate therapeutic drug for treating bacterial gill disease, a predominant disease of a variety of fish species. Research has been initiated to obtain the U.S. Food and Drug Administration's (FDA) approval for the use of chloramine-T on a variety of fish species. An attribute of a therapeutic aquaculture drug that must be characterized before the FDA approves its use is depletion of the drug's marker residue (the drug's parent compound or metabolite of highest concentration in an edible tissue). para-Toluenesulfonamide (p-TSA) is the primary degradation product and marker residue for chloramine-T in rainbow trout. To conduct residue depletion studies for chloramine-T in fish, a robust analytical method sensitive and specific for p-TSA residues in edible fillet tissue from a variety of fish was required. Homogenized fillet tissues from rainbow trout (Oncorhynchus mykiss), walleye (Stizostedion vitreum), and channel catfish (Ictalurus punctatus) were fortified at nominal p-TSA concentrations of 17, 67, 200, 333, and 1000 ng/g. Samples were analyzed by isocratic reversed-phase liquid chromatography (LC) with absorbance detection at 226 nm. Mean recoveries of p-TSA ranged from 77 to 93.17%; relative standard deviations ranged from 1.5 to 14%; method quantitation limits ranged from 13 to 18 ng/g; and method detection limits ranged from 3.8 to 5.2 ng/g. The LC parameters produced p-TSA peaks without coelution of endogenous compounds and excluded chromatographic interference from at least 20 chemicals and drugs of potential use in aquaculture.  相似文献

Chloramine-T is an effective drug for controlling fish mortality caused by bacterial gill disease. As part of the data required for approval of chloramine-T use in aquaculture, depletion of the chloramine-T marker residue (para-toluenesulfonamide; p-TSA) from edible fillet tissue of fish must be characterized. Declaration of p-TSA as the marker residue for chloramine-T in rainbow trout was based on total residue depletion studies using a method that used time consuming and cumbersome techniques. A simple and robust method recently developed is being proposed as a determinative method for p-TSA in fish fillet tissue. The proposed determinative method was evaluated by comparing accuracy and precision data with U.S. Food and Drug Administration criteria and by bridging the method to the former method for chloramine-T residues. The method accuracy and precision fulfilled the criteria for determinative methods; accuracy was 92.6, 93.4, and 94.6% with samples fortified at 0.5X, 1X, and 2X the expected 1000 ng/g tolerance limit for p-TSA, respectively. Method precision with tissue containing incurred p-TSA at a nominal concentration of 1000 ng/g ranged from 0.80 to 8.4%. The proposed determinative method was successfully bridged with the former method. The concentrations of p-TSA developed with the proposed method were not statistically different at p < 0.05 from p-TSA concentrations developed with the former method.  相似文献

Oxytetracycline (OTC) is a drug approved by the U.S. Food and Drug Administration (FDA) to control certain diseases in salmonids and catfish. OTC is also a likely control agent for diseases of other fish species and for other diseases of salmonids and catfish not currently on the label. One requirement for FDA to extend and expand the approval of this antibacterial agent to other fish species is residue depletion studies. The current regulatory method for OTC in fish tissue, based on microbial inhibition, lacks sensitivity and specificity. To conduct residue depletion studies for OTC in fish with a liquid chromatographic method, a bridging study was required to determine its relationship with the official microbial inhibition assay. Triplicate samples of rainbow trout fillet tissue fortified with OTC at 0.3, 0.6, 1.2, 2.4, 4.8, and 9.6 ppm and fillet tissue with incurred OTC at approximately 0.75, 1.5, and 3.75 ppm were analyzed by high-performance liquid chromatography (HPLC) and the microbial inhibition assay. The results indicated that the 2 methods are essentially identical in the tested range, with mean coefficients of variation of 1.05% for the HPLC method and 3.94% for the microbial inhibition assay.  相似文献

3-Trifluoromethyl-4-nitrophenol (TFM) is a pesticide used for the selective control of sea lampreys (Petromyzon marinus) in stream and river tributaries of the Great Lakes. To determine concentrations of TFM and TFM glucuronide in the edible fillet tissue of fish during sea lamprey control treatments, an analytical method was developed to determine the concentrations of these residues in rainbow trout (Oncorhynchus mykiss; RBT) and channel catfish (Ictalurus punctatis; CCF). Homogenized fillets were extracted with methanol-water (80 + 20). TFM and TFM glucuronide were isolated from coextractives by C18 solid-phase extraction. TFM glucuronide was hydrolyzed to TFM by the addition of beta-glucuronidase to the TFM glucuronide extract. The extracts were analyzed separately by liquid chromatography with UV-visible detection. Recoveries from TFM-fortified CCF and RBT tissues were 84.1 and 96.1%, respectively. The method detection limits (MDLs) are 2.4 ng/g for TFM-fortified tissues of CCF and 3 ng/g for those of RBT. Recoveries were 78.8 and 77% from TFM glucuronide-fortified CCF and RBT tissues, respectively. The MDLs for TFM glucuronide-fortified tissues are 3.5 and 6.9 ng/g for CCF and RBT, respectively.  相似文献

A rapid and direct liquid chromatographic (LC) technique is described for the determination of a eutectic mixture of diphenyl oxide and biphenyl such as Dowtherm A thermal heating fluid (THF) in oleochemicals and palm olein. Analysis is performed with an RP-18 column with fluorescence detection (excitation at 247 nm and emission at 310 nm). The isocratic mobile phase (1.0 mL/min) consists of methanol and water (90 + 10, v/v). A linear calibration model (correlation coefficient = 0.9999) was developed directly from used Dowtherm A THF with the biphenyl peak (4.70 min) as a marker. Average recoveries from spiked glycerin, fatty alcohol mixture, methyl ester mixture, fatty acids, and palm olein were 90.9-108.7%, with a detection limit of 0.1 microgram/mL. The technique requires no prior sample cleanup nor extraction steps and is good for quality assurance purposes.  相似文献

A simple reversed-phase high-performance liquid chromatographic method suitable for the simultaneous determination of benzocaine and benzyl benzoate in dermatological preparations is described. An internal standard method was employed, using C18 "bonded phase" silica column and a mobile phase consisting of acetonitrile - water (40:60, v/v), with absorption of the column effluent monitored at 254 nm. No sources of interference were observed. The simultaneous determination of both compounds by the method described is rapid and accurate.  相似文献

A new liquid chromatographic method has been developed for determination of nicarbazin in feeds. Approximately 40 g feed is extracted with 200 mL acetonitrile-water (80 + 20, v/v). An aliquot of the extract is filtered and assayed using a reversed-phase isocratic method that measures the 4,4'-dinitrocarbanilide moiety of nicarbazin at a wavelength of 340 nm. For medicated feeds, the method uses a standard linear range of 5 to 100 microg/mL. For lower levels, a linear range of 50 to 150 ng/mL can be used. The method has a limit of detection of 250 ng/g and a limit of quantitation of 500 ng/g in a 40 g feed sample. Recovery was 99.1%, with a range of 95.2 to 101.8%. In the typical U.S. dosing range of 27 to 113.5 g/ton, the precision of the method based on one analyst, one day, and 2 weighings ranged from 2.8% (113.5 g/ton) to 4.7% (27 g/ton).  相似文献

Two extraction methods were developed for the determination of triasulfuron in soil. Method I included extraction with methanol-phosphate buffer at pH 7 (2 + 1, v/v), liquid-liquid partition with dichloromethane, and cleanup on a liquid chromatographic Si adsorption solid-phase extraction tube. In Method II, Extrelut was added and the sample was then extracted with acetonitrile. In both cases, the extracts were analyzed by liquid chromatography (LC) with UV detection and the LC peak was confirmed by LC/mass spectrometry (MS). The 2 methods were tested on 3 soils having different physicochemical characteristics. Method I gave 83% average recovery and a determination limit of 0.4 microg/kg soil. Method II gave 67% average recovery and a determination limit of 2 microg/kg soil. Examples of application of Method I to field samples are reported.  相似文献

A method for the selective determination of trimethylamine (TMA) in aqueous matrices by liquid chromatography is reported. The proposed procedure is based on the derivatization of the analyte with 9-fluorenylmethyl chloroformate (FMOC) in a precolumn (Hypersil C18, 30 microm, 20 mm x 2.1 mm i.d.) connected on-line to the analytical column (LiChrosphere 100 RP18, 5 microm, 125 mm x 4 mm i.d.). Gradient elution was performed with a mixture of acetonitrile-water-0.05 M borate buffer (pH 9.0). The method has been applied to the direct determination of TMA in water within the 0.25-10.0 microg/ml concentration interval, and can also be adapted to the determination of TMA over the range 0.05-1.0 microg/ml by incorporating a preconcentration stage with C18 solid-phase extraction (SPE) cartridges. Good linearity, reproducibility and accuracy was achieved within the tested concentration intervals. The limits of detection at 262 nm were 50 and 5 ng/ml for the direct method and for the method involving preconcentration, respectively. The proposed conditions allowed the selective determination of TMA in the presence of other primary and secondary short-chain aliphatic amines. The utility of the described procedure has been tested by determining TMA in different water samples.  相似文献

A liquid chromatography (LC) method with UV detection is reported for the determination of the sulfonamide herbicide flumetsulam in soybeans. The ground soybean sample was partitioned between methanol and hexane. The hexane removed the lipids, and the methanol layer containing the analyte was further partitioned between dichloromethane and aqueous phosphate buffer at pH 7.0. The aqueous layer, containing the analyte, was acidified to pH 2.2 and partitioned with fresh dichloromethane. The dichloromethane layer containing the analyte was evaporated, and the residue was dissolved in the LC mobile phase for analysis. A polar embedded C18 column was used with a mobile phase of pH 2.2 aqueous phosphate buffer-acetonitrile (68 + 32), run isocratically with detection at 225 nm. The average recovery was 82% with a relative standard deviation (RSD) of 10%. A coefficient of determination of R2 = 0.9992 was achieved for the analyte calibration curve, from 0.005 to 1 microg/mL. The limit of detection, determined from 3 times the standard deviation of 7 replicate extractions of the lowest fortification level (0.01 microg/g), was 0.005 microg/g with an RSD of 22%. LC/electrospray ionization/mass spectrometry in the positive-ion mode was used for identity confirmation of flumetsulam in the fortified soybean extract. The ions at m/z 326, 348, and 129 were observed.  相似文献

An improved liquid chromatographic procedure is described for the quantitative determination of sulindac, sulindac sulfone, and sulindac sulfide from serum. The procedure makes use of acetonitrile extraction of the compounds of interest from acidified serum samples. Under these conditions extraction efficiencies in the 85 percent range are obtained for each of the compounds. The liquid chromatographic separation of the compounds of interest and the internal standard (indomethacin) is accomplished in an isocratic elution procedure using a nitrile (CN) stationary phase. The HPLC separation procedure is completed in less than 10 minutes, giving excellent resolution and peak shape.  相似文献

A method was developed for the determination of iprovalicarb, a new carbamic acid-amino acid fungicide, by liquid chromatography with UV detection. The method uses a reversed-phase C18 column with lambdamax at 215 nm and methanol-water (72 + 25, v/v) as the mobile phase. Cabbage head and leaves fortified with iprovalicarb were extracted with acetone. Partitioning of the fungicide into dichloromethane was followed by column cleanup on neutral alumina. Fortified soil samples were extracted with acetonitrile, and the extract was subjected to column cleanup. The average recoveries of iprovalicarb from cabbage head, leaves, and soil were 86.50, 82.0, and 84.3%, respectively, for fortification levels of 1 and 2 microg/g for cabbage head and leaves and 2 and 4 microg/g for soil.  相似文献

A liquid chromatographic method was developed using pulsed amperometric detection at a gold working electrode to measure residue levels of ethylenethiourea (ETU) in crops and groundwater. Use of the sequential pulsing program eliminates electrode fouling while preserving the sensitive and selective detection of ETU. Minimum detection limits in crops were 5-10 ppb (1.25-2.5 ng on-column) and 5 ppb (0.5 ng) in groundwater. The commercial availability of the pulsed electrochemical detector and its gold working electrode that remains functional with a minimum of conditioning is an improvement in method simplicity.  相似文献

Summary A simple and sensitive isocratic LC method is described for the determination of erythromycins in fermentation broths. A simple technique utilizing acetone-methyl ethyl ketone, 1∶1, as extraction solvent was coupled with suitable chromatographic conditions—compounds were separated on a 250 mm×4.6 mm i.d., 5 μm, reversed-phase column at 65°C with acetonitrile-0.2m K2HPO4 pH7.0-water, 35:5:60 (v/v), as mobile phase at a flow rate of 1.0 mL min−1. UV detection was performed at 215 nm. Separation of erythromycin F from polar components of the fermentation liquid was sufficient. Erythromycins A, B, C, D, and E, andN-desmethylerythromycin A were also separated, as were known decomposition products of erythromycin A and several unknown components. The method is suitable for monitoring the progress of erythromycin fermentation.  相似文献

A liquid chromatographic (LC) method was developed for the determination of Alternaria radicina and A. alternata toxins in carrots. Toxins were extracted from carrot with an acidified mixture of water-methanol-acetonitrile. The filtered extract was divided in 2 parts that were purified by solid-phase extraction on a C18 column for the analysis of radicinin (RAD), altertoxin-I (ATX-I), alternariol (AOH), and alternariol methyl ether (AME), and on a polymeric Oasis HLB column for tenuazonic acid (TeA). Toxins were quantified by reversed-phase LC with UV diode array detection by using 2 consecutive isocratic mixtures of acetonitrile-sodium dihydrogen phosphate solution. Mean recoveries of TeA, ATX-I, AME, RAD, and AOH from carrots spiked at levels between 0.5 and 3.0 microg/g were 69, 71, 90, 36, and 78%, with mean within-laboratory repeatability of 14, 5, 4, 6, and 18%, respectively. The mean between-laboratory reproducibilities for the determination of TeA, ATX-I, AME, and RAD in spiked samples were 25, 22, 6, and 12%, respectively. Limits of detection (signal-to-noise ratio of 3) for RAD, TeA, ATX-I, AME, and AOH were 0.006, 0.02, 0.02, 0.01, and 0.005 microg/g, respectively. RAD was detected (0.16-13.9 microg/g) in 3 out of 266 carrot samples produced under organic conditions in 3 European locations, whereas A. alternata mycotoxins were not found in any tested samples.  相似文献

A rapid, reversed-phase liquid chromatographic method was developed for the assay of nystatin in the bulk drug and a variety of dosage forms. Analysis was performed on a Symmetry C18 reversed-phase column using a mobile phase of methanol-water-dimethylformamide (DMF; 55 + 30 + 15, v/v/v), with detection by UV at 305 nm. Quantitation is based on the sum of the peak areas of the 2 major isomers of nystatin. The linearity of the assay was determined for a concentration range of 0.05 to 0.2 mg/mL (correlation coefficient > 0.999). Accuracies and precision showed good reproducibility.  相似文献
