A densitometric high performance thin-layer chromatographic (HPTLC) method was developed and validated for quantitative analysis of L-DOPA in tablets. Chromatographic separation was achieved on precoated silica gel F 254 HPTLC plates using a mixture of acetone-chloroform-n-butanol-acetic acid glacial-water (60:40:40:40:35 v/v/v/v/v) as mobile phase. Quantitative analysis was carried out at a wavelength of 497 nm. The method was linear between 100 and 500 ng/microL, with a correlation coefficient of 0.999. The intra-assay variation was between 0.26 and 0.65% and the interassay was between 0.52 and 2.04%. The detection limit was 1.12 ng/microL, and the quantification limit was 3.29 ng/microL. The accuracy ranged from 100.40 to 101.09%, with a CV not higher than 1.40%. The method was successfully applied to quantify L-DOPA in real pharmaceutical samples, including the comparison with HPLC measurements. The method was fast, specific, with a good precision, and accurate for the quantitative determination of L-DOPA in tablets.  相似文献

A method for the determination of mercury in workplace environments in a chlor-alkali plant was optimized. Mercury was collected on activated carbon with a personal sampling pump at a flow-rate of 0.5 l/min, then the carbon was mineralized by heating with potassium permanganate and sulphuric acid. The mercuric ion was next chelated with ammonium pyrrolidinedithiocarbamate and extracted with methyl isobutyl ketone at acidic pH (1.6–5). The chelate in the extract was decomposed with sulphuric acid and the mercuric ion reduced to the metal with stannous chloride. The mercury was determined by coldvapour atomic-absorption spectrometry (CVAAS). The calibration graph was linear up to 5 ng/ml Hg in the initial solution, the relative standard deviation was 4.5% (for 2 ng/ml) and the detection limit was 0.14 ng/ml. All the figures of merit are referred to the initial sample. The proposed method gave good accordance with CVAAS without extraction.  相似文献

A simple method is presented for the routine analysis of iodide in urine. After a one-step sample clean-up, iodide was separated by ion-pair reversed-phase liquid chromatography and detected electrochemically with a silver electrode. The coefficient of variation of a single analysis of iodide in a pooled urine sample (530 nmol/l) was 7.6%. The detection limit, derived from a signal-to-noise ratio of 3, was 3 pmol, corresponding to 0.06 mumol/l. The recovery of iodide added to urine was 96 +/- 7%. The accuracy of the method was assessed by analysing ten different samples with neutron activation analysis. The data obtained with the two methods showed a high correlation (r = 0.991) and did not differ significantly. Excretion of iodide in samples of 24-h urine from a free-living population was shown to have a log-normal distribution and to be higher in men than in women. The iodide/creatinine ratio was independent of sex and increased with age.  相似文献

Summary Samples of suspended particulate matter collected in Duisburg were investigated for phenolic compounds. The loaded filters were extracted with dichloromethane for 10 hours in a soxhlet apparatus. The concentrated extract was resolved in cyclohexane and separated into a non polar and a polar fraction on alumina. The polar fraction was taken up in dichloromethane and steam-distilled. The distillate was extracted with ether. After the evaporation of the ether, the residue was resolved in dichloromethane and allowed to react with benzoyl chloride. After adding toluene the solution was reduced in volume and analyzed in a GC-MS system. For the separation of phenols by means of HPLC the enrichment of the phenols was performed without benzoylization. Six phenols were detected: hydroxibenzene, methylhydroxibenzene, hydroxibiphenyl, hydroxifluorene, benzylhydroxibenzene and dihydroxibiphenyl.  相似文献

This study introduces a preparation method for polymer‐encased monolith frits with improved durability for liquid chromatography columns. The inner surface of the polyether ether ketone tubing is pretreated with sulfuric acid in the presence of catalysts (vanadium oxide and sodium sulfate). The tubing was rinsed with water and acetone, flushed with nitrogen, and treated with glycidyl methacrylate. After washing, the monolith reaction mixture composed of lauryl methacrylate, ethylene glycol dimethacrylate, initiator, and porogenic solvent was filled in the tubing and subjected to in situ polymerization. The tubing was cut into thin slices and used as frits for microcolumns. To check their durability, the frit slices were placed in a vial and a heavy impact was applied on the vial by a vortex mixer for various periods. The frits made in the presence of catalysts were found to be more durable than those made without catalysts. Furthermore, when the monolith‐incorporated tubing was used as a chromatography column, the column prepared in the presence of catalysts resulted in a better separation efficiency. The separation performance of the columns installed with the polyether ether ketone encased monolith frits was comparable to that of the columns installed with the commercial stainless‐steel screen frits.  相似文献

The indirect determination of nitrite was performed with a flow-injection assembly on the basis of the inhibition of the analytical output obtained in a luminometer by oxidation of acriflavine. The acriflavine solution merged with the nitrite and the resulting mixture was injected into a pure water stream. This solution merged with the oxidant solution (potassium permanganate in sulfuric acid medium) and the resulting chemiluminiscence was affected (inhibited) by the presence of nitrite after reaction with the aminoacridine. The method was applicable over the range 10-800 microg l(-1) of nitrite with a correlation coefficient of 0.9960. The relative standard deviation was 1.4% and the throughput was 76 samples h(-1). The influence of foreign substances was also tested. A solid-phase reactor, filled with Amberlite IRA-900, was inserted in the assembly for the on-line preconcentration of nitrite; the analytical output resulted in an increase of up to 11.5-fold. The method was applied to the determination of nitrites in residual waters, industrial formulations and soil samples.  相似文献

Deposition of positively charged nanosized latex particles onto planar silica and cellulose substrates was studied in monovalent electrolyte solutions at pH 9.5. The deposition was probed in situ with optical reflectometry in a stagnation point flow cell. The surface coverage can be estimated reliably with island film theory as well as with a homogeneous film model, as confirmed with atomic force microscopy (AFM). The deposition kinetics on the bare surface was of first order with respect to the particle concentration, whereby the deposition rate was close to the value expected for a perfect collector. The efficiency coefficient, which was defined as the ratio of the experimental and theoretical deposition rate constants, was in the range from 0.3 to 0.7. Subsequently, the surface saturated and a limiting maximum coverage was attained (i.e., blocking). These trends were in qualitative agreement with predictions of the random sequential absorption (RSA) model, where electrostatic interactions between the particles were included. It was observed, however, that the substrate strongly influenced the maximum coverage, which was substantially higher for silica than for cellulose. The major conclusion of this work was that the nature of the substrate played an important role in a saturated layer of deposited colloidal particles.  相似文献

以对乙酰氨基酚(PCT)为模板分子,甲基丙烯酸为功能单体,采用光引发原位聚合法在玻碳电极(GCE)表面聚合成膜,以甲醇-甲酸将模板分子洗脱,制得对乙酰氨基酚分子印迹膜修饰电极(MIP-GCE),建立了该电极直接测定PCT的分析方法。结果表明,该传感器具有较高的选择性和灵敏度,PCT浓度在5.0×10-5~1.0×10-3 mol?L-1范围内与其峰电流呈良好的线性关系,检出限为1.4×10-6 mol?L-1。应用该法测定药物中PCT的含量,在干扰物质共存情况下的回收率为96%~105%。  相似文献

建立离子色谱安培法检测尿液中胱氨酸的分析方法。将尿液稀释处理后,过OnGuardⅡRP前处理柱,去除尿液中的有机物,选用低容量阳离子交换柱IonPac CS17进行分离,以甲烷磺酸梯度淋洗,柱后用300 mmol/L NaOH溶液衍生,离子色谱脉冲安培法检测,外标法定量。胱氨酸的质量浓度在0.1~5.0 mg/L范围内与其色谱峰面积呈良好的线性,线性相关系数r^2=0.999 9,检出限为0.05 mg/L。测定结果的相对标准偏差为0.71%(n=6),样品加标回收率为94%~108%。该方法具有操作简单,专属性强,灵敏度高等优点,可用于临床快速检测尿液中的胱氨酸。  相似文献

A molecularly imprinted polymer (MIP) is synthesised by a noncovalent protocol in which ibuprofen was used as a template molecule. The polymer was evaluated chromatographically and it was seen that the MIP showed cross-reactivity. Subsequently, when this polymer was used as sorbent in SPE it was possible to selectively extract a mixture of nonsteroidal anti-inflammatory drugs from aqueous samples when a cleanup step with dichloromethane was performed. The performance of the MIP was evaluated with river water and water from a wastewater treatment plant, and compared with the performance of a commercial Isolute ENV+ sorbent.  相似文献

The possibility of entrapping polycrystalline TiO2 in a polymeric support, in order to couple the two unit operations, i.e. ultrafiltration and photodegradation of organic pollutants in aqueous solutions, was investigated. To this aim, polymeric membranes for ultrafiltration with entrapped TiO2 were prepared, characterised and tested. The polymeric support chosen was commercial polysulfone (PSf). The membrane preparation was carried out with the technique referred to as phase inversion. A three-component system, with a polymer, a solvent and a non-solvent was used. The best operative conditions were determined in order to obtain the desired membrane morphology. Permeability measurements and photostability tests were also carried out by using a system under pressure. Finally, a preliminary investigation was performed in order to evaluate the photocatalytic activity of the membranes with entrapped TiO2 for the oxidation of 4-nitrophenol as a model molecule in aqueous solutions.  相似文献

Abstract

A rapid, simple, and sensitive method was developed for the determination of three cresol isomers and phenol in urine. The sample was spiked with m-chlorophenol, as internal standard, and hydrolyzed with sulfuric acid at ambient temperature. The hydrolyzed solution was saturated with ammonium sulfate and extracted with ethyl acetate. The extract was injected without derivatization into a gas chromatograph fitted with a flame ionization detector and a glass column packed with 0.1% SP 1000 on Carbopack C. All four analytes, which eluted within 13 minutes, were quantified from standard curves.

The method was evaluated by analysis of spiked urine pools from laboratory workers and samples of urine from workers in a coal gasification plant.  相似文献

Study on biotinylation strategies for competitive immunoassay of estradiol (E2) was carried out. Two types of competitive enzyme immunoassay (EIA) with Biotin-Avidin amplification system were established and optimized.The E2-Biotin conjugate was used as a tracer in one assay, and biotinylated antibody was used as a tracer in the other. In both of EIAs, horseradish peroxidase-labelled Avidin (Avidin-HRP) was used with a spectrometric determination of enzyme activity. The precision, sensitivity and specificity were measured and compared. The results showed that although both were satisfactory in specificity, the EIA with hapten-Biotin showed to be superior to the EIA with biotinylated antibody in sensitivity and precision. The limit of detection of serum E2 was 8 and 50 pg/mL with E2-Biotin and biotinylated antibody as tracer, respectively.  相似文献

The quantitative extraction of cationic surfactant (CS+) in river sediments was studied. Further, the developed method was applied to the spectrophotometric determination of CS+ in urban river sediment samples by solid-phase extraction with membranes. A mixture of methanol and hydrochloric acid was proposed as an eluent. Dried sediment was digested in the eluent under ultrasonic irradiation. After elution, the eluent was evaporated to almost dryness. The residue was dissolved in a small volume of methanol and diluted to a certain volume with water. The pH of the solution was adjusted to 4-5 to separate iron and some other metals as precipitates of hydroxides. The solution was passed through two-piled membranes: first glass-fiber and then polytetrafluoroethylene (PTFE) membranes. A small volume of methanol was passed through the membranes to elute any CS+ retaining on the membranes. After passing the methanol solution through a cationic exchange resin column, the retained CS+ was eluted with methanol containing a high concentration of sodium chloride. Water, Bromophenol Blue (BPB) and hydrochloric acid were added to the solution. The solution was passed through a mixed cellulose ester membrane filter to retain an ion associate of CS+.BPB-. The retained ion associate was dissolved in a small volume of N,N-dimethylformamide together with the membrane filter, followed by the addition of triethanolamine to make the solution alkaline. The absorbance due to BPB2- was measured at 603 nm against a reagent blank. This method was applied to the determination of CS+ in river water and sediment. A cationic surfactant in sediments at 10(-5) mol kg-1 levels was detected with satisfactory precision. It was found that CS+ was about 500-fold enriched in the sediment from water at the place where domestic wastewater was discharged.  相似文献

This paper describes the development of a rapid and sensitive enzyme-linked electrochemical genosensor using a novel microfluidic-based platform. In this work, hybridization was performed on streptavidin-coated paramagnetic micro-beads functionalized with a biotinylated capture probe. The complementary sequence was then recognized via sandwich hybridization with a capture probe and a biotinylated signaling probe. After labeling the biotinylated hybrid with a streptavidin-alkaline phosphatase conjugate, the beads were introduced in a disposable cartridge composed of eight parallel microchannels etched in a polyimide substrate. The modified beads were trapped with a magnet addressing each microchannel individually. The presence of microelectrodes in each channel allowed direct electrochemical detection of the enzymatic product within the microchannel. Detection was performed in parallel within the eight microchannels, giving rise to the possibility of performing a multiparameter assay. Quantitative determinations of the analyte concentrations were obtained by following the kinetics of the enzymatic reaction in each channel. The chip was regenerated after each assay by removing the magnet and thus releasing the magnetic beads. The system was applied to the analytical detection of PCR amplified samples with a RSD% = 6. A detection limit of 0.2 nM was evaluated.  相似文献

The present study explains the molecular level interaction of valeraldehyde with collagen. Valeraldehyde is a monoaldehyde, which involves crosslinking with protein through covalent linkages. The role of valeraldehyde as a crosslinking agent for collagen stabilization was studied. Molecular modeling approaches was used to understand the interaction of collagen like peptide with valeraldehyde, which mimic the aldehyde tanning processes involved in protein stabilization. Crosslinking efficiency of valeraldehyde was found to increase with an increase in concentration due to the higher availability of aldehydic groups involved in crosslinking with collagen. Valeraldehyde interacted collagen membrane showed an increase in thermal stability by 25°C at pH 8. In the presence of valeraldehyde, collagen fibrils nucleation center was shifted from a lower to a higher range. Shift in the nucleation center was observed in the reduction of gelling time. Water accessibility in valeraldehyde interacted collagen membrane was reduced due to a higher crosslinking rate in the collagen. Modified collagen membrane by valeraldehyde at incubation of about 96 h showed higher resistance to collagenolytic activity of 81%. The amino groups reacting appear to be involved in crosslinking with valeraldehyde. Several interaction sites were identified and the docking energy obtained was ?5.539 kcal/mol. The participation of the aldehyde group with amino groups in collagen was observed, which plays a dominant role in the stabilization of peptide by valeraldehyde. It was found that complexes exhibit covalent bonding, hydrogen bonding and electrostatic interaction in the process of stabilization.  相似文献
