The sodium salts of (S)-alanine, (S)-phenylalanine, (S)-valine, and (S)-methionine are condensed with pivalaldehyde to imines 5 . Cyclization by treatment with benzoyl chloride in cold CH2Cl2 gives mainly (4:1 to > 99:1) the (2S,4S)-4-alkyl-3-benzoyl-2-(tert-butyl)-1,3-oxazolidin-5-ones ( 6 ; cis-configuration) in high yields (85–95%). The oxazolidinones 6 and 7 are deprotonated with lithium diethylamide (LDEA) in tetrahydrofuran (THF) and alkylated (Mel, benzyl bromide) or hydroxyalkylated (benzaldehyde) to 4,4-disubstituted oxazolidinones 9 and 10 , respectively, with high diastereoselectivity (9:1 to 50:1; relative topicity ul). Hydrolysis of three of the oxazolidinones to amino acids of known configuration and optical purity indicates that little if any racemization occurs in the process.  相似文献

The efficient synthesis of four benzyl 2-(S)-[(tert-butoxycarbonyl)amino]-ω-iodoalkanoates {benzyl 2-(S)-[(tert-butoxycarbonyl)amino]-3-iodopropanoate, benzyl 2-(S)-[(tert-butoxycarbonyl)amino]-4-iodobutanoate, benzyl 2-(S)-[(tert-butoxycarbonyl)amino]-5-iodopentanoate, benzyl 2-(S)-[(tert-butoxycarbonyl)amino]-6-iodohexanoate} from natural or protected l-amino acids is described.  相似文献

In the title compound, (2-chloro­benzyl)­tris­(pyridine-2-thiol­ato)-κ2N,S2N,SS-tin(IV), [Sn(C7H6Cl)(C5H4NS)3], two of the three pyridine-2-thiol­ato ligands (SPy) are bidentate and one is monodentate. The bonding C atom of the 2-chloro­benzyl group, the S atom of the monodentate SPy and the S and N atoms of the two bidentate SPy ligands form a distorted octahedron around the Sn atom. The three S atoms and the N atom of one of the bidentate SPy ligands occupy the equatorial positions, while the N atom of the second bidentate SPy ligand and the C(CH2) atom are axial. The axial N—Sn—C angle of 157.9 (1)° demonstrates the heavy distortion of the octahedron.  相似文献

Three homologous series of chiral swallow-tailed compounds, alkyl (S)-2-{6-[4-(4′-alkoxyphenyl)benzoyloxy]-2-naphthyl}propionates, (S)HNP(p,n,q) derived from (S)-2-(6-hydroxy-2-naphthyl)propionic acid in conjugation with a variety of swallow-tailed groups, attached to the external side of the chiral centre, have been synthesized and their mesomorphic and electro-optical properties studied. Both (S)HNP(p,1,2) and (S)HNP(p,1,3) exhibited an enantiotropic antiferroelectric SmC*A phase. This implys that the swallow-tailed groups in the molecules favour zigzag pairing of the molecules in the smectic phase. The maximum P S values of compounds (S)HNP(p,1,2) in the antiferroelectric phase were measured in the range 21–30 nC cm-2; those of compounds (S)HNP(p,1,3) were in the range 15–23 nC cm-2, indicating that these chiral compounds possess low polarity. The electro-optical response of the compounds in the antiferroelectric SmC*A phase displayed thresholdless V-shaped switching.  相似文献

Epoxidation of (?)-(1R,2R,4R)-2-endo-cyano-7-oxabicyclo[2.2.1]hept-5-en-2-exo-yl acetate ((?)-5) followed by saponification afforded (+)-(1R,4R,5R,6R)-5,6-exo-epoxy-7-oxabicyclo[2.2.1]heptan-2-one ((+)-7). Reduction of (+)-7 with diisobutylaluminium hydride (DIBAH) gave (+)-1,3:2,5-dianhydroviburnitol ( = (+)-(1R,2R,3S,4R,6S)-4,7-dioxatricyclo[3.2.1.03,6]octan-2-ol; (+)-3). Hydride reductions of (±)-7 were less exo-face selective than reductions of bicyclo[2.2.1]heptan-2-one and its derivatives with NaBH4, AlH3, and LiAlH4 probably because of smaller steric hindrance to endo-face hydride attack when C(5) and C(6) of the bicyclo-[2.2.1]heptan-2-one are part of an exo oxirane ring.  相似文献

The reaction of cyclohexene oxide with homochiral lithium amides, prepared from (S)-phenylglycine and (S)-valine has been studied and (R)-2-cyclohexen-1-ol 3 was prepared in a maximum of 72% ee. The optical purity was determined by 1H NMR measurement of the α-methoxy-α-(trifluoromethyl)phenyl acetic acid (MTPA) derivative of the corresponding alcohol.  相似文献

Treatment of (S)-3-isopropyl-2,5-dimethoxy-3,6-dihydropyrazine with trifluoroacetic acid in MeOD results in regioselective deuteration at its C6-position affording its corresponding (S)-[6-2H2]-isotopomer in excellent yield with no loss of stereochemical integrity at its C3-stereocentre. The lithium aza-enolate of this deuterated chiral template has been alkylated with a range of substituted benzyl bromides to afford (3S,6R)-[6-2H]-3-isopropyl-6-benzyl-bis-lactim ethers that were hydrolysed to afford their corresponding (R)-[α-2H]-phenylalanine methyl esters as hydrochloride salts in good yield.  相似文献

Summary Racemic methyl O-benzyllactate was reduced to the alcohol, transformed into the bromide and reacted with triethylphosphite to give the diethylphosphonate. Removal of protecting groups afforded a phosphonic acid which was purified as its cyclohexylammonium salt. (S)-Ethyl and (R)-isobutyl O-benzyllactate were reduced with LiAlD4 to the corresponding dideuteriated alcohols, which were transformed in the same way as the racemic compound into the chiral (2-hydroxy-[1,1-2H2]propyl)phosphonic acids. The optical purity of alcohols (S)- and (R)-6 b was determined by derivatisation with (+)-MTPA-Cl and1H-NMR-spectroscopy to be 98%. Exchange of the carbonyl-16-oxygen atom of 2-oxopropylphosphonate for oxygen-18 from H2 18O, reduction with NaBH4, deprotection and addition of cyclohexylamine yielded the salt (±)-18 of (2-[18O]hydroxypropyl)phosphonic acid.

Copper(II) complexes of the ligands N2-[(R)-2-hydroxypropyl]- and N2-[(S)-2-hydroxypropyl]-(S)-phenylalaninamide performed chiral separation of N-dansyl-protected and unmodified amino acids in HPLC (reversed phase). With the aim of investigating which species are potentially involved in the discrimination mechanism, the two ligands were synthesized and their complexation equilibria with Cu2+ studied by potentiometry and spectrophotometry in aqueous solution up to pH 11.7. The formation constants of the species observed, [CuL]2+, [CuL2]2+, [CuLH–1]+, [CuL2H–1]+, [CuL2H–2], and [CuL2H–3]?, were quite similar for both compounds and were compared to those of (S)-phenylalaninamide. Most probably, in [CuL2H–3]? the ligands behave as terdentate, with the deprotonated OH group occupying an apical position.  相似文献

The template reaction of {bis[(S)-2-(aminomethyl)pyrrolidine]}copper(II) with formaldehyde, nitroethane, and base in MeOH yields optically pure {1,7-bis[(S)-pyrrolidin-2-yl]-4-methyl-4-nitro-2,6-diazaheptane}- copper(II) ([Cu((S,S)-mnppm)]2+) in high yield. The same reaction with rac-2-(aminomethyl)pyrrolidine is also described. Preparative details and spectroscopic and electrochemical properties of the CuII complexes and of the free ligands are reported and compared with structural, spectroscopic and electrochemical data of the CuII complex of the unsubstituted parent ligand 1,7-bis[(S)-pyrrolidin-2-yl]-2,6-diazaheptane (ppm). The crystal structure of [Cu(ppm)]Cl ClO4 has been determined by X-ray diffraction methods.  相似文献

Treatment of (R) methionine sulfoxide with NaOD led to exchange of the C-4 methylene and C-5 methyl protons; exchange of the chiral C-2 proton did not occur. Reducation with mercaptoacetic acid gave (R)-[4-2H2, 5-2H3] methionine. The latter was converted into its carbobenzyloxy methyl ester sulfoxide, pyrolysis of which followed by deprotection yielded (R)-[4-2H2] vinylglcine as the hydrochloride.  相似文献

Both enantiomers of tert-butyl 2-(tert-butyl)-5,5-dimethyl-4-oxoimidazolidine-1-carboxylate ( 11 ; Bbdmoic) were prepared from L -alanine (Schemes 1 and 2). The parent heterocycle, 2-tert-butyl-5,5-dimethylimidazolidin-4-one ( 12 ; from 2-aminoisobutyramide, H-Aib-NH2, and pivalaldehyde) was also available in both enantiomeric forms by resolution with O,O′-dibenzoyltartaric acid. The compound (R)- or (S)- 11 was used as an auxiliary, but also as a chiral Aib building block in a dipeptide synthesis. The 3-propanoyl derivative 13 of (R)- 11 was used for the preparation of enantiomerically pure 2-methyl-3-phenylpropanoic acid (enantiomer ratio (e.r.) 99.5:0.5), by benzylation of the Zn-enolate (→ 14 ; Scheme 3). Oxidative coupling of the bis-enolate derived from heptanedioic acid and (S)- 11 (→ 23 ) and methanolysis of the auxiliary gave dimethyl trans-cyclopentane-1,2-dicarboxylate ( 26 ) with an e.r. of 93:7 (Scheme 5, Fig. 5). The 3-(Boc-Gly)-Bbdmoic derivative 29 was doubly deprotonated and, after addition of ZnBr2 alkylated with alkyl, benzyl, or allyl halides to give the higher amino-acid derivatives with excellent selectivities (e.r. > 99.5:0.5, Schemes 6 and 7). Michael additions of cuprates to [(E)-MeCH?CHCO]-Bbdmoic 36 occurred in high yields, but high diastereoselectivities were only observed with aryl cuprates (diastereoisomer ratio (d.r.) 99:1 for R = Ph, Scheme 8). Finally, 3-(Boc-CH2)-Bbdmoic 17 was alkylated through the ester Li-enolate with primary and secondary alkyl, allyl, and benzyl halides with diastereoselectivities (ds) ranging from 91 to 98%, giving acetals of Boc-Aib-Xxx-O(t-Bu) dipeptides (Scheme 4). The effectiveness of Bbdmoic is compared with that of other chiral auxiliaries previously used for the same types of transformations.  相似文献

Novel L ‐alanine and L ‐glutamic acid derivatized, carbazole‐containing N‐propargylamides [N‐(9‐carbazolyl)ethyloxycarbonyl‐L ‐alanine N′‐propargylamide and N‐(9‐carbazolyl)ethyloxycarbonyl‐L ‐glutamic acid‐γ‐benzyl ester N′‐propargylamide] were synthesized and polymerized with (nbd)Rh+6‐C6H5B?(C6H5)3] (nbd = norbornadiene) as a catalyst to obtain the corresponding polymers with moderate molecular weights in high yields. Polarimetry, circular dichroism, and ultraviolet–visible spectroscopy studies revealed that both poly[N‐(9‐carbazolyl)ethyloxycarbonyl‐L ‐alanine N′‐propargylamide] and poly[N‐(9‐carbazolyl)ethyloxycarbonyl‐L ‐glutamic acid‐γ‐benzyl ester N′‐propargylamide] took a helical structure with a predominantly one‐handed screw sense in tetrahydrofuran, CHCl3, and CH2Cl2. The helix content of poly[N‐(9‐carbazolyl)ethyloxycarbonyl‐L ‐alanine N′‐propargylamide] could be tuned by heat or the addition of a protic solvent, and the helical sense of poly[N‐(9‐carbazolyl) ethyloxycarbonyl‐L ‐glutamic acid‐γ‐benzyl ester N′‐propargylamide] was inverted by heat in CHCl3 or in mixtures of tetrahydrofuran and CH2Cl2. Poly[N‐(9‐carbazolyl) ethyloxycarbonyl‐L ‐alanine N′‐propargylamide] and poly[N‐(9‐carbazolyl)ethyloxycarbonyl‐L ‐glutamic acid‐γ‐benzyl ester N′‐propargylamide] also took a helical structure in film states. 2-(Trimethylsilyl)ethyl Esters as Carboxyl Protecting Group; Application in the Synthesis of (?)-(S)-Curvularin The mould metabolite curvularin (VIII) has been synthesized with the help of a new carboxyl protecting group that can be removed selectively with fluoride ions. 2-(Trimethylsilyl)ethyl 7-hydroxy-octanoate (III) was acylated with 3,5-dibenzyloxy-phenacetyl chloride (IV) to form V with two different ester groups. Tetrabutyl-ammonium fluoride in tetrahydrofuran cleaved the 2-(trimethylsilyl)ethyl ester in V selectively to form the carboxylate anion of VI together with ethylene and trimethylsilyl fluoride. Curvularin dibenzyl ether (VII) was formed by intramolecular acylation of VI. Removal of the benzyl ether groups in VII by hydrogenolysis led to (±)-curvularin (VIII). The naturally occurring (?)-enantiomer was formed when (+)-(S)-III served as starting material.  相似文献

A stereospecific requirement for hallucinogenesis applies to certain molecules containing an asymmetric center. Thus, only the R-isomers of substituted phenylisopropylamines and lysergic acid diethylamide are psychotomimetic. The enantiomers of a minor hallucinogen, S(+)- and R(-)-3-(2-aminopropyl)indole (α-methyltryptamine) ( 6a and 6b ) were synthesized via a 5-step manipulation from D - and L -tryptophan ethyl ester hydrochloride, respectively. Optical purity of these two isomers was determined by pmr spectroscopy of their complexes with a europium chiral shift reagent using the indole C2 H signal.  相似文献

Synthesis of (?)-(6R)- and (+)-(6S)-Tetrahydro-6-[(Z)-pent-2-enyl]-2H-Pyran-2-one, lactones from Jasminum grandiflorum L. and from Polianthes tuberosa L. (?)-(2S)-Ethyl 2-hydroxyhexanedioate ((2S)- 2 ) was obtained by kinetic resolution of racemic ethyl 2-hydroxy-hexanedioate with baker's yeast. The key intermediates (+)-(5R)- and (?)-(5S)-ethyl 5,6-epoxyhexanoate ((5R)- and (5S)- 6 , resp.) are proved to be useful synthons for the total synthesis of chiral 6-alkyl-δ-lactones, as exemplified by the preparation of both enantiomers of jasmine lactone ((6R)- and (6S)- 10 , resp.).  相似文献

The Rh1(diolefin)complexes [Rh(nbd)( 2 )][PF6] [Rh(1,5-cod)( 2 )][PF6], and [Rh((Z)-α -acetamidocinnamic acid)( 2 )][PF6] ( 2 = the chiral P,N-ligand (S)-1-[bis(p-methylphenyl)phospino]-2-[p-methoxybenzyl)amino]-3-methylbutane have been prepared and characterized. These complexes exit as a mixture of isomers arising from different five-membered-ring conformations and diastereoisomers due to both the prochiral nitrogen and olefin ligands. The three-dimensional solutions structures of these complexes have been studied with the specific aim of understanding how the chiral pocket is built. Aspects of the exchange dynamics and their possible relevance to homogeneous hydrogenation are discussed The solid-state structure for the nbd complex, [Rh(nbd)( 2 )][PF6], as well as detailed one- and two-dimensional 31P-, 13C-, and 1H-NMR results are presented.  相似文献
